Isolation and identification of anti-inflammatory compounds from ethyl acetate fraction of Ecklonia stolonifera and their anti-inflammatory action

Background: Pentacyclic triterpenoids are a biologically active class of phytoconstituents with diverse pharmacological activity including anti-inflammatory action. Objective: In the current study, we isolated 3-Acetylmyricadiol, a pentacyclic triterpenoid, from the ethyl acetate bark-extract of Myrica esculenta and evaluated it for anti-inflammatory potential. Methods: The ethyl acetate bark-extract of the M. esculenta was subjected to column chromatography to isolate 3-Acetylmyricadiol. MTT assay was performed to check cell viability. The production of proinflammatory mediators like Nitric oxide, IL-6, TNF-α was observed after administration of 5, 10, 20 μM of 3-Acetylmyricadiol in LPS-activated Raw 246.7 macrophages by the reported methods. Results: MTT assay indicated more than 90% cell viability up to 20 μM of 3-Acetylmyricadiol. The administration of 3-Acetylmyricadiol inhibited the production of Nitric oxide, IL-6, TNF-α in a dose-dependent manner significantly in comparison to LPS treated cells. The maximum effect was observed at 20 μM of 3-Acetylmyricadiol which resulted in 52.37, 63.10, 55.37 % inhibition of Nitric oxide, IL-6, TNF-α respectively. Conclusion: Our study demonstrated the anti-inflammatory action of 3-Acetylmyricadiol and can serve as a potential candidate in the development of the clinically efficient anti-inflammatory molecule.

Hedyotis diffusa Willd and Scutellaria barbata D. Don (HDSB) were the core couplet in medicines that were commonly used for the purpose of anti-inflammation and anticancer treatments in China. However, biological properties of this couplet have not been fully elucidated. In this study, we screened fractions of HDSB for their anti-inflammatory activities and explored pertinent molecular mechanisms on murine macrophage RAW264.7 cell model. Ethyl acetate fraction from the aqueous extract of the couplet at equal weight ratio (EA11) showed the strongest inhibition of the nitrite accumulation in supernatant of RAW264.7 cells stimulated with lipopolysaccharide (LPS)/interferon-γ (IFN-γ). In addition, EA11 inhibited iNOS and IL-1β expression in a concentration-dependent manner while promoting the expression of HO-1 and PPAR-γ. Anti-inflammatory capability is most likely facilitated by its inhibitory effect on JNK signaling pathway and miR-155 expression. This study suggests that EA11 may be represented as a potential anti-inflammatory therapeutic candidate.

Abstract Viscum album L. has been used in the indigenous systems of medicine for treatment of headache and some inflammatory diseases. In order to evaluate this information, antinociceptive and anti-inflammatory activities of the five flavonoids (5,7-dimethoxy naringenin or 4′,6′- dimethoxy chalcononaringenin) derivatives, isolated from the ethyl acetate fraction of the extract from V. album ssp. album, were investigated, namely 5,7-dimethoxy-flavanone-4′-O- β-ᴅ-glucopyranoside (1), 2′-hydroxy-4′,6′-dimethoxy-chalcone-4-O-β-ᴅ-glucopyranoside (2), 5,7-dimethoxy-flavanone-4′-O-[2″-O-(5‴-O-trans-cinnamoyl)-β-ᴅ-apiofuranosyl]-β-ᴅ-glucopyranoside (3), 2′-hydroxy-4′,6′-dimethoxy-chalcone-4-O-[2″-O-(5‴-O-trans-cinnamoyl)-β-ᴅ-apiofuranosyl]- β-ᴅ-glucopyranoside (4), 5,7-dimethoxy-flavanone-4′-O-[β-d-apiofuranosyl- (152)]-β-ᴅ-glucopyranoside (5). For the antinociceptive activity assessment the p-benzoquinone- induced writhing test and for the anti-inflammatory activity the carrageenan-induced hind paw edema model in mice were used. The ethyl acetate fraction in a dose of 250 mg/kg as well as compounds 2 and 5 in a 30 mg/kg dose were shown to possess remarkable antinociceptive and anti-inflammatory activities per os without inducing any apparent acute toxicity as well as gastric damage

There has been burgeoning interest in plant-based feed additives following restrictions placed on the use of antibiotic feed additives in many countries. Phytogenic feed additives are recommended to have a range of useful properties to support the growth and development of poultry to a similar level as that obtained by supplementing feed with antibiotics. The aim of this study was to evaluate the antibacterial, anti-lipoxygenase and antioxidant activity, and in vitro safety of fractions and isolated compounds from leaves of Senna singueana. Antibacterial activities of the fractions and isolated compounds were determined against a panel of bacteria using a two-fold serial microdilution assay and qualitative bioautography assays. Anti-lipoxygenase activity was evaluated using the ferrous oxidation-xylenol orange (FOX) method. Antioxidant activity was assessed qualitatively and quantitatively using radical scavenging assays. Dichloromethane and ethyl acetate fractions from solvent-solvent partitioning had the best antibacterial activity with MIC values ranging from 156 to 313 μg/ml. Fractions obtained from column chromatography had significant to weak antibacterial activity with MIC values ranging from 50 to 1,250 μg/ml. Bioautography showed clear bands of bacterial inhibition, indicating the presence of a number of active compounds in several fractions. The ethyl acetate fraction and all the tested column fractions had potent anti-lipoxygenase activity with IC50 values of ≤2.5 μg/ml which were lower than that of quercetin (positive control), indicating anti-inflammatory potential. The ethyl acetate fraction and several column fractions had powerful antioxidant activity with IC50 values of ≤5 μg/ml in the ABTS assay. Cytotoxicity values against Vero kidney cells ranged from LC50 = 40.0–989.3 μg/ml. Bioassay-guided fractionation led to the isolation and identification of a known bioactive compound, luteolin. S. singueana is a promising candidate for the development of poultry phytogenic feed additives.
