AbstractThe unique Himalayan ecosystems are repositories to the wild populations of diverse flora and fauna. The high value medicinal and aromatic plant species (MAPs) are an example of the same. Since time immemorial, these MAPs have been traditionally used by the local inhabitants and have eventually developed a high market value all over the world. Increasing market demand engenders over-extraction of species, unsustainable collection further catalyses decline in wild populations. The current communication raises high conservation concern on the rapid population decline of Fritillaria cirrhosa D.Don in the Western Himalaya. Harvested and traded with a new trade name i.e., ‘Jangli lehsun’ probably to disguise common Allium species, the species is facing tremendous decline in wild populations due to its illegal harvesting and trade in Himachal Pradesh. Further, F. cirrhosa faces threat due to unorganized, over-extraction, unsustainable and premature harvesting of the bulbs, coupled with illegal hidden markets functioning parallelly. Considering that this valuable species is under multiple threats being a medicinally important plant, priority should be given for its conservation through in-situ such as identification of medicinal plant conservation areas and ex-situ methods for its propagation and multiplication. Further, to ensure the long-term conservation of Fritillaria cirrhosa, prioritized conservation strategies such as strengthening of the Biodiversity Management Committees, capacity building through awareness programs for the key stakeholders and sustainable harvesting would be the practical solution.

Investigations were carried out to study the effects of light-emitting diode (LED) lights on growth and development of isosteroidal alkaloids in embryogenic calli of Fritillaria cirrhosa D. Don, an important traditional Chinese medicine herb. Calli were cultured in glass bottles, each containing 100 mL of Murashige and Skoog’s basal medium supplemented with 2% sucrose and 0.4% gellan gum powder, a gelling agent. These bottles were incubated in a specially designed plant growth chamber equipped with eight different LED lights consisting of single or combinations of four different light spectra emitting blue (450 nm), green (525 nm), red (660 nm), and far-red (730 nm) light. After three months of incubation, morphological changes in embryogenic calli were recorded, and LC-MS/MS analysis of cultures was carried out for peimisine, sipeimine, peiminine, and peimine. The highest number of somatic embryos and the maximum fresh weight was recorded in calli incubated under red (9R), infrared (9IR), and a combination of red+blue+infrared (3R3B3IR), respectively, in decreasing order. The highest contents of peimisine, peiminine, and peimine were recorded under red (9R) and infrared (9IR) lights, respectively. Eight LED lights had significant effects on the morphogenesis of embryogenic calli of F. cirrhosa D. Don and contents of isosteroidal alkaloids.

The genus Fritillaria belongs to the widely distributed family Liliaceae. The bulbs of Fritillaria ussuriensis and Fritillaria cirrhosa are valuable herbaceous medicinal ingredients. However, they are still used indiscriminately in herbal medicine. Identification and molecular phylogenic analysis of Fritillaria species is therefore required. Here, we report the complete chloroplast (cp) genome sequences of F. ussuriensis and F. cirrhosa. The two Fritillaria cp genomes were 151,524 and 151,083 bp in length, respectively, including a pair of inverted repeat regions (52,678 and 52,156 bp) separated by a large single copy region (81,732 and 81,390 bp) and small single copy region (17,114 and 17,537 bp). A total of 111 genes in F. ussuriensis and 112 in F. cirrhosa comprised 77 protein-coding genes in F. ussuriensis and 78 in F. cirrhosa, 30 tRNA genes, and four rRNA genes. The gene order, content, and orientation of the two Fritillaria cp genomes exhibited the general structure of flowering plants, and were similar to those of other Fritillaria species. Comparison of the six Fritillaria species’ cp genomes indicated seven highly divergent regions in intergenic spacers and in the matK, rpoC1, rpoC2, ycf1, ycf2, ndhD, and ndhF coding regions. We established the position of the six species through phylogenic analysis. The complete chloroplast genome sequences of two Fritillaria species will be useful genomics resources for identification of Fritillaria species and for studying the phylogenetic relationship among Fritillaria species within the Liliaceae family.
