Tribulus terrestris and Pedalium murex are the two popular herbal plants originated from a very famous Ayurvedic plant named Gokshuru. These plants are well known for their medicinal importance. In Ayurveda, T. terrestris is known as laghu gokhshuru and P. murex is known as brihat gokshuru. The fruits of T. terrestris plant are slightly astringent in taste while the fruits of P. murex are somewhat sweeter in taste. The fruits are used to treat various diseases such as urinary disorder adrenal stone. In this current paper, we have discussed the pharmacological, Ayurvedic and folk uses of these plants in every region of the world. From various reported studies, it was found that the T. terrestris plant carries much medicinal importance while the P. murex fruits is only used as a substitute or as an adulterant of the T. terrestris plant. It was found that the T. terrestris plant is mainly used to cure urinogenital disorders and kidney disorders such as renal and gall bladder stone. On the other hand, the P. murex plant carries aphrodisiac property and mainly used to cure sexual disorders like infertility, erectile dysfunctioning in both male and females. It was also found that these plants have some similar phytochemical constituents which possess aphrodisiac, anti-inflammatory, demulcent, diuretic properties, and also used to cure other diseases such as cough, asthma, cardiac disease, vesical calculi, and gall bladder stone. These plants are known for major therapeutic properties such as anti-ulcerative, antiurolithic, anticancer, aphrodisiac, analgesic, stomachic, anti-hypertensive, diuretic, urinary anti-infective, cardio tonic, antibacterial, anti-inflammatory, nephroprotective, antispasmodic, anthelmintic, and anti-carcinogenic.

Aim: To test the effectiveness of marketed polyherbal formulations on lipopolysaccharide induced inflammatory conditions in macrophages. Background: Usage of herbal compounds among patients suffered by arthritis and cancer is increasing every year. Many anti-inflammatory herbal products available in the market should be screened thoroughly for their possible mechanism of action. Objective: Joint Pain Spl (JPS) is a polyherbal dietary food supplement composed of 13 herbal plants and Rumalaya Forte (RF) is a polyherbal formulation comprising of 6 herbal plants were tested for its cytotoxicity, as well as antioxidant and anti-inflammatory activity in LPS treated IC-21 peritoneal macrophages. Methods: Commercially available JPS and RF powder was used to prepare the extract. The aqueous and methanol extracts were quantified for the presence of phenolic and flavonoid compound and confirmed with HPLC. In vitro DPPH free scavenging activity was performed. Cytotoxicity was tested by MTT assay. Anti-inflammatory activity was tested using lipopolysaccharide stimulated IC-21 peritoneal macrophage cells. Results: The phytochemical screening showed the presence of phenolic and flavonoid compounds in JPS and RF. The aqueous and methanol extracts of JPS and RF possesses significant DPPH free radical scavenging activity. MTT assay revealed that 90.64% (aqueous extract) and 92.21% (methanol extract) of exposed macrophages are viable even after 24h exposure of maximal tested concentrations of herbal formulations. Pre-treatment of JPS and RF on LPS induced IC-21 macrophages showed an reduction in nitric oxide production (maximal 79.95%) and high level of superoxide anion scavenging activity (maximal 82.5%) over control. Conclusion: The two tested poly herbal formulations such as JPS and RF possesses anti-inflammatory activity by modulating free radical generation in IC-21 macrophages. Thus the presence of the phenolic and flavonoid compounds may contribute to the antioxidant activity.

Salsola cyclophylla, an edible halophyte, is traditionally used for inflammation and pain. To confirm the claimed anti-inflammatory and analgesic properties, a detailed study on respective pharmacological actions was undertaken. The activities are contemplated to arise from its phytoconstituents. The LC-MS analysis of S. cyclophylla 95% aqueous-ethanolic extract revealed the presence of 52 compounds belonging to phenols, flavonoids, coumarins, and aliphatics class. A high concentration of Mn, Fe, and Zn was detected by atomic absorption spectroscopic analysis. The ethyl acetate extract showed the highest flavonoid contents (5.94 ± 0.04 mg/g, Quercetin Equivalents) and Fe2+-chelation (52%) potential with DPPH radicals-quenching IC50 at 1.35 ± 0.16 mg/mL, while the aqueous ethanolic extract exhibited maximum phenolics contents (136.08 ± 0.12 mg/g, gallic acid equivalents) with DPPH scavenging potential at IC50 0.615 ± 0.06 mg/mL. Aqueous ethanolic extract and standard quercetin DPPH radicals scavenging’s were equal potent at 10 mg/mL concentrations. The aqueous ethanolic extract showed highest analgesic effect with pain reduction rates 89.86% (p = 0.03), 87.50% (p < 0.01), and 99.66% (p = 0.0004) after 60, 90, and 120 min, respectively. Additionally, aqueous ethanolic extract exhibited the highest anti-inflammation capacity at 41.07% (p < 0.0001), 34.51% (p < 0.0001), and 24.82% (p < 0.0001) after 2, 3, and 6 h of extract’s administration, respectively. The phytochemical constituents, significant anti-oxidant potential, remarkable analgesic, and anti-inflammatory bioactivities of extracts supported the traditionally claimed anti-inflammatory and analgesic plant activities.

To investigate the effect of farnesol on allergic asthma, three farnesol doses were extra-added into AIN-76 feed consumed by ovalbumin- (OVA-) sensitized and -challenged mice continuously for 5 weeks, at approximately 5, 25, and 100 mg farnesol/kg, BW/day. The results showed that there were no significant differences in body weight, feed intake, and visceral organ weights between the farnesol supplementation and dietary control groups. Farnesol supplementation decreased interleukin (IL)-6/IL-10 level ratios in bronchoalveolar lavage fluid (BALF). Farnesol supplementation significantly (P<0.05) restored the cytokine secretion ability of peritoneal macrophages that was suppressed as a result of OVA sensitization and challenge and slightly decreased tumor necrosis factor (TNF-α)/IL-10 cytokine secretion ratios. Farnesol supplementation slightly (P>0.05) decreased IL-4 but significantly (P<0.05) increased IL-2 levels secreted by the splenocytes in the presence of OVA, implying that farnesol might have a systemic antiallergic effect on allergic asthmatic mice. Farnesol supplementation significantly (P<0.05) increased IL-10 levels secreted by the splenocytes in the presence of OVA, suggesting that farnesol might have an anti-inflammatory potential to allergic asthmatic mice. Overall, our results suggest that farnesol supplementation may be beneficial to improve the Th2-skewed allergic asthmatic inflammation.

Summary Cussonia barteri Seem (Araliaceae) is a deciduous tree growing in savannah of Africa. Ethnomedicinally, it is used in Africa as an analgesic, anti-malarial, anti-inflammatory, anti-anaemic, anti-diarhoea, anti-poison, ani-pyschotic and anti-epileptic agent. This review provides a brief summary on the phytochemical screenings, ethnomedicinal and pharmacological applications of various parts of C. barteri. Leaves, stem bark and seed of C. barteri have been shown to be rich in saponins, flavonoids, phenols, sugars and alkaloids. Some of these constituents have been isolated and elucidated from C. barteri. Several compounds isolated from plant include triterpenes, saponins, polyenyne and quinic esters. Phytochemical constituents are also partly responsible for biological activities of C. barteri. Extracts and components isolated from the plant have demonstrated neuropharmacological, anti-larvicidal, anti-microbial, anti-inflammatory and antioxidant activities. Overall, the insights provided by this review reinforce the potential of C. barteri for drug development and create the need for further scientific probe of constituents of the plant with the aim of developing novel drug candidates.

Cardiovascular disease is the most common cause of death. Oxidative stress and inflammation are pathophysiological processes involved in the development of cardiovascular diseases, so anti-inflammatory and antioxidant agents that modulate redox balance have become the targets of research to evaluate their molecular mechanisms and therapeutic properties. Astaxanthin, a carotenoid of the xanthophyll group, has potent antioxidant effects due to its molecular structure and its arrangement in the plasma membrane, factors that favor the neutralization of reactive oxygen and nitrogen species. This carotenoid also stands out for its anti-inflammatory activity, possibly interrelated with its antioxidant effect, as well as for its modulation of lipid and glucose metabolism. Considering the potential positive effects of astaxanthin on cardiovascular health evidenced by preclinical and clinical studies, this paper describes the molecular and cellular mechanisms related to the antioxidant and anti-inflammatory properties of this carotenoid in cardiovascular diseases, especially atherosclerosis.

Introduction: Recently there is considerable awareness and interest in the field of herbal medicine due to its natural origin and lesser side effects compared to Allopathy. Selected herbal plants like Symplocos racemosa, commonly known as lodhra, are found mainly in plains and lower hills of Bengal. The word ‘Lodhra’ means ‘Propitious’. Symplocos racemosa is an important Indian traditional drug used in many Ayurvedic and herbal formulations for treatment of liver as well as uterine disorders and leucorrhea. Ethnobotanical Literature indicates use of Symplocos racemosa in treatment of eye disease, skin disease, ear disorders, liver and bowel complaints, tumours,uterine disorders, spongy and bleeding gums, asthma, fever, snakebite, gonorrhoea and arthritis. Aim: To analyse the anti-inflammatory activity of Symplocos racemosa using protein denaturation assay. Materials and Methods: 2 g of Lodhra bark powder is mixed with 100 ml distilled water & boiled for 20 min at 50°C. The extract is filtered using whatman filter paper & concentrated to 10 ml.1 ml each of Bovine serum albumin is added to various fixations of plant extract (10μL - 50 μL) and the anti - inflammatory activity was evaluated by analysing the percentage inhibition. Results: From this study, it is evident that Lodhra has significant anti-inflammatory activity. At 50μℓ concentration, the plant extract shower higher anti- inflammatory activity of 76%. Conclusion: Symplocos racemosa extract has proved to exhibit effective anti- inflammatory activity. Further studies have to be carried to analyse the other properties of this herb, which can be incorporated successfully in the pharmaceutical industry.

Abstract Background: High intensity and endurance exercises lead to exercise-induced oxidative stress (EIOS), exercise-induced muscle damage (EIMD), and inflammation, which are the influencing factors on muscle soreness, localized swelling, and sport performance. Therefore, the purpose of this study is to determine the effectiveness of Tribulus terrestris (TT) as an herbal supplement with antioxidant and anti-inflammatory properties on the nutritional, oxidative stress, and anti/inflammatory status, as well as the sport performance of recreational runners.Methods/design: This study is a double-blind, randomized, placebo-controlled trial, which will be conducted among recreational runners of Tabriz stadiums, Iran. Thirty-four recreational runners will be selected, and participants will be assigned randomly to two groups: to receive 500 mg TT supplement or placebo capsules twice daily for two weeks. Both groups will do the high-intensity interval training (HIIT) workouts during the study. Baseline and post-intervention body composition, muscle fatigue, and soreness parameters will be assessed. In addition, assessment of malondialdehyde (MDA), total antioxidant capacity (TAC), superoxide dismutase (SOD), high-sensitivity C-reactive protein (hs-CRP), interleukin-6 (IL-6), interleukin-10 (IL-10), creatine kinase (CK), lactate dehydrogenase (LDH), insulin-like growth factor-1 (IGF-1) and brain-derived neurotrophic factor (BDNF) will be done during three blood samplings.Discussion: This study will be the first to assess the potential effects of TT in recreational runners. Our results will contribute to the growing body of knowledge regarding TT supplementation on the nutritional, oxidative stress, anti/inflammatory status and sport performance in recreational runners.Trial registration: Iranian Registry of Clinical Trials (www.irct.ir) (ID: IRCT20150205020965N8).
