Effects of the addition of Calanopia elliptica, Artemia franciscana, and Brachionus rotundiformis in a nursery biofloc system on water quality, growth, gut morphology, health indices, and transcriptional response of immune and antioxidant-related genes in Penaeus vannamei

Keberhasilan kegiatan budidaya perikanan harus ditunjang dengan ketersediaan benih yang berkesinambungan. Oleh karena itu, diperlukan juga ketersediaan pakan alami larva berupa rotifera (Brachionus rotundiformis). Desain percobaan berupa rancangan faktorial dengan dua faktor dan lima ulangan diaplikasikan dalam penelitian ini. Sebagai perlakuan berupa suhu ruang penyimpanan (suhu kamar, suhu ruang AC, dan suhu refrigerator/lemari es) dan pakan mikroalga konsentrat (monospesies dan multispesies). Bakteri probiotik juga digunakan sebagai pengontrol kualitas air. Spesies mikroalga yang digunakan adalah Nannochloropsis sp., Dunaliella sp., Isochrysis sp., dan Pavlova sp. Parameter yang diukur adalah kelimpahan rotifera dan parameter kualitas air media kultur (pH, salinitas, DO, dan NH3). Analisis data terdiri atas analisis regresi, analisis ragam, dan uji keparalelan. Hasil pengukuran parameter kualitas air selama penyimpanan menunjukkan kondisi media yang relatif stabil dan merupakan kisaran optimum bagi pertumbuhan B. rotundiformis. Kelimpahan maksimum tertinggi dari B. rotundiformis baik pada perlakuan pakan monospesies maupun multispesies alga adalah pada suhu kamar. Dari interaksi kedua perlakuan, diperoleh kelimpahan akhir tertinggi pada suhu ruang AC–pakan multispesies. Hal ini menunjukkan bahwa rotifera dapat disimpan lebih lama pada suhu ruang AC dengan pemberian pakan multispesies alga.The success of any aquaculture practices should be supported by sustainable supply of fish fry. Therefore, the availability of rotifers (Brachionus rotundiformis) as natural feed for fish larvae is required. The research was arranged in factorial design with two treatments and five replications. Treatments consisted of different room storage temperatures (refrigerator, room temperature, and room with air conditioner/AC) and microalgae concentrate added as rotifer feed (monospecies and multispecies algae). Probiotic bacteria was used to control water quality. Mikroalgae species consisted of Nannochloropsis sp., Dunaliella sp., Isochrysis sp., and Pavlova sp. Parameters measured were rotifer density and water quality of rotifer media (pH, salinity, DO, and NH3). Data analysis included regression analysis, analysis of varians and parallel testing. The results of water quality parameters during rotifer storage showed that media conditions were relatively stable and optimal for B. rotundiformis growth. The result of treatments interaction showed that the highest maximum density of rotifer at the end of the research was achieved by rotifer stored in air conditioned room fed with multispecies algae. This showed that rotifers can be stored longer in room storage with AC and fed by multispecies algae.

The biofloc system is an ecologically sustainable shrimp culture system. The conglomerates of beneficial bacteria, algae and protozoa in pond water serve as a water quality management system and as a feed additive to the shrimps. This study aimed to characterise the microbial communities associated with the biofloc pond water and the surface of Penaeus vannamei Boone, 1931, reared in it using the Illumina Miseq sequencing technology. The multiple alpha diversity measures indicated the shrimp surface samples to be richer in diversity than the pond water samples. Analysis of the bacterial community revealed that Proteobacteria, Bacteroidetes, Planctomycetes and Cyanobacteria formed the principal phyla. There was a shift in the relative abundance of bacterial communities at each time point. The operational taxonomic units (OTU) analyses revealed that 18.38 % OTUs were shared by the pond water samples, the shrimp surface samples shared 29.35 % at the three different time points. PICRUST analysis revealed that the bacterial communities in the biofloc rearing water, and shrimp surface, were likely involved in intensive microbial metabolism and core housekeeping functions. The information generated will help understand the bacterial community composition associated with optimal water quality and shrimp health in a biofloc culture system.

Title (Bahasa Indonesia): Viabilitas rotifer Brachionus rotundiformis strain tumpaan pada salinitas berbeda The purpose of the research was to analyze the viability of eggs of Tumpaan-strained rotifer, Brachionus rotundiformis, at different salinities (10 and 20 ppt). Rotifer collection was done in the area of reclamation plan and household wastewater disposal located in the coastal village of Tumpaan 1, South Minahasa Regency. At the time of sample collection, water quality parameters of the sampling site were also measured. After multiplication through clone culture in the salinity of 10 ppt and 20 ppt, the viability of the rotifer was then tested by daily observing the number of live rotifers, the number of eggs carried and the number of youngsters produced. The data were then calculated using the life table method. Results showed that water quality of the rotifer collection site is suitable for the rotifer to live. The rotifers held in 10 ppt salinity had higher survivorship and population growth (fertility rate and Ro) than those in 20 ppt salinity. This reflects that beside the quality of feed, rotifer growth is affected by salinity as well. Tujuan penelitian ini untuk menganalisis viabilitas tetasan telur dari rotifer Brachionus rotundi-formis strain Tumpaan pada salinitas berbeda (10 dan 20 ppt). Pengambilan rotifer dilakukan di suatu areal rencana reklamasi dan tempat pembuangan penelitian air limbah rumah tangga bertempat di pesisir Desa Tumpaan Satu Kabupaten Minahasa Selatan. Pada saat pengambilan hewan uji, parameter kualitas air di lokasi sampling juga diukur. Setelah diperbanyak dengan kultur klon pada salinitas 10 dan 20 ppt, rotifer kemudian diuji viabilitas dengan melakukan pengamatan setiap hari data jumlah rotifer yang hidup, jumlah telur yang dibawah dan jumlah anak yang dihasludgean. Data kemudian dihitung menggunakan metode life table.Hasil pengukuran kualitas air dari perairan tempat pengambilan rotifer termasuk layak untuk kehidupan rotifer. Rotifer yang diuji pada salinitas 10 ppt memiliki kemampuan hidup serta pertumbuhan populasi (nilai Ro dan laju fertilitas) lebih tinggi dibandingkan pada salinitas 20 ppt. Hal ini menunjukkan bahwa selain kualitas pakan, pertumbuhan rotifer juga dipengaruhi oleh salinitas.
