A complete mathematical model for measuring the parameters of the scattering matrix for a measurement object [Sx] in the form of a four-port network is developed, in which the mathematical model of the eight-port error is described by 16 parameters of the scattering matrix [E]. In addition, in comparison with the 12-parameter model of the eight-port error network, four parameters are included, which allow taking into account leaks, parasitic transmissions of microwave modules under study (microwave microassemblies). Due to the use of matrix analysis methods, the equations in matrix form are obtained that connect the matrices of the measurement object [Sи] and the actual values of the matrix parameters [Sx], with the aim of enabling the solution of these equations instead of the scattering matrix [E] to use a transmition matrix [T] in the form of cellular matrices [Taa], [Tab], [Tba], [Tbb].

Abstract This article aims at presenting ISIS expansion in North Africa in the first quarter of 2016, with its subsequent implication in the wider framework of Jihadist proliferation worldwide. It can be argued that, while losing real estate in the Middle East, ISIS has started a permanent search for extra-cellular matrices or an ongoing process of de- and reterritorialization. The allegiance and support pledged by other African-based terrorist groups or organizations such as Boko Haram, al-I’tisam of the Koran and Sunnah in Sudan, al-Huda Battalion in Maghreb of Islam, The Soldiers of the Caliphate, al-Ghurabaa, Djamaat Houmat ad-Da’wa as-Salafiya and al-Ansar Battalion in Algeria, Islamic Youth Shura Council, Islamic State Libya (Darnah), in Libya, Jamaat Ansar Bait al-Maqdis, Jund al-Khilafah and Mujahideen Shura Council in the Environs of Jerusalem in Egypt, Okba Ibn Nafaa Battalion, Mujahideen of Tunisia of Kairouan and Jund al-Khilafah in Tunisia and al-Shabaab Jubba Region Cell Bashir Abu Numan in Somalia is an alarming hypothesis of Jihadism reaching “the threshold of inevitability”- syntagm existent in the network theories of David Singh Grewal- turning a whole region, continent of even world into what Nassim Nicholas Taleb would call Extremistan.

ABSTRACTSofosbuvir (SOF) is a highly efficacious and well-tolerated uridine nucleotide analog that inhibits the hepatitis C virus (HCV) NS5B polymerase enzyme. SOF is administered as a prodrug, which undergoes intracellular phosphorylation by host enzymes to a monophosphate, diphosphate, and finally a pharmacologically active triphosphate. In order to fully understand the clinical pharmacology of SOF, there is a great need to determine the intracellular phosphate concentrations of the drug. We describe the validation and utilization of a method to characterize SOF's disposition into variousin vivocell types, including hepatocytes, peripheral blood mononuclear cells (PBMC), and red blood cells (RBC). Standard bioanalytical validation criteria were applied to lysed cellular matrices, with a validated linear range of 50 to 50,000 fmol/sample for each phosphate moiety. The assay was utilized to collect the first data demonstrating concentrations of phosphorylated anabolites formed in PBMC, hepatocytes, and RBCin vivoduring SOF therapy. Median concentrations in PBMC were 220 (range, 51.5 to 846), 70.2 (range, 25.8 to 275), and 859 (range, 54.5 to 6,756) fmol/106cells in the monophosphate, diphosphate, and triphosphate fractions, respectively. In contrast, RBC triphosphate concentrations were much lower than those of PBMC, as the median concentration was 2.91 (range, 1.14 to 10.4) fmol/106cells. The PBMC triphosphate half-life was estimated at 26 h using noncompartmental approaches, while nonlinear mixed-effect modeling was used to estimate a 69 h half-life for this moiety in RBC. The validated method and the data it generates provide novel insight into the cellular disposition of SOF and its phosphorylated anabolitesin vivo.

Molecular beacons (MBs) represent a class of nucleic acid probes with unique DNA hairpin structures that specifically target complementary DNA or RNA. The inherent “OFF” to “ON” signal transduction mechanism of MBs makes them promising molecular probes for real-time imaging of DNA/RNA in living cells. However, conventional MBs have been challenged with such issues as false-positive signals and poor biostability in complex cellular matrices. This paper describes the novel engineering steps used to improve the fluorescence signal and reduce to background fluorescence, as well as the incorporation of unnatural nucleotide bases to increase the resistance of MBs to nuclease degradation for application in such fields as chemical analysis, biotechnology, and clinical medicine. The applications of these de novo MBs for single-cell imaging will be also discussed.
