Watermelon (Citrullus lanatus) consists of high moisture content and is favoured for its juice products. The popular fruit has a tempting taste, sweet aroma and attractive flesh colour. It is enriched with phytochemicals and antioxidant properties that are beneficial to human health. Due to convenience, the majority of individuals are likely to consume watermelon juice. However, little is known about the fruit juice storage and temperatures that may affect its beneficial properties. This study investigated the effect of storage temperature at room temperature, refrigerator cold, refrigerator freeze and freeze-dried, and analyzed the juice physico-chemicals (weight loss, pH, ash, moisture, total soluble solid, browning and turbidity), phytochemicals (total phenolic, total flavonoid, lycopene and β-carotene) and antioxidant scavenging activities during 9 days of storage. The results showed that watermelon juice was affected by storage temperatures and conditions with significant changes in physico-chemical appearance and decrease in total phytochemical content, thus consequently affecting their antioxidant activities during 9 days of storage. Although fresh watermelon juice can be consumed for its high nutritional values, freeze-drying is the preferable technique to retain its benefits and to delay juice degradation.

Background: Epilepsy is a neurologic condition that is occurs globally and is associated with various degrees of seizures. Levetiracetam is an approved drug that is commonly used to treat seizures in juvenile epileptic patients. Accurate quantification of the drug’s active compound and determining its stability in the stomach after oral administration are important tasks that must be performed. Results & methodology: Levetiracetam was extracted from drug samples and quantified by gas chromatography mass spectrometry using calibration standards. Stability of levetiracetam was studied under various storage conditions and in simulated gastric conditions. The calibration plot determined for levetiracetam showed good linearity with a coefficient of determination value of 0.9991. The limits of detection and quantification were found to be 0.004 and 0.014 μg·ml-1, respectively. The structural integrity of levetiracetam did not change within a 4-h period under the simulated gastric conditions, and no significant degradation was observed for the different storage temperatures tested. Discussion & conclusion: An accurate and sensitive quantitative method was developed for the determination of levetiracetam in drug samples. The stability of the drug active compound was monitored under various storage and gastric conditions. The levetiracetam content determined in the drug samples were within ±10% of the value stated on the drug labels.

AbstractFresh fruits like bananas are very susceptible to mechanical damage during postharvest handling which can result in a substantial decline in quality. The study aims to evaluate the effect of bruise damage and storage temperatures on the quality of banana fruits after 48 h storage. Each ‘Grand Naine’ banana fruit was impacted once by using a drop impact test using three different heights (10, 30, and 50 cm) and storage temperatures (13 and 22 °C) after 48 h of storage. Different quality analyses were measured like bruise measurements (impact energy, bruise area, bruise volume, and bruise susceptibility), weight loss, total soluble solids (TSS), color (L*, a*, b*, hue°, chroma, yellowness index, yellowness value) headspace gases (respiration and ethylene production rate). The results showed that bruise measurements (bruise area, bruise volume, and bruise susceptibility) were highly affected by drop height. The quality parameters like weight, color, total soluble solids and headspace gases were affected by drop height and storage condition. Weight loss, total soluble solids, respiration rate, and ethylene production rate increased as drop height and storage temperature rise. Storage at ambient conditions (22 °C) accelerated bruising occurrence in banana fruits. Fewer changes were observed after 48 h of storage. The least value of yellowness index was observed on the non-bruised banana fruits (84.03) under 13 °C storage conditions. The findings of the study can provide baseline data to understand the mechanical damage mechanism on fruit quality, hoping to create awareness and educate farming communities and consumers. Storage temperature management is another approach that needs to be followed to reduce the occurrence of mechanical damage in fresh produce.

Anaerobic digestates derived from agricultural mesophilic biogas plants are mainly used as organic fertilizers. However, animal derived pathogens could persist in the anaerobic digestates (ADs) posing an epidemiological risk. The present study investigated whether storage of ADs could reduce Salmonella Typhimurium, Listeria monocytogenes, and ESBL carrying Escherichia coli and whether reduction rates are dependent on temperature and substrate. Quantified bacterial suspensions were used to inoculate ADs derived from five biogas plants using different input materials to investigate the substrate dependence of the pathogen reduction. ADs were stored over six months with four different temperature profiles each representing six consecutive months, and, thus, the four seasons. Pathogen reduction during storage was shown to be strongly dependent on the temperature but also on the type of AD. This influence was higher at low temperatures. At higher temperatures (spring and summer profiles), a 5-log reduction was achieved after twelve weeks for S. Typhimurium, after twenty weeks for E. coli (ESBL) and after twenty-four weeks for L. monocytogenes in all ADs, respectively. In contrast at lower temperatures (autumn and winter profiles), a 5-log reduction was reached after twenty-four weeks for S. Typhimurium and not reached for ESBL-E. coli and L. monocytogenes. In conclusion, storing the ADs after the biogas process improves the hygienic quality and reduce the risk of introducing pathogens to the environment, but each case should be evaluated individually considering the composition of the ADs and the storage temperatures.

Tacrolimus is an immunosuppressant used to treat a large variety of inflammatory or immunity-mediated ophthalmic diseases. However, there are currently no commercial industrial forms available that can provide relief to patients. Various ophthalmic formulations have been reported in the literature, but their stability has only been tested over short periods. The objective of this study was to evaluate the physicochemical stability of a preservative-free tacrolimus formulation (0.2 and 1 mg/mL) at three storage temperatures (5 °C, 25 °C and 35 °C) for up to nine months in a multidose eyedropper. Analyses performed were the following: visual inspection and chromaticity, turbidity, viscosity, size of micelles, osmolality and pH measurements, tacrolimus quantification by a stability-indicating liquid chromatography method, breakdown product research, and sterility assay. In an in-use study, tacrolimus quantification was also performed on the drops emitted from the eyedroppers. All tested parameters remained stable during the nine month period when the eyedrops were stored at 5 °C. However, during storage at 25 °C and 35 °C, several signs of chemical instability were detected. Furthermore, a leachable compound originating from a silicone part of the eyedropper was detected during the in-use assay. Overall, the 0.2 mg/mL and 1 mg/mL tacrolimus ophthalmic solutions were physicochemically stable for up to nine months when stored at 5 °C.

ABSTRACT: Eggshell quality is directly linked to the good acceptability of the product by consumers, whereas the industry is concerned with economic losses. Prolonged periods of laying, diseases, nutrition, bird age and genetics can influence egg quality. This study aimed to examine the influence of the environment, bird age and storage period on the physical characteristics of the shell of eggs from commercial laying hens. The experiment involved 288 eggs from Hy-line hens, which were distributed in a completely randomized design with a triple 3×2×4 factorial arrangement represented by three ages (31, 62 or 88 weeks), two storage temperatures (ambient or refrigerated), and four storage periods (one, 10, 20 or 30 days), totaling 24 treatments. Four replications were used with three eggs each. Eggshell strength, percentage and thickness were measured. Differences were observed, with the eggs stored at room temperature exhibiting lower shell strength. Laying eggs at an advanced age (88 weeks) resulted in decreased eggshell strength as compared with the other ages, whereas the eggs from young layers (31 weeks) had a thicker shell. The advancing age of the laying hens reduced eggshell quality, regardless of the storage environment. Nonetheless, the eggs stored in the refrigerator showed better eggshell quality and integrity than those stored at ambient temperature.

Abstract A novel self-supporting multi-layer magnetorheological elastomer-based (MRE-based) composite with large magnetic field-induced responsiveness has been designed and fabricated. We characterized its morphological properties, evaluated the impact of fabrication conditions on its field-induced responsiveness, investigated attenuation of its field-induced responsiveness under different storage temperatures along with time and analyzed this mechanism from the perspective of rheology. The results showed that the MRE-based composite had homogeneous dispersing of the magnetic fillers and a clear interface between different layers. The field-induced responsiveness of the MRE-based composite could be affected by the fabrication conditions, and it attenuated at different rates when subjected to different storage temperatures along with time; its attenuation period lasted a few days under room temperature while over one month under low temperature (4℃). The rheological analysis results indicated a long-term cross-linking process over the storage period along with the attenuation of field-induced responsiveness, which might lead to increasing elasticity (indicated by the loss factor tan δ) and rigidity (indicated by the storage modulus G') of the MRE-based composite along with the storage period. What's more, emerging feature of Payne effect could be found on MRE-based composite during cyclic shear, which indicated decline of the mechanical properties due to strain-induced inherent friction. On the other hand, the iron fillers in MRE layer could enhance the shear modulus and lead to MR effect (up to 187%) for the whole composite, which benefits to the magnetic field-induced responsiveness, due to the relative strengthen of the MRE layer against the assist layer. This work presents a better understanding on the attenuation of the field-induced responsiveness, which is important for the future application of the MRE-based composite.
