Changes of surface charge and extracellular polymer (ECP) content were investigated in batch experiments for three anaerobic sludges, each of which had been enriched at 35°C and pH 639-7.3 for more than 40 batches using propionate, butyrate and glucose, individually, as the sole substrate. Results showed that both ECP and the negative surface charge were dependent on the growth phase of microorganisms. They increased at the beginning of all batches when the microorganisms were in the prolific-growth phase, having high substrate concentration and food-to-microorganisms ratio. Both later gradually returned to their initial levels when the microorganisms were in the declined-growth phase, as the substrate became depleted. The negative surface charge increased linearly with the total-ECP content in all series with slopes of 0.0187, 0.0212 and 0.0157 meq/mg-total-ECP for sludge degrading propionate, butyrate and glucose, respectively. The change of surface charge for the first two sludges was mainly due to the increase of proteinaceous fraction of ECP; but, for glucose-degrading sludge, that could be due to the increases of both proteinaceous and carbohydrate fractions of ECP. The negative-charged nature of anaerobic sludge implies that cations should be able to promote granulation of anaerobic sludge.

The aquatic plant Pistia stratiotes L. is environmentally hazardous and requires effective methods for its utilization. The harmfulness of these plants is determined by their excessive growth in water bodies and degradation of local aquatic ecosystems. Mechanical removal of these plants is widespread but requires fairly resource-intensive technology. However, these aquatic plants are polymer-containing substrates and have a great potential for conversion into bioenergy. The aim of the work was to determine the main patterns of Pistia stratiotes L. degradation via granular microbial preparation (GMP) to obtain biomethane gas while simultaneously detoxifying toxic copper compounds. The composition of the gas phase was determined via gas chromatography. The pH and redox potential parameters were determined potentiometrically, and Cu(II) concentration photocolorimetrically. Applying the preparation, high efficiency of biomethane fermentation of aquatic plants and Cu(II) detoxification were achieved. Biomethane yield reached 68.0 ± 11.1 L/kg VS of Pistia stratiotes L. biomass. The plants’ weight was decreased by 9 times. The Cu(II) was completely removed after 3 and 10 days of fermentation from initial concentrations of 100 ppm and 200 ppm, respectively. The result confirms the possibility of using the GMP to obtain biomethane from environmentally hazardous substrates and detoxify copper-contaminated fluids.

Production of bio-based materials in biorefineries is coupled with the generation of organic-rich process-wastewater that requires further management. Anaerobic technologies can be employed as a tool for the rectification of such hazardous by-products. Therefore, 5-hydroxymethylfurfural process-wastewater and its components were investigated for their biodegradability in a continuous anaerobic process. The test components included 5-hydroxymethylfurfural, furfural, levulinic acid, and the full process-wastewater. Each component was injected individually into a continuously operating anaerobic filter at a concentration of 0.5 gCOD. On the basis of large discrepancies within the replicates for each component, we classified their degradation into the categories of “delayed”, “retarded”, and “inhibitory”. Inhibitory represented the replicates for all the test components that hampered the process. For the retarded degradation, their mean methane yield per 0.5 gCOD was between 21.31 ± 13.04 mL and 28.98 ± 25.38 mL. Delayed digestion was considered adequate for further assessments in which the order of conversion to methane according to specific methane yield for each component from highest to lowest was as follows: levulinic acid > furfural > 5-hydroxymethylfurfural > process-wastewater. Disparities and inconsistencies in the degradation of process-wastewater and its components can compromise process stability as a whole. Hence, the provision of energy with such feedstock is questionable.
