Immunotoxicology sensu lato comprises not only toxicity toward immune cells, but also biological reactions from immune cells exposed to toxicants, reactions that may have deleterious effects at the organismal level. Within this wide frame, a specific case of interest is represented by the response of macrophages to particulate materials, with the epitome examples of asbestos and crystalline silica. For such toxicants that are both persistent and often encountered in an occupational setting, i.e. at low but repeated doses, there is a need for in vitro systems that can take into account these two parameters. Currently, most in vitro systems are used in an acute exposure mode, i.e., with a single dose and a readout made shortly if not immediately after exposure. We describe here how adequate changes of the culture methods applied to the murine macrophage cell line J774A.1 enable longer periods of culture (several days), which represents a first opportunity to address the persistence and dose-rate issues. To respond to this, the protocol uses a reduction in the concentration of the animal serum used for cell culture, as well as a switch from fetal to adult serum, which is less rich in proliferation factors. By doing so, we have considerably reduced cell proliferation, which is a problem with cell lines when they are supposed to represent slowly-dividing cells such as resident macrophages. We also succeeded in maintaining the differentiated functions of macrophages, such as phagocytosis or inflammatory responses, over the whole culture period. Furthermore, the presence of serum, even at low concentrations, provides excellent cell viability and keeps the presence of a protein corona on particulate materials, a feature that is known to strongly modulate their effects on cells and is lost in serum-free culture. Besides data showing the impact of these conditions on macrophages cell line cultures, illustrative examples are shown on silica- and cobalt-based pigments.

The present study aimed to use remote sensing technology to estimate the concentration of particulate materials in the water entering the reservoirs of dams and consequently investigate the possibility of estimating the amount of sediment carried to the reservoir by flood during the life of the dam and its annual estimate. Using an advanced spectrometer device (ASD), the reflectance values of water containing different amounts of particulate sediments were measured in the range of 400–2500 nm; then, these reflectance values were represented for the Landsat 8 satellite OLI bands using their spectral response functions. In the study of interband correlation with the number of particulate materials, band 2 (blue) and band 5 (near-infrared) were identified to prepare a specific and appropriate model. The specificity of the reflectance difference in the two abovementioned bands was presented as an exponential relationship between the concentration of particulate materials and spectral reflectance. In this model, the RMSE parameter for the maximum possible sediment concentration was equal to 1.57 and the parameter R2 was equal to 0.91. In the second step, at the same time as the satellite passed, the area was visited and the sediments of the Ardak dam reservoir were sampled by recording their location. To complete this research, two measures were performed simultaneously, calculating the concentration of particulate materials sampled in the laboratory environment and their location on the image. Then, the number of particulate materials is estimated by taking into account the coordinates recorded from the images on which the relevant corrections have been made. According to the extracted exponential model, the results of estimating the concentration of particulate matter obtained from the model and Landsat satellite images with the concentration of particulate matter obtained from sampling showed its complete compatibility with field surveys to validate this research.

This paper demonstrates a new method to classify mineral phases in 3D images of particulate materials obtained by X-ray computed micro-tomography (CT), here named mounted single particle characterization for 3D mineralogical analysis (MSPaCMAn). The method allows minimizing the impact of imaging artefacts that make the classification of voxels inaccurate and thus hinder the use of CT to characterize natural particulate materials. MSPaCMAn consists of (1) sample preparation as particle dispersions; (2) image processing optimized towards the labelling of individual particles in the sample; (3) phase identification performed at the particle level using an interpretation of the grey-values of all voxels in a particle rather than of all voxels in the sample. Additionally, the particle’s geometry and microstructure can be used as classification criteria besides the grey-values. The result is an improved accuracy of phase classification, a higher number of detected phases, a smaller grain size that can be detected, and individual particle statistics can be measured instead of just bulk statistics. Consequently, the method broadens the applicability of 3D imaging techniques for particle analysis at low particle size to voxel size ratio, which is typically limited due to unreliable phase classification and quantification. MSPaCMAn could be the foundation of 3D semi-automated mineralogy similar to the commonly used 2D image-based semi-automated mineralogy methods.
