Acid hydrolysis-generated pectic oligomers have been shown to affect ripening of tomato fruit by inducing both acceleration of reddening and increased ethylene biosynthesis (Campbell & Labavitch, 1991 Plant Physiol 97:706-713). In the present work, homogeneous size classes of these oligomers were demonstrated to have different impacts on ethylene production of tomato fruit pericarp discs. Endogenous oligomeric material of the same size classes was isolated from ripening tomato tissues and also tested for biological activity. They promoted some aspects of ripening as shown by increased ACC and ethylene production, which suggests that pectic oligomers are potential regulators of the ripening process in tomatoes. A metabolic origin for these oligomers is suggested by the fact that they are produced by in vitro polygalacturonase I treatment of polygalacturonic acid or tomato pectin.

The accumulation of carotenoids such as lycopene and beta-carotene greatly influences the quality of ripe tomato (Lycopersicon esculentum) fruit because cellular levels of these compounds determine the intensity of red color. As well, lycopene has anti-cancer properties and beta-carotene is a Vitamin A precursor. Recent work has demonstrated phytochrome regulation of the carotenoid pathway but the mechanism is not completely understood. This work investigates phytochrome regulation of 1-deoxy-D-xylulose 5-phosphate synthase (DXS) and phytoene synthase (PSY), two key enzymes of carotenogenesis. A simple procedure for the assay of PSY from crude pericarp extracts was developed and mRNA levels of DXS and PSY1 genes were measured by relative RT-PCR. Discs from mature green tomatoes were ripened in total darkness, or in darkness interrupted by brief daily treatments of red light, or red light followed by far red light. After ten days of incubation, lycopene levels of red light-treated discs had reached ≈12 mg/100 g fresh weight; nearly a 50% increase over discs ripened in total darkness. This increase was not observed in discs treated with red light followed by far red light, demonstrating the red/far red reversibility (and thus phytochrome control) of carotenoid accumulation. Similar patterns of phytochrome control are observed for PSY activity but not for DXS and PSY1 transcript levels, suggesting the mechanism of control may be at the level of post-translational modification of PSY. Potential applications of this regulation of carotenoid accumulation will be discussed.

Abscission of cluster tomatoes commonly limits product marketability in the retail environment. Ripening and exogenous ethylene exposure are assumed to play important roles in cluster tomato fruit abscission. `Clarance' and `DRW7229' fruit harvested at either mature green or partially ripened stages did not abscise during storage for 2 weeks at 20 °C and 95% to 100% relative humidity (RH), although respiration and ethylene production indicated that all fruit reached the postclimacteric stage. Exogenous ethylene (1 or 10 ppm) exposure for 8 days at 20 °C and 95% to 100% RH also did not induce fruit abscission for either cultivar, although pedicel and sepal yellowing were observed. Fruit from clusters stored at 20 °C and 20% or 50% RH abscised if sepal shrivel became noticeable before the fruit reached the full red ripeness stage, while no abscission occurred in fruit that reached the full red stage prior to the appearance of sepal shrivel; no fruit stored in 95% to 100% RH abscised. Fruit that ripened prior to the appearance of sepal shrivel were “plugged” (i.e., tissue underlying the stem scar was pulled out) if manual fruit detachment from the pedicel was attempted. These results indicate that there is an interaction of water loss and fruit ripening in promoting abscission zone development in cluster tomatoes.
