AbstractMycobacterium tuberculosis is the cause of one of the most important infectious diseases in humans, which leads to 1.4 million deaths every year1. Specialized protein transport systems—known as type VII secretion systems (T7SSs)—are central to the virulence of this pathogen, and are also crucial for nutrient and metabolite transport across the mycobacterial cell envelope2,3. Here we present the structure of an intact T7SS inner-membrane complex of M. tuberculosis. We show how the 2.32-MDa ESX-5 assembly, which contains 165 transmembrane helices, is restructured and stabilized as a trimer of dimers by the MycP5 protease. A trimer of MycP5 caps a central periplasmic dome-like chamber that is formed by three EccB5 dimers, with the proteolytic sites of MycP5 facing towards the cavity. This chamber suggests a central secretion and processing conduit. Complexes without MycP5 show disruption of the EccB5 periplasmic assembly and increased flexibility, which highlights the importance of MycP5 for complex integrity. Beneath the EccB5–MycP5 chamber, dimers of the EccC5 ATPase assemble into three bundles of four transmembrane helices each, which together seal the potential central secretion channel. Individual cytoplasmic EccC5 domains adopt two distinctive conformations that probably reflect different secretion states. Our work suggests a previously undescribed mechanism of protein transport and provides a structural scaffold to aid in the development of drugs against this major human pathogen.

Background: Anterior talofibular ligament (ATFL) injuries are the most common in ankle torsional injuries. ATFL and peroneal tendons are both important stabilizers of lateral ankle joint. We aimed to evaluate peroneal tendons and ATFL. Methods: Fifteen nonpaired leg of fresh frozen cadavers were assessed in this study. After harvesting, ATFL diameters were measured at three points by calipers, these are fibular side, intermediate side and talar side. The mean of these three measurements were assessed and tissue a 15 lb load was applied to the peroneal tendons for 10 minutes, and the transverse diameters were measured by folding the thickest part of the tendon in a double-strand. Results: 5 single bundle, 8 double-bundle and 2 three bundles of ATFL were obtained after dissection. . There was no correlation between ATFL diameter, peroneus longus, peroneus brevis and total tendon diameters of peroneus longus and peroneus brevis in women (p> 0.05). A strong correlation was found between ATFL diameter, peroneus longus (r: 0.95), peroneus brevis (r: 0.81) and total tendon diameters of peroneus longus and peroneus brevis (r: 0.92) in men. Conclusion: Relationship between the diameter of the ATFL and peroneal tendons diameters were evaluated and a correlation was observed in males, while no correlation was observed in females.

New economic development, which is a result of globalisation and internationalisation, has exerted tremendous pressure on the Indian human resource functions how to evolve itself and add the value to the outcome of the firm. The present paper precisely discusses the globalisation and its impact on the HRM practices of Indian firm in the light of new economic reforms. This study aims to examine the far-reaching changes in the pattern of HR practices owing to ever-increasing globalisation in India. The data were collected through the selfdeveloped questionnaire from 282 senior HR managers of Indian firms. The principal component analysis with varimax rotation generated three components from fifteen HR practices, namely Formal, Motivational and Strategic HR practices. Structural Equation Modelling (SEM) technique has been applied to understand the relationship between globalisation and three bundles of HR practices by using the SPSS Amos 22 software. The study findings suggest that the role of globalisation cannot be neglected. Indeed, The HR policies and practices of Indian firms are strongly influenced by the pace of globalisation of the Indian economy.

ObjectiveTo assess quality of care for children presenting with acute abdominal pain using validated indicators.DesignAudit of care quality for acute abdominal pain according to 21 care quality indicators developed and validated in four stages.Setting and participantsMedical records of children aged 1–15 years receiving care in 2012–2013 were sampled from 57 general practitioners, 34 emergency departments (ED) and 28 hospitals across three Australian states; 6689 medical records were screened for visits for acute abdominal pain and audited by trained paediatric nurses.Outcome measuresAdherence to 21 care quality indicators and three bundles of indicators: bundle A-History; bundle B-Examination; bundle C-Imaging.ResultsFive hundred and fourteen children had 696 visits for acute abdominal pain and adherence was assessed for 9785 individual indicators. The overall adherence was 69.9% (95% CI 64.8% to 74.6%). Adherence to individual indicators ranged from 21.6% for assessment of dehydration to 91.4% for appropriate ordering of imaging. Adherence was low for bundle A-History (29.4%) and bundle B-Examination (10.2%), and high for bundle C-Imaging (91.4%). Adherence to the 21 indicators overall was significantly lower in general practice (62.7%, 95% CI 57.0% to 68.1%) compared with ED (86.0%, 95% CI 83.4% to 88.4%; p<0.0001) and hospital inpatient settings (87.9%, 95% CI 83.1% to 91.8%; p<0.0001).ConclusionsThere was considerable variation in care quality for indicator bundles and care settings. Future work should explore how validated care quality indicator assessments can be embedded into clinical workflows to support continuous care quality improvement.

Part II of the book focuses on understanding the link between the nature of the reform at stake, the process engaged to achieve reform, and the end result (adoption or non-adoption of reform). The chapter presents the comparison of three bundles of reforms in France, Ireland, and Italy between 2000 and 2015, and formulates propositions about the prediction of the outcomes of reform. The chapter discusses link(s) between antecedent conditions and the likelihood of a given type of reform and process emerging. Next, theoretical propositions centre on the interaction between the nature of the reform (divisive or consensual, depending on the level of support for the reform), and the nature of the process of reform (majoritarian, supermajoritarian, or externalized, depending on the level of inclusion). The chapter explains why the effect of veto players and of multifaceted reforms varies according to the type of reform and type of process chosen.

Cada haz cortical llega a estar rodeado por una vaina de fibras que, a menudo, mostraba la presencia de una capa gelatinosa (fibras G). A medida que avanza el crecimiento, cada haz se puede dividir en dos o tres haces. Algunos de los haces son mayores y muestran elementos vasculares bien diferenciados debido a su asociación con frutos en desarrollo, mientras que los más pequeños, con pocos vasos, pueden ser trazas foliares o de yemas de flores que caen antes de la fecundación. El xilema secundario del haz cortical está compuesto de vasos, fibras y células del parénquima axial, mientras que el floema consiste en tubos cribosos, células de acompañamiento y células del parénquima axial.The development of cortical bundles, in the branches and peduncles of Couroupita guianensis (Lecythidaceae), initiates close to the apical meristem concomitant with the normal vascular bundles. Each cortical bundle becomes surrounded by a sheath of fibres, which most often showed presence of gelatinous layer (G-fibres). As growth progresses, theses bundle may divide into two-three bundles. Some of the bundles are larger and show well differen­tiated vascular elements due to their association with developing fruits while narrower bundles, with few vessels, may be leaf traces or flower bud traces that fell down before fertilization. The secondary xylem of cortical bundle is composed of vessels, fibres and a­xial parenchyma cells while phloem consists of sieve tubes, companion cells and axial parenchyma cells.

The study showed significant differences between the average weight lens and the average amount protein in the lens between that Kestrel Falco tinnunculus L. and the Collared Dove Streptopelia decaocto F. , also the study electrical migration of lens proteins having one bundle of crystalline –? in Kestrel compared with three bundles in Collared Dove, two bundles of crystalline – ? in both , and crystalline – ? appeared as one bundle in both birds.

It is well known that the three bundles of bamboo strips found at Guōdiàn containing versions of materials appearing in the Lǎozǐ included no passages from the last fifteen chapters of the received text. This article examines closely those final ­fifteen chapters, arguing that they articulate a coherent philosophical position funda­mentally different from and even opposed to that of the Guōdiàn materials. The foundation of the last fifteen chapters is an anthropomorphic conception of heaven that supports the good, a view explicitly rejected in the rest of the Lǎozǐ. These last fifteen chapters support the use of punishments, show no awareness of skepticism about language or moralizing categories, and they have no concern with internal practices of self-cultivation. The article concludes with some reflections on the background of the last fifteen chapters and on the significance of this argument for interpreting the Lǎozǐ, particularly the split between so-called “religious” and “philosophical” readings.
Il est bien connu que les trois faisceaux de fiches de bambou découverts à Guodian portant des recensions de matériaux figurant dans le Laozi ne contiennent aucun ­passage des quinze derniers chapitres du texte reçu. Cet article propose un examen soigneux de ces quinze derniers chapitres et affirme qu’ils énoncent une position philo­sophique cohérente, fondamentalement différente de celle des textes de Guodian, voire en opposition avec eux. Les quinze derniers chapitres se fondent sur une conception anthropomorphique d’un Ciel encourageant le bien. Or cette vue est explicitement rejetée par le reste du Laozi. Ils encouragent le recours aux châtiments, ne manifestent aucun scepticisme au regard du langage et des catégories morales, et se désintéressent des pratiques internes du perfectionnement de soi. L’article se conclut par quelques réflexions sur l’arrière-plan des quinze derniers chapitres et sur la portée des arguments de l’auteur pour l’interprétation du Laozi, notamment la coupure entre les lectures dites “religieuses” et “philosophiques”.

An octagonal FPSO has been proposed for marginal oil and gas development in shallow waters. A shuttle tanker will be deployed near the FPSO during offloading operations. This new concept simplifies the design and manufacturing processes, yet maintains full production, storage, and offloading functions of a conventional ship-shaped FPSO. However, design of the mooring system for this floating unit imposes technical challenges due to: 1) high environmental loads expected on this unit, 2) large dynamic offsets of the unit in shallow waters, and 3) inadequate performance of catenary mooring systems in shallow waters. Thus, development of a viable station keeping solution becomes a key issue to the new concept FPSO design. In this paper, an innovative mooring system is designed to meet the challenges. The FPSO mooring system consists of pile anchors, bridle chains, anchorage buoys, and polyester ropes. Nine mooring lines are grouped into three bundles which evenly spread around the FPSO. The shuttle tanker is attached to the FPSO with a nylon rope hawser at the bow and secured to pre-installed anchorage buoys at the stern with two other nylon ropes. Analyses have been performed for the FPSO mooring system. It is concluded that the proposed mooring system is fully functional and effective.
