Presently Packaging plays a significant role for Biosimilar product. The process of selecting materials and the type of packaging also offers an opportunity for the Packaging scientist to look for new biological delivery choices. Most injectable protein products were supplied in some sort of glass vial, prefilled syringe, and cartridge. Those product having high Ph content there is a chance of “delamination “from inner surface of glass vial. With protein-based drugs, the biggest issue is the effect of packaging derivatives on the protein’s three-dimensional and surface structure. These are any effects that relate to denaturation or aggregation of the protein due to oxidation or interactions from contaminants or impurities in the preparation. The potential for these effects needs to be carefully considered in choosing the container and the container closure system to avoid putting patients in jeopardy.

Abstract A laboratory, diet-overlay pesticide bioassay was developed using a susceptible population of the tarnished plant bug, Lygus lineolaris (Palisot de Beauvois), to study its susceptibility to neonicotinoid, sulfoxamine, organophosphate, and pyrethroid insecticides (thiamethoxam, sulfoxaflor, acephate, and permethrin, respectively). The diet-overlay bioassay was compared to the traditional glass-vial surface residue bioassay. We measured LC50 values by feeding tarnished plant bug adults known doses of insecticides dispensed on top of diet in a 10% solution of honey water for thiamethoxam and 10% acetone in water solutions for permethrin, acephate, and sulfoxaflor. Both the diet-overlay and glass-vial bioassays used dose-response (mortality) regression lines to calculate LC50 values for each insecticide at 6-, 24-, 48-, and 72-h post-exposure. Data variability from the glass-vial bioassay was higher for permethrin, sulfoxaflor, and thiamethoxam than the diet-overlay bioassay, for all evaluation times. In contrast, there was lower variability among replicates to acephate in the glass-vial assay compared to the diet-overlay assay. Control mortalities observed on diet-overlay bioassay were lower (0–5%) than those observed on the glass-vial bioassay (4–27%). The use of green beans, floral-foam, rolling glass vials, and insect handling made the existing standard method tedious to manipulate and difficult to handle large numbers of individuals. The nonautoclaved solid diet provides an opportunity to significantly reduce cost and variability associated with procedures of other bioassay methods. In general, the baseline data provide a basis for future comparison to determine changes in resistance over time.

Abstract Soybean aphid, Aphis glycines Matsumura, remains the most economically damaging arthropod pest of soybean in the midwestern United States and southern Canada. Foliar applications of a limited number of insecticide modes of action have been the primary management tactic, and pyrethroid resistance was documented recently with full concentration–response leaf-dip and glass-vial bioassays. Full concentration–response bioassays can be cumbersome, and a more efficient assessment tool was needed. In this study, we implemented a diagnostic-concentration glass-vial bioassay using bifenthrin and λ-cyhalothrin. Bioassays were conducted with field-collected soybean aphid populations to assess the geographic extent and severity of resistance to pyrethroids. In 2017, 10 of 18 and 11 of 21 field populations tested with bifenthrin and λ-cyhalothrin, respectively, had mean proportion mortalities less than the susceptible laboratory population. In 2018, 17 of 23 and 13 of 23 field populations tested with bifenthrin and λ-cyhalothrin, respectively, had mean proportion mortalities less than the susceptible laboratory population. Populations collected after reported field failures of a pyrethroid insecticide generally had mean proportion mortalities less than the susceptible laboratory population. In both years, there was a strong correlation between chemistries, which suggests cross-resistance between these insecticides. The diagnostic-concentration glass-vial bioassays reported here will provide the foundation for an insecticide resistance monitoring program with the ability to determine practical levels and geographic extent of insecticide resistance.

Purpose: To assess the stability of insulin detemir at controlled room temperature (RT) at 25°C in different packaging systems over 7 days. Methods: The degradation characteristics of insulin detemir were determined based on the assay results in different packaging systems (pinhole glass vial, closed glass vial, glass syringe, and plastic syringe) at RT using a reverse-phase high-performance liquid chromatography (HPLC) assay method for insulin injection. Each packaging system was compared to insulin detemir stored in the original packaged closed glass vial at 2°C to 8°C. Results: Insulin detemir stored in a closed glass vial and a glass syringe showed minor degradation at the end of day 7 (98.96% ± 1.49% and 99.78% ± 0.10%, respectively). Insulin detemir stored in plastic syringe decreased to 94.90% ± 2.50% by day 3 and to 93.52% ± 0.29% by day 7. Storage in pin-hole glass vial showed an increase in the assay (152.13% ± 0.12%) by day 7. Conclusion: Stability studies in different packaging systems demonstrated that insulin detemir remained stable for at least 7 days in a closed glass vial or glass syringe, but for only 3 days in a plastic syringe at RT. This study will allow pharmacists in the hospital setting to deliver patient-specific insulin doses into an insulin syringe with confidence in the stability.

Abstract. Divergent ice nucleation (IN) efficiencies of quartz, an important component of atmospheric mineral dust, have been reported in previous studies. We show here that quartz particles obtain their IN activity from milling and that quartz aged in water loses most of its IN efficiency relative to freshly milled quartz. Since most studies so far reported IN activities of commercial quartz dusts that were milled already by the manufacturer, IN active samples prevailed. Also, the quartz surface – much in contrast to that of feldspars – is not prone to ammonia-induced IN enhancement. In detail we investigate the influence of solutes on the IN efficiency of various silica (SiO2) particles (crystalline and amorphous) with special focus on quartz. We performed immersion freezing experiments and relate the observed variability in IN activity to the influence of milling, the aging time and to the exposure conditions since milling. Immersion freezing with silica particles suspended in pure water or aqueous solutions of NH3, (NH4)2SO4, NH4HSO4, Na2SO4 and NaOH, with solute concentrations corresponding to water activities aw=0.9–1.0, were investigated in emulsified droplets by means of differential scanning calorimetry (DSC) and analyzed in terms of the onset temperature of the heterogeneous freezing signal Thet and the heterogeneously frozen water volume fraction Fhet. Quartz particles, which originate from milling coarse samples, show a strong heterogeneous freezing peak in pure water with Thet equal to 247–251 K. This IN activity disappears almost completely after aging for 7 months in pure water in a glass vial. During this time quartz slowly grew by incorporating silicic acid leached from the glass vial. Conversely, the synthesized amorphous silica samples show no discernable heterogeneous freezing signal unless they were milled. This implies that defects provide IN activity to silica surfaces, whereas the IN activity of a natural quartz surface is negligible, when it grew under near-equilibrium conditions. For suspensions containing milled quartz and the solutes (NH4)2SO4, NH4HSO4 or Na2SO4, Thet approximately follows ThetΔawhet(aw), the heterogeneous freezing onset temperatures that obey Δawhet criterion, i.e., ThetΔawhet(aw)=Tmelt(aw+Δawhet) with Δawhet being a constant offset with respect to the ice melting point curve, similar to homogeneous IN. This water-activity-based description is expected to hold when the mineral surface is not altered by the presence of the solutes. On the other hand, we observe a slight enhancement in Fhet in the presence of these solutes, implying that the compliance with the Δawhet criterion does not necessarily imply constant Fhet. In contrast to the sulfates, dilute solutions of NH3 or NaOH (molality ≥5×10-4 mol kg−1) reveal Thet by 3–8 K lower than ThetΔawhet(aw), indicating a significant impact on the mineral surface. The lowering of Thet of quartz suspended in dilute NH3 solutions is opposite to the distinct increase in Thet that we found in emulsion freezing experiments with aluminosilicates, namely feldspars, kaolinite, gibbsite and micas. We ascribe this decrease in IN activity to the increased dissolution of quartz under alkaline conditions. The defects that constitute the active sites appear to be more susceptible to dissolution and therefore disappear first on a dissolving surface.
