We investigated the genetic diversity of the population of captive forest musk deer (Moschus berezovskii) in Barkam Musk Deer Breeding Centre using twelve microsatellite markers, and then analyzed the change in genetic structure of successive generation groups from the population. The data provide a new understanding for the evaluation and usage of the breeding management system. Microsatellite marker analysis detected 141 alleles with an average of 11.75 alleles for each marker. The average expected heterozygosity (HE) was 0.731. Performing an F-statistical analysis on the data showed that the genetic diversity of population decreased, and the inbreeding coefficient significant increased with the increase of generation, and FIS of the 1st generation is significantly lower than that of the second to fifth generation (p < 0.01). The result suggested that the captive population was facing the pressure of inbreeding (FIS = 0.115) and the subsequent loss of genetic diversity. Therefore, it is necessary to improve the breeding management system of the captive population by preventing close relatives from mating or inducing new individuals from the exotic population.

The diversity status of local goats of Karnataka was studied by using microsatellite marker analysis. The genomic DNA from unrelated local goats were PCR- amplified with a panel of 23 microsatellite markers. Microsatellite PCR products were multiplexed and run on capillary based genetic analyser and the raw data obtained was analysed. Totally 158 alleles were observed and the number of alleles ranged from three (ILSTS005 and OarJMP29) to 13 (RM088). The number of effective alleles ranged from 2.25 (ILSTS005) to 8.40 (RM088) in all the 23 loci studied. The mean observed heterozygosity (Ho) was 0.4698±0.2214 [range 0 (ETH225) to 0.8462 (ILSTS034)] and the mean expected heterozygosity (He) was 0.7471± 0.1098 [range 0.5656 (ILSTS005) to 0.9138 (SRCRSP 8)] indicating the heterogenous nature of the local goat population of Karnataka.

AbstractThe research on domestic donkey has little information and criteria comparing to other livestock, companion animal in South Korea. We analyzed genetic database of domestic donkey using microsatellite marker to clarify domestic donkey identification and paternity test. It is the first microsatellite marker analysis on domestic donkey in South Korea. We studied 179 horse samples from 50 Thoroughbred, 50 Jeju Halla horse, including 79 donkeys then analyzed 15 microsatellite marker (AHT4, AHT5, ASB2, ASB17, ASB23, CA425, HMS1, HMS2, HMS3, HMS6, HMS7, HTG4, HTG10, LEX3 and VHL20). We observed genetic diversity from biostatic analysis of them.The number of alleles on total average is 6.08 observed from 1 (ASB17), 2 (HMS1) to 14 (AHT5). The observed heterozygosity (OHet) is from 0.0000 (ASB17, HMS1) to 0.8608 (ASB23) which is mean value of 0.4861, the expected heterozygosity (EHet) is from 0.0000 (CA425) to 0.9104 (AHT5) with mean value of 0.5915, and the Polymorphism Information Content (PIC) on each group of microsatellite marker is from 0.0000 (ASB17) to 0.8968 (AHT5) observed as a mean value of 0.5374. Among 15 markers AHT4, AHT5, ASB23, CA425, HMS2, HMS3, HTG4, HTG10, LEX3 is observed above 5.000.The results on 15 microsatellite marker analysis of 3 horse groups is that the donkey had 0.5915 EHet, 0.4861 OHet on average, Thoroughbred had 0.6721 EHet, 0.6587 OHet on average, and the Jeju halla horse had 0.7898 EHet, 0.7093 OHet on average observed. Furthermore the mean alleles value is observed as 6.08, 4.83, 8.00 in donkey, Thoroughbred, Jeju halla horse breed in each.

Microsatellite markers or simple sequences repeats are DNA - based molecular techniques that areused to see the different among accessions and inbred lines. There are three methods to analysis the results ofthe polymerase chain reaction of microsatellite markers namely polyacrylamide gel electrophoresis (PAGE),capillary electroforesis, and Metaphor Agarose Gel Electroforesis (MAGE), and the Use of MAGE assessedmore easily and economically the polymorphic pattern of DNA markers. This study aimed to obtain fast,effective and efficient in term of easy and cheap technique to identify microsatellite markers of some blackrice cultivars and F2 populations from crosses between black with white rice. The results showed that MAGEsuccessfully separated clearly SSRs alleles with different sizes of less than 25 bp .
