Efficient and reliable propagation and rooting of foetid juniper (Juniperus foetidissima Willd.), as an endangered plant under in vitro condition

Meizotropis pellita is a rare, endangered and threatened plant species found in Patwadanger forest. In nature this species grows from rootstock in precise and sensitive habitats. However, phenolic compounds provoke browning reaction which is responsible for lethal browning in plant tissue culture and hindering in vitro germination. The aim of this study was to minimize the effect of phenolic compound during seed germination. We evaluate different antibrowning treatments in Meizotropis pellita seeds. The seeds were pretreated in Polyvinylpyrrolidone and then treated seeds were cultured on MS media and sterilized seeds were directly inoculated on PVP containing MS media. These culture media incubate under dark and light conditions at different durations respectively. Our results showed that addition of antibrowning agent, PVP inhibit onset of browning. Dark and light condition also played a crucial role on onset of browning. Delayed and low intensity of browning were observed in case of seeds that were inoculated on MS media containing PVP and were kept in dark. The present research offers a positive in vitro seed germination protocol for Meizotropis pellita conservation.

Protocols for the in vitro propagation of two endangered plant species in the Czech Republic, Gentiana verna L. and Jurinea cyanoides (L.) Rchb., were established. In G. verna, the induction of organogenesis on vegetative shoots was successful on the basal 6% agar WPM medium with 200 mg&middot;l<sup>&ndash;1</sup>of glutamine, 200 mg&middot;l<sup>&ndash;1 </sup>of casein hydrolysate, 30 g&middot;l<sup>&ndash;1 </sup>of sucrose, 0.2 mg&middot;l<sup>&ndash;1 </sup>of BAP, and 0.1 mg&middot;l<sup>&ndash;1 </sup>of IBA. The multiplication of primary explants was achieved on 6% agar basal MS medium with the same concentrations of substances mentioned above. In J. cyanoides, the induction of organogenesis and multiplication of its vegetative shoots were successful on MS medium with the same concentration of added substances used in G. verna. On the basal MS medium of 1/3 concentration with 3 mg&middot;l<sup>&ndash;1 </sup>of IBA 70&ndash;75% rooting efficiency of G. verna microcuttings and 40% rooting efficiency of J. cyanoides microcuttings were reached. The mortality during acclimatization did not exceed 20% for G. verna and 25% for J. cyanoides. &nbsp; &nbsp;
