Evaluation of the Percentage of Peripheral T Cells with Two Different T Cell Receptor α-Chains and of their Potential Role in Autoimmunity

The current study examines the possibility that the TCR repertoire of L3T4-, Lyt-2- cells in lpr/lpr mice is enriched for self-reactive specificities. T cells utilizing V beta 17a and V beta 8.1 gene products have been shown to be clonally eliminated in nonautoimmune mice expressing I-Ek and Mlsa/H-2k, respectively, because of their potential self reactivity. We quantitated these V beta specificities in lymph nodes and spleens of lpr/lpr mice. The results indicate that lpr-dependent L3T4-/Lyt-2- T cells, similar to normal peripheral T cells, have undergone a repertoire modification such that potential self-reactive V beta specificities have been eliminated. Evidence for tolerance in this population provides insight into the development of these aberrant cells, and may also have important implications for normal T cell development in the thymus.

Recognition of conventional foreign antigen by T cells is determined by the expression of multiple variable regions of both alpha and beta chains of the T cell receptor (TCR) alpha/beta heterodimer. In contrast, there exists a class of antigens that appears to interact with the TCR alpha/beta heterodimer through the variable region on the beta chain (V beta), independent of other TCR components, a property that has led to their designation as superantigens. The goal of the present study was to analyze V alpha use in V beta 6+ T cells responsive to the superantigen, Mlaa. Results indicate that while deletion of T cells expressing V beta 6 in Mlsa-expressing mice is essentially complete and therefore appears to occur regardless of V alpha usage, in vitro Mlsa stimulation of T cells from Mlsa-negative mice results in significant skewing of V alpha use among responding V beta 6+ T cells. This indicates that V alpha expression influences recognition of the superantigen, Mlsa by mature peripheral T cells.

CD3-ζ/η–deficient mice have small thymuses containing cells that show a profound reduction in the surface levels of T cell receptors and terminate their differentiation at the CD4+CD8+ stage. Rather unexpectedly, CD3− or very low single positive T cells accumulate over time in the spleen and lymph nodes of CD3-ζ/η–deficient mice after a process dependent on MHC expression. Fusion of these peripheral T cells with a CD3-ζ–positive derivative of the BW5147 TCR-α−/β− thymoma resulted in hybridomas that do express an heterogeneous set of T cell receptor α/β dimers at their surface and at density comparable to those found in hybridomas derived from wild-type peripheral T cells. We have investigated the specificities of these T cell receptors using spleen cells from congenic and mutant mouse strains, and showed that the majority of them readily recognized self-MHC class I or class II molecules. These results demonstrate that by increasing the density and/or output of the T cell receptors expressed in peripheral T cells, one can confer them with the capacity to respond to normal density of self-MHC molecules.
