The study aimed to produce fluorescent protein transgenic medaka Oryzias curvinotus (Nichols & Pope, 1927) as a novel strain of ornamental fish. These fish were produced by transferring a plasmid consisting of a fluorescent reporter gene and a strong promoter into one-cell stage embryos. For this purpose, myosin light chain 2, but not other promoters, was mainly used. The study also evaluated the stability of the transgenic medaka germline acquiring vivid fluorescent phenotypes via the transgenesis of the cyan fluorescent protein (CFP) gene under the control of O. curvinotus skeletal alpha-actin (OCacta) promoter. The pOCacta-CFP plasmid, containing a OCacta promoter and CFP reporter gene, was transferred into the one-cell stage of O. curvinotus embryos by a microinjection technique. As a result, 36 of 1386 microinjected O. curvinotus embryos exhibited CFP signals in their trunks. The expressed CFP signals in O. curvinotus embryos and adults were detected under a microscope using a green fluorescent protein (GFP) filter (450–490 nm wavelength), and blue LED light (400–450 nm wavelength). Five O. curvinotus founders showing clear CFP signals were selected and crossed with non-transgenic counterparts to produce subsequent generations. Among strains, the frequency of germline transmission from founder to F1 was highly variable. Only two of the five founders successfully pass the transgene to the F1 generation. At present, the progeny of subsequent generations is being produced and tested for the expression of CFP signals, and therefore, stable lines are ongoing.

 Ion chì là một trong những cation có tác động lên sức sống, khả năng sinh trưởng, dị tật về xương sống trong quá trình sống của cá. Nghiên cứu được tiến hành nhằm đánh giá tác động của ion chì (Pb2+) ở các nồng độ 0 µg/L; 50 µg/L; 100 µg/L; 150 µg/L lên quá trình phát triển cá Sóc giai đoạn ấu trùng sau nở từ 1-10 ngày tuổi. Kết quả cho thấy: (i) ở các nồng độ Pb2+ được kiểm tra khác nhau, nồng độ tối thiểu Pb2+ ảnh hưởng đến tỉ lệ sống của cá Sóc là 50 µg/L; (ii) ở mọi nồng độ được kiểm tra Pb2+, tỉ lệ sống của cá bị ảnh hưởng nhiều nhất vào ngày thứ 5 tại nồng độ 150 µg/L. Dựa vào kết quả thu được, chúng tôi thiết lập một phương trình để dự đoán tỉ lệ sống của ấu trùng cá và tìm được nồng độ LD50 Pb2+ gây chết cá ấu trùng sau 10 ngày nuôi là 98 µg/L và LT50 của Pb2+ ở nồng độ 150 µg/L là 9,76 ngày. Kết quả nghiên cứu cho thấy, nồng độ Pb2+ càng cao càng làm giảm tỉ lệ sống của cá, tăng dị tật xương sống cá ấu trùng và ảnh hưởng cao nhất tại nồng độ 150 µg/L.

Species of the genus Oryzias are models for studies of adaptation to osmotic environments because they demonstrate inter-species adaptability. Oryzias curvinotus, a fresh and brackish water species, is distributed from the coastal waters of southern China to northern Vietnam. The correlation of O. curvinotus’ distribution with physical water conditions in the Tien Yen estuary of northern Vietnam was recorded during monthly collections for one year. For the current study, a total of 2916 individuals of O. curvinotus were caught, of which 942 were selected to be measured and have their developmental stage recorded: 41 larvae, 749 post-larvae and 152 juveniles or adults. A developmental description and an identification to the species level are also provided. Temperature, salinity and turbidity at collection sites ranged from ca. 17 to 34°C, from ca. 0 to 16 psu and from 0 to 524 NTU, respectively. A peak catch per unit effort was found in April and at the stations at which salinity was about 0.6 to 2.5 psu. In stations where salinity was greater than 17 psu or at 0 psu, few specimens were collected. Examination of distribution indicated a tendency of the species to be present along the estuary, with the center part as the main habitat of this species. The function of an estuarine environment for fishes, which inhabit fresh and brackish waters, is discussed.
