Background: Leukemia is one of the most occurring haematological pathologies in the world which develops due to the impairment in the hematopoietic machinery. Cellular death via apoptosis is severely impaired in this pathophysiological condition and leads to the progression of the disease. Objective: We have tried to unearth the efficacy of Withanolide D, a steroidal lactone derived from Withania somnifera or Ashwagandha on some of the apoptotic machinery components, i.e. TERT, BCL2 and PUMA in the experimental leukemic mice. Materials and Methods: LD50 and EC50 values of Withanolide D were estimated. Three groups of animals were taken for experimental purpose i.e. Group I = Leukemic, (L); Group II = Control, (C); Group III = Leukemic treated with Withanolide D, (L + WD). Group III received Withanolide D via oral route and other two groups received equal volume of distilled water. Various cytological, immunofluorescence and flow cytometric studies were taken into consideration post administration. Result: Leukemic group showed increased cellular proliferation and decreased cellular death as compared to control. Post Withanolide D administration TERT, BCL2 and PUMA expression started to shift towards normal status. This shift in the expressional values of increased apoptosis rate and decreased cellular proliferation revealed by cytological, immunofluorescence studies and flow cytometric investigations. Conclusion: As Withanolide D decreased the suppression of apoptosis and impaired the progression of the disease, so, we can conclude that Withanolide D of Ashwagandha may hold a promise towards a new therapeutic strategy in leukemia.

COVID 19, which has lead to the death of millions of people, is still spreading worldwide. The development of any new drug after proper trial is much time consuming.  This present global pandemic situation has lead the researchers around the world to run behind various existing antiviral and immunomodulatory natural compounds to overcome this contagious disease.  Withania somnifera (ashwagandha) that is being used in Ayurvedic medicine for several ailments since several years is also said to pocess anti viral activity. Thus many of its metabolites are being studied individually for its efficacy against the dreadful disease. Withaferin A, a steroidal lactone from ashwagandha is one such prime metabolite which beyond acting as an antioxidant or antimicrobial agent, is also said to pocess anti-inflammatory to anti carcinogenic properties.  Thus because of its wide spectrum of medicinal properties it has now become an attractive drug candidate for several preclinical studies. This increase in the demand for withaferin A has chanelled its way towards in vitro propagation of the plant Withania somnifera and trials on various strategies to increase the yield in terms of plant biomass as well as the withaferin content in the plants thus making it a better alternative to field grown plants. Thus this article reviews in depth on the important medicinal properties of withaferin A, its demand in Ayurveda industry and the in vitro strategies being carried out to overcome the demand.

Abstract The outbreak and continued spread of the novel coronavirus disease 2019 (COVID-19) is a preeminent global health threat that has resulted in the infection of over 6 million people worldwide. In addition, the pandemic has claimed the lives of over 350,000 people worldwide. Age and the presence of underlying comorbid conditions have been found to be key determinants of patient mortality. One such comorbidity is the presence of an oncological malignancy, with cancer patients exhibiting an approximate two-fold increase in mortality rate. Due to a lack of data, no consensus has been reached about the best practices for the diagnosis and treatment of cancer patients. Interestingly, two independent research groups have discovered that Withaferin A (WFA), a steroidal lactone with anti-inflammatory and anti-tumorigenic properties, may bind to the viral spike (S-) protein of SARS-CoV-2. Further, preliminary data from our research group has demonstrated that WFA does not alter expression of ACE2 in the lungs of tumor-bearing female mice. Downregulation of ACE2 has recently been demonstrated to increase the severity of COVID-19. Therefore, WFA demonstrates real potential as a therapeutic agent to treat or prevent the spread of COVID-19 due to the reported interference in viral S-protein to host receptor binding and its lack of effect on ACE2 expression in the lungs.
