Applications of Bacteriophage Cocktails to Reduce Salmonella Contamination in Poultry Farms

Current study investigated the occurrence of aflatoxin contamination in poultry feed and feed materials in different poultry farms and feed factories in Bangladesh. A total of 100 samples of finished feed and raw feed materials were collected and tested through direct competitive Enzyme-Linked Immunosorbent Assay (ELISA) for total aflatoxin detection. Overall, 97% samples (n=97/100) in our study, were found positive for aflatoxin contamination. Among finished feed categories, layer grower feed contained highest level of aflatoxin with a mean value of 21.64 ppb whereas layer feed was less susceptible for aflatoxin contamination (mean value 9.49 ppb). Between raw feed materials, maize samples were highly contaminated (n=15/15, 100%) with aflatoxin while 86.67% soybean samples showed positive result. Twenty one percent (21%) of the samples in our study contained aflatoxin concentration more than the acceptable limit employed by USFDA and many other countries which might pose severe health risk to poultry and human consumer. Abstract Background Several outbreaks of highly pathogenic avian influenza (HPAI) caused by influenza A virus of subtype H5N8 have been reported in wild birds and poultry in Europe during autumn 2020. Norway is one of the few countries in Europe that had not previously detected HPAI virus, despite widespread active monitoring of both domestic and wild birds since 2005. Results We report detection of HPAI virus subtype H5N8 in a wild pink-footed goose (Anser brachyrhynchus), and several other geese, ducks and a gull, from south-western Norway in November and December 2020. Despite previous reports of low pathogenic avian influenza (LPAI), this constitutes the first detections of HPAI in Norway. Conclusions The mode of introduction is unclear, but a northward migration of infected geese or gulls from Denmark or the Netherlands during the autumn of 2020 is currently our main hypothesis for the introduction of HPAI to Norway. The presence of HPAI in wild birds constitutes a new, and ongoing, threat to the Norwegian poultry industry, and compliance with the improved biosecurity measures on poultry farms should therefore be ensured. [MK1]Finally, although HPAI of subtype H5N8 has been reported to have very low zoonotic potential, this is a reminder that HPAI with greater zoonotic potential in wild birds may pose a threat in the future. The exploration of novel nonantibiotic interventions in the field, such as the use of bacteriophages, is necessary to avoid the presence of Salmonella. Bacteriophages are a group of viruses widely distributed in nature, strictly associated with the prokaryotic cell. Researchers have demonstrated the success of phage therapy in reducing Salmonella counts in poultry products. However, the impact that phage concentration in the environment may have against certain Salmonella serovars is not well understood. Therefore, the aim of this study was to assess Salmonella phage prevalence in commercial poultry farms in terms of the production type: layers or broilers. The most prevalent Salmonella serovars isolated in poultry production were used for phage isolation. Salmonella specific phages were isolated from 141 layer and broiler farms located in the Valencia region during 2019. Analysis of the samples revealed that 100% presented Salmonella phages, the most prevalent being against serovar S. Enteritidis (93%), followed by S. Virchow (59%), S. Typhimurium (55%), S. Infantis (52%) and S. Ohio (51%). These results indicate that poultry farms could represent an important source of Salmonella phages. Nevertheless, further studies are needed to assess the epidemiology of phages against other serovars present in other countries and their diversity from the point of view of molecular studies.
