Anti-inflammatory effects of ethanolic extract of Rosmarinus officinalis L. and rosmarinic acid in a rat model of neuropathic pain

The use of alternative medicine to treat pain has been increased, and the combination of several medicinal plants for its relief is a common practice in traditional medicine. The present study is aimed at determining whether a combination of Syzygium aromaticum (S. aromaticum) and Rosmarinus officinalis L. (R. officinalis) potentiates their antinociceptive and anti-inflammatory effects. These effects were explored using the formalin and carrageenan assays in rats, respectively. Animals received local pretreatment with S. aromaticum oil or R. officinalis ethanolic extract (0.1–100 μg/paw) alone or combined in a 1 : 1 rate. Concentration-response curves were built to compare pharmacological responses after an individual administration of S. aromaticum, R. officinalis, or their combination. The pharmacological interaction was investigated by an isobolographic study using the EC50 of each component in a fixed 1 : 1 ratio. S. aromaticum and R. officinalis administered alone showed significant and concentration-dependent antinociceptive and anti-inflammatory effects, but R. officinalis was more potent than S. aromaticum in both the antinociceptive and anti-inflammatory effects (EC50 = 7.96 ± 0.6 μg/paw vs. EC50 = 41.6 ± 1.7 μg/paw; EC50 = 1.97 ± 0.3 μg/paw vs. EC50 = 26.9 ± 2.5 μg/paw, respectively). The isobolographic analysis of the combination of these species in a 1 : 1 ratio showed a synergistic interaction between S. aromaticum and R. officinalis since Zmix (experimental value) was lower than Zadd (theoretical value) for both the antinociceptive effect (Zmix = 0.45 ± 0.1 < Zadd = 24.8 ± 1.3) and the anti-inflammatory effect (Zmix = 5.2 ± 0.6 < Zadd = 14.4 ± 2.2), suggesting a potentiation for both pharmacological effects. These results prove evidence of the efficacy of mixture herb-herb used in folk medicine for pain therapy. It also emphasizes the requirement of pharmacological studies to explore the efficacy and safety of herb interactions.

In this study, the phytochemical composition and the possible prophylactic effects of an aqueous ethanol extract of Haematoxylon campechianum flowers (HCF) on peripheral neuropathic pain in a chronic constriction injury (CCI) rat model are investigated. Rats with induced CCI were subjected to neuropathic pain behaviour tests and evaluated by chemical, thermal, and mechanical sensation tests and functional recovery of the brain stem and sciatic nerve at 7- and 14-day intervals. The effect of the extract on acute pain and inflammation is also investigated. The extract exerted both peripheral and central analgesic and anti-inflammatory properties in addition to antipyretic effects that are clear from targeting COX, LOX and PGE. It was found that CCI produced significant thermal and mechanical hyperalgesia, cold allodynia and deleterious structural changes in both sciatic nerve and brain stem. Treatments with HCF extract significantly improved cold and thermal withdrawal latency, mechanical sensibility and ameliorated deleterious changes of sciatic nerve and brain stem at different dose levels. The extract also ameliorated oxidative stress and inflammatory markers in brain stem and sciatic nerve. It suppressed the apoptotic marker, p53, and restored myelin sheath integrity. The effects of HCF extract were more potent than pregabalin. Fifteen secondary metabolites, mainly gallotannins and flavonoids, were characterized in the extract based on their retention times and MS/MS data. The identified phenolic constituents from the extract could be promising candidates to treat neuropathic pain due to their diverse biological activities, including antioxidant, anti-inflammatory and neuroprotective properties.

Background: This study was a preliminary study aimed to investigating the in vitro activity of rosmarinic acid contained on Rosemary leaves (Rosemarinus officinalis L.) on the Candida albicans. Rosmarinic acid can be an inhibitor of the RTPase enzyme in the fungus Candida albicans. Objective: This study is expected to provide information in the development of science about the antifungal activity of ethanol extract of Rosemary leave (Rosmarinus officinalis L.) againsts C.albicans. The recent research was an laboratory experimental. Experiments on an ethanol extract of Rosemary leaves (Rosemarinus officinalis L.) and post-test design of the control group. Methods: The extraction method used in this study was a multilevel extraction method. The antifungal activity testing method uses discs and MIC methods. Replication was done six times at two concentration namely, 80% (b/v) and 100% for each measurement. Results: The data analysis was carried out using SPSS with 95% of confidence level with the Kruskal Wallis analysis then completed with the Mann Whitney U test. The results of the MIC experiment showed all the various concentrations of ethanol extract performed Candida albicans growth on the surface of the media. Extract with 80% (b/v) concentration showed greater antifungal activity. Furthermore the MIC was shown by 80% (b/v) concentration of the ethanol extract of Rosemary leaves against Candida albicans. The test results of the antifungal activity of the Rosemary leaf ethanol extract showed the presence of inhibitory diameter in the ethanol extract of the Rosemary leaf concentration of 80% (b/v) with a mean ± SD of 10.6 ± 0.57735 and RSD of 5.774%. Conclusion: The ethanol extract of Rosemary leaves (Rosmarinus officinalis L.) has an inhibitory effect on the fungus Candida albicans at a concentration of 80% (b / v) Keywords: Antifungal activity, Candida albicans, Rosemary, RTPase, ethanol extract
