The geological context of this study is established in the iron oxide-copper-gold (IOCG) deposit of Sossego (Canaã dos Carajás, Brazil), where hydrothermal alterations in shear zones concentrated the metals of interest and added new characteristics to the metavolcanic-sedimentary and granite rocks. The mineral transformation of rocks by hypersaline fluids enriched in metals and silica also modifies some metallurgical properties, such as abrasiveness. Special bench tests on rock drill cores are used in mapping the abrasiveness of rocks, with the Bond abrasion test being more commonly used in the mining industry, but it has a restrictive sampling protocol and mass requirement for geometallurgical studies. As a counterpoint, the test of the Laboratoire Central des Ponts et Chaussées/Central Laboratory of Bridges and Roads (LCPC) requires a smaller amount of fine material and a finer granulometric range. The study on the use of LCPC was implemented in 40 samples, using Bond Ai as a reference. The results showed a strong correlation between both methodologies (R2 = 95%), validating the use of LCPC to quantify abrasiveness in the Sossego mine. It was also possible to classify the most abrasive lithologies.

Objectives The performance of blood cultures (BC) relies on optimal sampling. Sepsis guidelines do not specify which sampling protocol to use, but recommend two sets of BC bottles, each set containing one aerobic and one anaerobic bottle. For the single-site sampling (SSS) protocol, only one venipuncture is performed for all four bottles. The predominating multi-site sampling (MSS) protocol implies that BC bottles are collected from two separate venipuncture sites. The aim of this study was to compare SSS and MSS. Primary outcomes were number of BC sets collected, sample volume and diagnostic performance. Methods This was a retrospective clinical study comparing BC results in an emergency department before and after changing the sampling protocol to SSS from MSS. All BC samples were incubated in the BacT/ALERT BC system. Results The analysis included 5,248 patients before and 5,364 patients after the implementation of SSS. There was a significantly higher proportion of positive BCs sampled with SSS compared to MSS, 1,049/5,364 (19.56%) and 932/5,248 (17.76%) respectively ( P =0.018). This difference was due to a higher proportion of solitary BC sets (two BC bottles) in MSS. Analyzing only patients with the recommended four BC bottles, there was no difference in positivity. SSS had a higher proportion of BC bottles with the recommended sample volumes of 8-12 ml than MSS ( P <0.001). Conclusions Changing the sampling protocol to SSS from MSS resulted in higher positivity rates, higher sample volume and fewer solitary BC sets. These advantages of SSS should be considered in future sepsis guidelines.

AbstractDuodenoscopy-associated infections occur worldwide despite strict adherence to reprocessing standards. The exact scope of the problem remains unknown because a standardized sampling protocol and uniform sampling techniques are lacking. The currently available multi-society protocol for microbial culturing by the Centers for Disease Control and Prevention, the United States Food and Drug Administration (FDA) and the American Society for Microbiology, published in 2018 is too laborious for broad clinical implementation. A more practical sampling protocol would result in increased accessibility and widespread implementation. This will aid to reduce the prevalence of duodenoscope contamination. To reduce the risk of duodenoscopy-associated pathogen transmission the FDA advised four supplemental reprocessing measures. These measures include double high-level disinfection, microbiological culturing and quarantine, ethylene oxide gas sterilization and liquid chemical sterilization. When the supplemental measures were advised in 2015 data evaluating their efficacy were sparse. Over the past five years data regarding the supplemental measures have become available that place the efficacy of the supplemental measures into context. As expected the advised supplemental measures have resulted in increased costs and reprocessing time. Unfortunately, it has also become clear that the efficacy of the supplemental measures falls short and that duodenoscope contamination remains a problem. There is a lot of research into new reprocessing methods and technical applications trying to solve the problem of duodenoscope contamination. Several promising developments such as single-use duodenoscopes, electrolyzed acidic water, and vaporized hydrogen peroxide plasma are already applied in a clinical setting.

(1) Background: The influence of tap water fittings construction and internal pipe-work on the release of heavy metals was investigated. (2) Methods: A statistical approach was applied for the examination of the chemistry of tap water in five different cities in southern Poland. In total, 500 samples were collected (from 100 to 101 samples in each city). The sampling protocol included information on the construction of the water supply network and the physicochemical parameters of measured tap water. (3) Results: The statistical analysis allowed to extract the crucial factors that affect the concentrations of trace elements in tap water. Age of connection, age of tap, age of pipe-work as well as material of connection, material of pipe-work and material of appliance reveal the most significant variability of concentrations observed for As, Al, Cd, Cu, Fe, Mn, Pb, and Zn. Calculated cancer risks (CRs) decrease with the following order of analysed elements Ni > Cd > Cr > As = Pb and can be associated with the factors that affect the appearance of such elements in tap water. The hazard index (HI) was evaluated as negligible in 59.1% of the sampling points and low in 40.1% for adults. For children, a high risk was observed in 0.2%, medium in 9.0%, negligible in 0.4%, and low for the rest of the analysed samples.

Abstract. Northern African beaches are among the most vulnerable areas under the extreme climate change hazard. Mainly sedimentary low-lying platform, the coasts are supplied by terrestrial yields, which are increasingly interrupted by dams. Unfortunately, the sediment fluxes are rarely measured and monitored, so that it is quite impossible today to assess the contribution of continental sediments to the coast and its variability. The aim of our study is to determine the sampling protocol of delta sedimentation plain and nearshore seabed for better understanding of the anthropogenic driver in contrast to climate change. We adopt a multi-criteria analysis based both on the geomorphologic feature and the historic evolution from the River to the littoral plain. The shoreline evolution reveals an alarming retreat trend reaching −20 m ± 0.15 m yr−1 after the human-induced change where ∼50 % of sediment discharge has been trapped upstream the dam, including quite all the coarse material, like sand. The shoreline retreat and the decreasing sediment rate of fluvial flow are all due to the dam construction.

Abstract Objective Salivary cortisol and cortisone are used as biomarkers of physiological stress. Careful sampling of saliva for profiling of awakening response and the diurnal slope can be challenging in free-living environments, and validated sampling protocols are lacking. Therefore, we investigated (1) the level of compliance to a three-day home-based salivary sampling protocol, and (2) the within subject day-to-day variability of cortisol and cortisone outcomes and the required measuring days to obtain high reproducibility. Results Nineteen healthy adults (mean age: 42, 50% females) participated. Participants collected in total 434 salivary samples out of 456 scheduled (four samples per day over three consecutive days at two time points). We found high level of compliance to the proposed free-living salivary sampling protocol with 18 (95%) and 16 (84%) participants being compliant to numbers and timing of samples, respectively. The area under the curve for the morning salivary samples and peak-to-bed slope had moderate reproducibility for cortisol and cortisone (intraclass correlation coefficient: 0.51–0.68, and mean coefficient of variation: 14.7%-75.3%). Three-to-four measuring days were required for high reproducibility of the area under the curve for the morning salivary samples and peak-to-bed slope using this free-living salivary sampling protocol. Abstract Several studies have detected SARS-CoV-2 in the stool of infected people as in urban wastewater. The quantification of SARS-CoV-2 in wastewater appears today as a powerful tool that can help in Wastewater-based epidemiology (WBE). The goal is to improve the prediction of new waves of COVID-19 outbreaks and provide an early warning of the evolution of the infection. In this research, we highlighted some practical and scientific aspects that emerged during an extensive ongoing monitoring campaign carried out on a large number of wastewater treatment plants located in the province of Trento (North Italy) and aimed at the detection of SARS-CoV-2 in raw municipal wastewater. The open issues underline are related to the collection and storage (sampling protocol, storage and heat treatment), to the molecular analysis (enrichment phase), and to the mathematical calculation of SARS-CoV-2 load in wastewater, suitable for WBE (standard curve to obtain the concentration of genomic units and flow rate measurements). This study provides some insights that can help in the implementation of surveillance plans in other regions.

Sampling, sample preparation, and assay protocols aim to achieve an acceptable estimation variance, as expressed by a relatively low nugget variance compared to the sill of the variogram. With gold ore, the typical heterogeneity and low grade generally indicate that a large sample size is required, and the effectiveness of the sampling protocol merits attention. While sampling protocols can be optimised using the Theory of Sampling, this requires determination of the liberation diameter (dℓAu) of gold, which is linked to the size of the gold particles present. In practice, the liberation diameter of gold is often represented by the most influential particle size fraction, which is the coarsest size. It is important to understand the occurrence of gold particle clustering and the proportion of coarse versus fine gold. This paper presents a case study from the former high-grade Crystal Hill mine, Australia. Visible gold-bearing laminated quartz vein (LV) ore was scanned using X-ray computed micro-tomography (XCT). Gold particle size and its distribution in the context of liberation diameter and clustering was investigated. A combined mineralogical and metallurgical test programme identified a liberation diameter value of 850 µm for run of mine (ROM) ore. XCT data were integrated with field observations to define gold particle clusters, which ranged from 3–5 mm equivalent spherical diameter in ROM ore to >10 mm for very high-grade ore. For ROM ore with clusters of gold particles, a representative sample mass is estimated to be 45 kg. For very-high grade ore, this rises to 500 kg or more. An optimised grade control sampling protocol is recommended based on 11 kg panel samples taken proportionally across 0.7 m of LV, which provides 44 kg across four mine faces. An assay protocol using the PhotonAssay technique is recommended.
