ABSTRACT Introduction Laghupanchamula is a compound formulation prepared by combinations of roots of five herbs. Two variants of Laghupanchamula have been described in Ayurvedic classics where beside four common herbs fifth one is either Gokshura (Tribulus terrestris L.) or Eranda (Ricinus communis L.). The objective of the study is to make comparison between two variants of Laghupanchmula with respect to important group of phytochemicals and antioxidant activity to corroborate the science behind their therapeutic utility. Materials and methods Standard methods have been followed for quantitative determination of total quantity of phenols, tannins, flavonoids, and flavonols, and in vitro antioxidant activity in variants of Laghupanchmula formulations. Qualitative high-performance liquid chromatography (HPLC) analysis has also been performed to establish presence/absence of important chemical constituents in formulations. Results Additional quantity of phenols, tannins, flavonoids, and flavonols has been observed in Laghupanchamula variant containing Eranda than the variant containing Gokshura. Greater antioxidant activity has also been found in formulation containing Eranda. The HPLC analysis revealed the presence of shikimic acid, gallic acid, catechin in both formulations, but rutin has been found only in formulation containing Eranda. Conclusion From the results of experiments it has been observed that the formulation containing Eranda has more antioxidant activity as it contains more quantities of phenol, tannins, flavonoids, and flavonols than the formulation containing Gokshura. Objective: The present investigation was based on the design and evaluation of floating tablets of Pedalium murex and Tribulus terrestris fruit extracts as the medicinal source and the excipients that can enhance the bioactivity and prolong the gastric retention time.Methods: The fruit extracts were prepared from the powdered dry fruits of Pedalium murex and Tribulus terrestris using Soxhelt apparatus for 8 h. Direct compression technique was used for the formulation of polyherbal floating effervescence tablets which consists of different compositions of Hydroxy Propyl Methyl Cellulose (HPMC K4M, HPMC K15M), Micro Crystalline Cellulose (MCC) and Sodium bicarbonate (NaHCO3). The formulations were evaluated for thickness, hardness, friability, average weight variation, drug content, floating lag time, duration of floating and in vitro drug release. The data obtained from the in vitro dissolution studies were fitted in different models.Results: All the tablets were satisfactory during the preformulation studies while F11 polyherbal formulation showed the maximum floating time of 15 h, minimum floatation lag time of 35 s and drug release of 100.12%. The dissolution kinetic studies for the optimum formulation was found to follow Korsemeyer and Peppas model with R2 value, rate constant KK and n as 0.9819, 1.0492 and 1.7385 respectively with a significance of P<0.05 and showed better results compared to that of film coated herbal tablets.Conclusion: Thus this polyherbal floating effervescence tablets can be used not only as an effective drug release method for herbal drugs to enhance their bioactivity but also as a replacement for film coated herbal tablets.

Tribulus terrestris and Pedalium murex are the two popular herbal plants originated from a very famous Ayurvedic plant named Gokshuru. These plants are well known for their medicinal importance. In Ayurveda, T. terrestris is known as laghu gokhshuru and P. murex is known as brihat gokshuru. The fruits of T. terrestris plant are slightly astringent in taste while the fruits of P. murex are somewhat sweeter in taste. The fruits are used to treat various diseases such as urinary disorder adrenal stone. In this current paper, we have discussed the pharmacological, Ayurvedic and folk uses of these plants in every region of the world. From various reported studies, it was found that the T. terrestris plant carries much medicinal importance while the P. murex fruits is only used as a substitute or as an adulterant of the T. terrestris plant. It was found that the T. terrestris plant is mainly used to cure urinogenital disorders and kidney disorders such as renal and gall bladder stone. On the other hand, the P. murex plant carries aphrodisiac property and mainly used to cure sexual disorders like infertility, erectile dysfunctioning in both male and females. It was also found that these plants have some similar phytochemical constituents which possess aphrodisiac, anti-inflammatory, demulcent, diuretic properties, and also used to cure other diseases such as cough, asthma, cardiac disease, vesical calculi, and gall bladder stone. These plants are known for major therapeutic properties such as anti-ulcerative, antiurolithic, anticancer, aphrodisiac, analgesic, stomachic, anti-hypertensive, diuretic, urinary anti-infective, cardio tonic, antibacterial, anti-inflammatory, nephroprotective, antispasmodic, anthelmintic, and anti-carcinogenic.
