Sex-specific effects of met-enkephalin treatment on vasopressin immunoreactivity in the rat supraoptic nucleus

Suckling in the lactating rat (days 6-10 of lactation) was used to characterize the effects of intracerebral oxytocin (Oxt) antisense oligonucleotide treatment on the hypothalamo-neurohypophysial system. Vehicle, mixed bases, Oxt antisense, or vasopressin antisense oligonucleotides (2.5 micrograms/0.5 microliter each) were infused directly into the left and right supraoptic nucleus (SON), 4 h before a litter of 10 pups was allowed to suckle for 30 min. In the Oxt antisense group, there was a significant reduction in the number of milk ejection reflexes (to 34.6 +/- 4.88%, P < 0.001), as well as in the weight gain of the litter (to 18.8 +/- 6.98%, P < 0.03) compared with vehicle (100%)-, mixed base-, or vasopressin antisense-treated animals, which did not differ from each other. The time to onset of suckling, however, was unchanged. Compared with presuckling values, plasma Oxt was significantly increased in all four groups 30 min after onset of suckling (vehicle, to 325 +/- 117%; mixed bases, to 258 +/- 48.2%; vasopressin antisense, to 330 +/- 55.7%), but this increase was less pronounced in Oxt antisense-treated rats (to 157 +/- 20.5%; P < 0.05 vs. vasopressin antisense). In contrast to these changes in neuroendocrine functions during suckling, Oxt as well as vasopressin content and Oxt immunoreactivity in posterior pituitaries and the SON, respectively, did not differ among groups. Our data indicate rapid and specific effects of an Oxt antisense oligonucleotide infused into the SON on neuroendocrine, suckling-related parameters that are not due to depleted stores of Oxt in the hypothalamo-neurohypophysial system.

Abstract. Legally irrelevant information like facial features is used to form judgments about rape cases. Using a reverse-correlation technique, it is possible to visualize criminal stereotypes and test whether these representations influence judgments. In the first step, images of the stereotypical faces of a rapist, a thief, and a lifesaver were generated. These images showed a clear distinction between the lifesaver and the two criminal representations, but the criminal representations were rather similar. In the next step, the images were presented together with rape scenarios, and participants (N = 153) indicated the defendant’s level of liability. Participants with high rape myth acceptance scores attributed a lower level of liability to a defendant who resembled a stereotypical lifesaver. However, no specific effects of the image of the stereotypical rapist compared to the stereotypical thief were found. We discuss the findings with respect to the influence of visual stereotypes on legal judgments and the nature of these mental representations.

FVIII was separated into low molecular weight FVIII (LMW FVIII) and high molecular weight FVIII (HMW FVIII) by gel chromatography in the presence of high salt concentration or by high salt elution of LMW FVIII from FVIII bound to anti HMW FVII-Sepharose. Specific antibodies were raised in rabbits against HMW FVIII and LMW FVIII. After removal of the contaminating anti HMW activities the rabbit anti LMW FVIII was still able to neutralize the FVIII coagulant activity of normal plasma and of IMW FVIII with canparable efficiency and it had no effect on the VIIIR:WF of FVIII in normal plasma or in HMW FVIII. Anti LMW FVIII does not bind to HMW FVIII and does not precipitate FVIII as tested by counter immunoelectrophoresis. Rabbit anti HMW FVIII precipitates FVIII in normal plasma, inhibits VIIIR:WF activity, while it has no effect on the FVIII coagulant activity of LMW FVIII. The coagulant activity of FVIII in normal plasma is slightly inhibited by anti HMW FVIII presumably by non-specific effects (sterical hindrance). It is concluded that inhibitory antibodies against VIII:C raised in rabbits recognize antigenic structures only present on LMW FVIII. Antibodies against HMW FVIII raised in rabbits appears to recognize structures only present on HMW FVIII.

ABSTRACT Acute stress, chronic stress plus hydration, cortisone treatment, cortisone treatment plus dehydration were used as methods of investigation and the relation between the neurosecretory activity of the hypothalamic supraoptic nucleus and paraventricular nucleus and the neurosecretory material around the hypophysial portal vessels of the median eminence on the one hand and the corticotrophin release on the other hand, has been studied in the rat. Whereas stress stimulates both the activity of the above mentioned cells of the hypothalamus and the ACTH release, stress plus hydration causes a depression of these hypothalamic cells but nevertheless causes a marked ACTH release. Cortisone inhibits the activity of the cells in the supraoptic nucleus and the paraventricular nucleus as well as the ACTH release whereas cortisone plus dehydration causes stimulation but inhibits the ACTH release. In some stress and cortisone treatment groups the variations of the neurosecretory material around the hypophysial portal vessels and of the ACTH release were found to show a correlation. It is concluded that the activity of the cells of the supraoptic nucleus and the paraventricular nucleus and the ACTH release do not seem to have any definite connection, whereas some observations indicate that the neurosecretory material in the region of the median eminence around the hypophysial portal vessels may have some significance in ACTH release.
