The present study clarified changes in the contents of polar metabolites (amino acids, organic acids, saccharides, cyclitols, and phosphoric acid) in leaf senescence in Ginkgo biloba with or without the application of methyl jasmonate (JA-Me) in comparison with those in naturally senescent leaf blades and petioles. The contents of most amino acids and citric and malic acids were significantly higher in abaxially, and that of myo-inositol was lower in abaxially JA-Me-treated leaves than in adaxially JA-Me-treated and naturally senescent leaves. The levels of succinic and fumaric acids in leaves treated adaxially substantially high, but not in naturally senescent leaves. In contrast, sucrose, glucose, and fructose contents were much lower in leaf blades and petioles treated abaxially with JA-Me than those treated adaxially. The levels of these saccharides were also lower compared with those in naturally senescent leaves. Shikimic acid and quinic acid were present at high levels in leaf blades and petioles of G. biloba. In leaves naturally senescent, their levels were higher compared to green leaves. The shikimic acid content was also higher in the organs of naturally yellow leaves than in those treated with JA-Me. These results strongly suggest that JA-Me applied abaxially significantly enhanced processes of primary metabolism during senescence of G. biloba compared with those applied adaxially. The changes in polar metabolites in relation to natural senescence were also discussed.

Lolium spp. are troublesome weeds mainly found in winter cereal crops worldwide, including Europe. In recent years resistant mechanisms have been evolved to several important herbicides. In this study we investigated the mechanisms responsible for conferring glyphosate resistance in some Lolium spp. populations. A holistic approach was used, based on dose-response experiments, determination of shikimic acid concentration in plant leaf tissue, as well as molecular analyses. More specifically, in three Lolium spp. populations the existence of a mutation in the Pro-106 codon of the 5-enolpyruvylshikimate-3 phosphate synthase (EPSPS) gene was investigated as well as the relative transcript levels of four ABC-transporter genes were monitored at three time points after glyphosate application. The results demonstrated that glyphosate resistance is a multifactor phenomenon. Relative transcript levels of the ABC-transporter genes were abundant at very early time points after glyphosate treatments. Dose-response experiments and shikimate analyses were in accordance with the findings of the quantitative PCR (qPCR) analyses. We suggest that relative expression ratio of ABC-transporter genes can be a useful tool to rapidly identify Lolium spp. populations resistant to glyphosate.

The biochemical compounds of red raspberry (Rubus idaeus L.) fruits cultivated with conventional growing technology and on a nutrient substrate were studied during 2019–2020 at the Federal Horticultural Research Center for Breeding, Agrotechnology and Nursery, Moscow, Russia. The antioxidant activity, phenolic compounds, and ash constituents of the fruits and the metabolites of the alcoholic extract of the raspberries were determined. The effect of growing technologies, i.e., conventional vs. nutrient substrate, on the accumulation of macro- and microelements in raspberry fruits was established. In red raspberries grown on nutrient substrate, the antioxidant activity decreased by 25 times (aqueous extract) and 1.5 times (alcoholic extract). The K and Na contents and Se contents of red raspberries grown on nutrient substrate were 1.5 and 3 times higher than those of raspberries of grown with conventional technology. Raspberries grown with conventional technology contained 2 times more Ca, Ni, and Mn and 7.4 times more Fe than raspberries grown on nutrient substrate. The total amount of elements in raspberries grown through soilless cultivation was 5.5% higher than that in berries grown conventionally. A total of 48 compounds were identified in the alcoholic extracts, and only 29 substances were found in berries grown on a nutrient substrate. Sugar and citric acid constituted the largest share of red raspberry components. Fructose and turanose disaccharide synthesis in raspberries grown on nutrient substrate was 20% higher than that in conventionally grown raspberries. A total of 48 organic compounds with different biological activities were identified. They included five substances with antimicrobial activity, three phenolic substances, eight organic acids, four sugar acids, nine amino acids, and 19 sugars and their derivatives. At the same time, 42 compounds were found in raspberries grown with traditional technology, and 21 compounds were identified in raspberry fruits grown on nutrient substrate. Three fatty acids, namely, ɑ-linoleic acid (polyunsaturated omega-6 fatty acid), palmitic acid, and stearic acid (saturated fatty acid), along with cinnamic acid, shikimic acid, and chrysin were found in berries grown conventionally.

Abstract Hydroxycinnamoyl shikimates were reported in 2005 to be intermediates in monolignol biosynthesis. 3-Hydroxylation of p-coumarate, originally thought to occur via coumarate 3-hydroxylase (C3H) from p-coumaric acid or its CoA thioester, was revealed to be via the action of coumaroyl shikimate 3′-hydroxylase (C3′H) utilizing p-coumaroyl shikimate as the substrate, itself derived from p-coumaroyl-CoA via hydroxycinnamoyl-CoA: shikimate hydroxycinnamoyltransferase (HCT). The same HCT was conjectured to convert the product, caffeoyl shikimate, to caffeoyl-CoA to continue on the pathway starting with its 3-O-methylation. At least in some plants, however, a more recently discovered caffeoyl shikimate esterase (CSE) enzyme hydrolyzes caffeoyl shikimate to caffeic acid from which it must again produce its CoA thioester to continue on the monolignol biosynthetic pathway. HCT and CSE are therefore monolignol biosynthetic pathway enzymes that have provided new opportunities to misregulate lignification. To facilitate studies into the action and substrate specificity of C3H/C3′H, HCT, and CSE enzymes, as well as for metabolite authentication and for enzyme characterization, including kinetics, a source of authentic substrates and products was required. A synthetic scheme starting from commercially available shikimic acid and the four key hydroxycinnamic acids (p-coumaric, caffeic, ferulic, and sinapic acid) has been developed to provide this set of hydroxycinnamoyl shikimates for researchers.

The process of seed germination is crucial not only for the completion of the plant life cycle but also for agricultural production and food chemistry; however, the underlying metabolic regulation mechanism involved in this process is still far from being clearly revealed. In this study, one indica variety (Zhenshan 97, with rapid germination) and one japonica variety (Nipponbare, with slow germination) in rice were used for in-depth analysis of the metabolome at different germination stages (0, 3, 6, 9, 12, 24, 36, and 48 h after imbibition, HAI) and exploration of key metabolites/metabolic pathways. In total, 380 annotated metabolites were analyzed by using a high-performance liquid chromatography (HPLC)-based targeted method combined with a nontargeted metabolic profiling method. By using bioinformatics and statistical methods, the dynamic changes in metabolites during germination in the two varieties were compared. Through correlation analysis, coefficient of variation analysis and differential accumulation analysis, 74 candidate metabolites that may be closely related to seed germination were finally screened. Among these candidates, 29 members belong to the ornithine–asparagine–polyamine module and the shikimic acid–tyrosine–tryptamine–phenylalanine–flavonoid module. As the core member of the second module, shikimic acid’s function in the promotion of seed germination was confirmed by exogenous treatment. These results told that nitrogen flow and antioxidation/defense responses are potentially crucial for germinating seeds and seedlings. It deepens our understanding of the metabolic regulation mechanism of seed germination and points out the direction for our future research.

The polyphagous brown marmorated stink bug (Halyomorpha halys Stål.) is an important pest in many countries. Recently it was noticed that it can feed on and cause damage to strawberries (Fragaria × ananassa Duch.). The metabolic response of strawberries to brown marmorated stink bug attacks was studied. Brown marmorated stink bugs attacked strawberry fruit which had 18% lower total sugar content compared to that of the control treatment. However, organic acid content had no significant difference among the three treatments, with the exception of shikimic acid, which had the highest content in the attacked fruit. Thirty-one phenolic compounds were identified. Results showed a strong effect on secondary metabolites due to H. halys attacks. Halyomorpha halys treatment had 27% higher total analyzed phenolic content compared to the indirect Halyomorpha halys treatment. The brown marmorated stink bug significantly increased total ellagic acid derivatives (33.1% to 37% higher), hydroxycinnamic acids (22.3% higher) and anthocyanins’ (39% higher) contents. Fruit attacked by Halyomorpha halys also had higher catechin and epicatechin content than that of the control treatments. This pest had a significant influence on the plant’s secondary metabolism, and this improved our understanding of how a strawberry plant reacts to the attacks of this very important pest. H. halys-infested fruit are not suitable for commercial production, due to the production of off-flavors.

Rhodiolacrenulata (Hook.f. & Thomson) H.Ohba is an alpine medicinal plant that can survive in extreme high altitude environments. However, its changes to extreme high altitude are not yet clear. In this study, the response of Rhodiola crenulata to differences in altitude gradients was investigated through chemical, ICP-MS and metabolomic methods. A targeted study of Rhodiola crenulata growing at three vertical altitudes revealed that the contents of seven elements Ca, Sr, B, Mn, Ni, Cu, and Cd, the phenolic components, the ascorbic acid, the ascorbic acid/dehydroascorbate ratio, and the antioxidant capacity were positively correlated with altitude, while the opposite was true for total ascorbic acid content. Furthermore, 1165 metabolites were identified: flavonoids (200), gallic acids (30), phenylpropanoids (237), amino acids (100), free fatty acids and glycerides (56), nucleotides (60), as well as other metabolites (482). The differential metabolite and biomarker analyses suggested that, with an increasing altitude: (1) the shikimic acid-phenylalanine-phenylpropanoids-flavonoids pathway was enhanced, with phenylpropanoids upregulating biomarkers much more than flavonoids; phenylpropanes and phenylmethanes upregulated, and phenylethanes downregulated; the upregulation of quercetin was especially significant in flavonoids; upregulation of condensed tannins and downregulation of hydrolyzed tannins; upregulation of shikimic acids and amino acids including phenylalanine. (2) significant upregulation of free fatty acids and downregulation of glycerides; and (3) upregulation of adenosine phosphates. Our findings provide new insights on the responses of Rhodiola crenulata to extreme high altitude adversity.
