ObjectivesTo investigate the genetic variants that are responsible for peripheral neuroblastic tumors (PNTs) oncogenesis in one family case.Materials and MethodsOne family was recruited, including the healthy parents, sister affected by neuroblastoma (NB), and brother who suffered from ganglioneuroma (GN). Whole-genome sequencing (WGS) of germline DNA from all the family members and RNA-seq of tumor RNA from the siblings were performed. Mutants were validated by Sanger sequencing and co-IP was performed to assess the impact of the mutant on chemosensitivity in the SH-SY5Y cell line.ResultsA novel compound heterozygous mutation of BRCA2 was locked as the cause of carcinogenesis. One allele was BRCA2-S871X (stop-gain) from the siblings’ mother, the other was BRCA2-N372H (missense) from their father. This novel compound heterozygous mutations of the BRCA2 gene associated with PNTs by disordering DNA damage and response (DDR) signal pathway. Moreover, chemosensitivity was reduced in the NB cell line due to the BRCA2-N372H mutant.ConclusionIn summary, these results revealed a novel germline compound heterozygous mutation of the BRCA2 gene associated with familial PNTs.

In sociology, it is recognized that the family is the Foundation of society, ensuring both its stability and its development. However, there are situations in which the family itself needs support and assistance not only from loved ones, but also from state structures and social protection bodies. This article is devoted to the study of the family in which a disabled child was born and brought up. The aim of the study was to study the dynamics of relations between parents before and after the birth of a child, the analysis of social and emotional aspects of relations, the characteristics of the situation that contributes to the destruction of the family. Case study was chosen as a research strategy. Three methods were included in this monographic technique: in-depth interview and included observation and semantic differential method. The family can be traced to changes in the relationship from the joyful, carefree to mutual resentment, scandals and accusations. The birth of a child leveled the emotional background of the relationship of parents, making it more smooth, calm, but indifferent. Areas of stress are: long waiting for the baby, the news of the disease of the unborn child, the birth of a child with cerebral palsy, problems of care, communication and education.

Lymphatic malformation (LM) is a benign congenital childhood growth that presents primarily at birth, with the remainder becoming evident by age 2 years. LM can cause devastating complications such as respiratory failure, dyspnea, dysphagia, organ compression, and nerve compression. Surgery is the preferred treatment option, although resection is not always an option due to the anatomic location of the malformation. Other treatments that have been tried with success include sclerotherapy, radiofrequency ablation, propranolol, and sirolimus. Nurses have an important role in caring for the child with LM, as with any complex disease. Nurses provide care at the bedside, education to the family, case coordination, and psychosocial support. LM is a rare disease in infancy necessitating pediatric nurses to support families through prolonged treatments and potential complications.
