Variation in protein composition among wheat (Triticum aestivum L.) cultivars to identify cultivars suitable as reference material for wheat gluten analysis

Fusarium head blight (FHB) is found in both temperate and semi-tropical regions and causes losses in wheat (Triticum aestivum L.) resulting in reduced yield, deteriorated grain quality, and contamination of the grains with mycotoxins, primarily deoxynivalenol (DON). In this study, we focused on the identification of protein components in Fusarium-inoculated and non-inoculated wheat samples along with the major antioxidant enzymes that Fusarium can encounter during FHB infection process in six winter wheat varieties when FHB symptoms started to occur. Our hypothesis was that wheat antioxidants and H2O2 may play a role against Fusarium infections, consequently preserving protein grain components. Results showed that in more resistant varieties (Olimpija and Vulkan) DON content of inoculated flour was less accumulated and thus the major gluten network was not significantly attacked by Fusarium spp. The maximum increase in guaiacol peroxidase (POD) activity in response to FHB treatment was detected in the FHB-resistant varieties Olimpija and Vulkan, while the lowest increase in H2O2 content was detected in the FHB-susceptible variety Golubica. A particular reduction in the content of both total glutenin and high-molecular-weight glutenin subunits was detected in susceptible wheat varieties after serious artificial infection, along with increased DON accumulation. The defense mechanism in wheat varieties differed, where resistant varieties’ protein composition depended on POD activity as a detoxification agent.

The main goal of our study was to present research data on genes encoding high molecular weight glutenin subunits (HMW-GS) associated with high flour bread-making quality. This is the leading research objective in our institute in the area of wheat gluten in cultivars that have not been studied so far in that respect, but which can potentially be a valuable source of new information. Identification and characterization of high molecular weight glutenin subunits (HMW-GS) were performed using sequencing and SDS-PAGE and STS-PCR methods. Genes located in the vicinity of the Glu-1 locus have been identified and characterized in 28 Polish cultivars of Triticum aestivum. The results were then analyzed using the following computer programs: Finch TV, BLAST, MEGA 4, Molecular Imager® Gel Doc™ XR, and Quantity One software (Bio-Rad). Three alleles (a, b, c) have been identified in the Glu-A1 locus, 6 alleles (a, b, c, d, e, k) in the Glu-B1 locus, and 2 alleles (a, d) in Glu-D1 using the SDS-PAGE method. The amplification of specific HMW-GS sequences generated one product of 450 bp in 1Dx5 in 13 cultivars of old wheat and of 435 bp in 1Dx2 in 15 cultivars. The amplification products of primers for 1Dy10 and 1Dy12 genes were 422 bp and 552 bp in size, respectively.

Scope of the study was to find more effective N fertilisation doses and applications to reach not only higher quantity but better quality grains as well as to be able to help preserving the continuity of feed and food quality improvement, since wheat is one of the most consumed crops all over the world.Samples of winter wheat (Triticum aestivum L.) varieties harvested from the experimental field of the Szent István University in two consecutive crop seasons had been examined in the laboratory of the Crop Production Institute. Effects of nitrogen (N) application on the performance of grain protein were tested. Five high quality winter wheat varieties were studied regarding grain quality traits affected by applying undivided and split doses of N. The evaluated samples show that increasing doses of N topdressing and increasing time of application have beneficial effects on the yield and the value of protein content. Wheat grain protein value ranged between 9.9% of the untreated Mv Karéj and Alföld, where the 120+40 kg ha-1 N was applied resulting in the highest value of 16.0%. Similarly, the gluten values among untreated and N applied plots were in a wide range. Mv Karéj had the lowest wheat gluten value on untreated plot with 18.4% and Alfold had the highest value with 36.8% on the plot where the 120+40 kg ha-1 N was applied. There were no significant changes recorded on test weight and thousand kernel weight. In the case of baking quality, there were significant differences between varieties. The best records were obtained in the case of Mv Toborzö followed by Mv Karéj.
