The aim of this study was to evaluate physiological responses to short-term heat stress in the leaves of traditional (Bistrica) and modern (Toptaste) plum cultivars. In this study, detached plum leaves were incubated at 25 °C (control) and 40 °C (stress). After 1 h of exposure to heat (40 °C), chlorophyll a fluorescence transients were measured, and several biochemical parameters were analyzed. Elevated temperature caused heat stress in both plum cultivars, seen as a decrease in water content (WT), but in the leaves of the cultivar Bistrica, an accumulation of proline and phenols, as well as an accumulation of photosynthetic pigments, suggest the activation of a significant response to unfavorable conditions. Conversely, in the leaves of Toptaste, a significant accumulation of malondialdehyde (MDA) and an activation of guaiacol peroxidase (GPOD), all together with a decreased soluble proteins content, indicate an inadequate response to maintaining homeostasis in the leaf metabolism. The impact of an elevated temperature on photosynthesis was significant in both plum cultivars as reflected in the decrease in performance indexes (PIABS and PItotal) and the maximum quantum yield of PSII (Fv/Fm), with significantly pronounced changes found in Toptaste. Unlike the traditional plum cultivar, Bistrica, in the modern cultivar, Toptaste, short-term heat stress increased the minimal fluorescence (F0) and absorption (ABS/RC), as well as Chl b in total chlorophylls. Additionally, the inactivation of RCs (RC/ABS) suggests that excitation energy was not trapped efficiently in the electron chain transport, which resulted in stronger dissipation (DI0/RC) and the formation of ROSs. Considering all presented results, it can be presumed that the traditional cultivar Bistrica has better tolerance to heat stress than the modern cultivar Toptaste. The cultivar, Bistrica, can be used as a basis in further plum breeding programs, as a source of tolerance for high temperature stress.

Abstract BACKGROUND Our study aimed to characterize and prospect immobilization strategies for a novel fungal peroxidase - POD (EC 1.11.1.7) and to insert it in the context of pollutant remediation, since these compounds pose risks to human and environmental health. The enzymatic extract was obtained by submerged fermentation of the fungus Trichoderma koningiopsis in an alternative substrate, consisting of fresh microalgal biomass. The immobilization efficiency was evaluated by monitoring the residual activity (RA) and the discoloration potential (DP) of a synthetic dye solution. Concomitantly, the catalytic properties of free POD were explored, and the most promising storage strategy to maintain the enzymatic activity was studied. RESULTS The novel guaiacol peroxidase expressed specific activity of up to 7801 U mg−1 in the free form, showing stability when subjected to up to 80°C in a pH range between 4.0-8.0. Furthermore, the bioproduct immobilized on magnetic nanoparticles expressed up to 689% RA and 100% DP. An increase in the RA of the enzyme, both in free and immobilized form, was also observed after storage for up to 8 months. The synthesized magnetic nanozymes showed good reusability, maintaining 13546 U mg−1 after ten cycles and removing 94% of color in a second batch. Toxicological evaluation with Allium cepa indicated that the enzymatic process of color removal with immobilized POD was essential to eliminate genotoxic effects. CONCLUSION T. koningiopsis peroxidase production and immobilization presented in our work are promising for the enzyme market and for the wastewater treatment technologies, considering its high bioxidative potential.

The effect of continuous lighting (CL, 24 h) and light spectrum on growth and nutritional quality of arugula (Eruca sativa), broccoli (Brassica oleracea var. italic), mizuna (Brassica rapa. var. nipposinica), and radish (Raphanus sativus var. radicula) were investigated in growth chambers under light-emitting diode (LED) and fluorescent lighting. Microgreens were grown under four combinations of two photoperiods (16 h and 24 h) providing daily light integral (DLI) of 15.6 and 23.3 mol m−2 day−1, correspondingly) with two light spectra: LED lamps and fluorescent lamps (FLU). The results show that fresh and dry weights as well as leaf mass per area and robust index of harvested arugula, broccoli, mizuna, and radish seedlings were significantly higher under CL compared to 16 h photoperiod regardless of light quality. There were no visible signs of leaf photodamage. In all CL-treated plants higher chlorophyll a/b and carotenoid-to-chlorophyll ratios were observed in all plants except mizuna. CL treatment was beneficial for anthocyanin, flavonoid, and proline accumulation. Higher activities of antioxidant enzymes (catalase, superoxide dismutase, ascorbate peroxidase, and guaiacol peroxidase) were also observed in CL-treated plants. In most cases, the effects were more pronounced under LED lighting. These results indicate that plants under mild oxidative stress induced by CL accumulated more non-enzymatic antioxidants and increased the activities of antioxidant enzymes. This added nutritional value to microgreens that are used as functional foods providing health benefits. We suggest that for arugula, broccoli, mizuna, and radish, an LED CL production strategy is possible and can have economic and nutritional benefits.

Abstract Peach trees initiate flowering and then dense budding when the temperatures in winter are steadily low. When temperatures during the winter are high or when the chilling accumulation needs of the cultivar are not met, it is necessary to apply chemicals that stimulate flowering and budding in a uniform manner. This study aimed to evaluate alternative products for breaking the dormancy of ‘Kampai’ peach trees in a subtropical region. The experiment was conducted with ‘Kampai’ peach trees in the 2018, 2019 and 2020 production with the following treatments: (1) negative control composed only of water (control); (2) positive control composed of hydrogen cyanamide at a dose of 1.5% (commercial product Dormex®) plus 4.5% mineral oil; (3) Erger G® organomineral fertilizer supplemented with calcium nitrate at a dose of 3%; (4) potassium nitrate at a dose of 5%; and (5) copper sulphate at a dose of 0.3%. We evaluated affected budding capacity, flowering, the production cycle, peach production, the quality of peach trees and the enzymatic activities of catalase and guaiacol peroxidase. Hydrogen cyanamide and the organomineral fertilizer Erger G® promoted earlier flowering and an earlier production cycle. On the other hand, hydrogen cyanamide and copper sulphate stimulated flower opening and peach production. The chemicals used decreased catalase activity (24 h after application) and increased guaiacol peroxidase activity (6 h after application). The application of copper sulphate may be an option to break the dormancy of peach trees in the subtropics.

AbstractThe use of biopesticides has gradually become essential to ensure food security and sustainable agricultural production. Nevertheless, the use of single biopesticides is frequently suboptimal in agricultural production given the diversity of biotic and abiotic stresses. The present study investigated the effects of two biopesticides, physcion and chitosan-oligosaccharide (COS), alone and in combination, on growth regulation and antioxidant potential of maize seedlings by seed coating. As suggested from the results, physcion significantly inhibited the growth of the shoots of maize seedlings due to the elevated respiration rate. However, COS significantly reduced the growth inhibition induced by physcion in maize seedlings by lowering the respiration rate and increasing the content of photosynthetic pigments and root vigor, which accounted for lower consumption of photosynthesis products, a higher photosynthetic rate and a greater nutrient absorption rate. Thus, an improved growth was identified. As indicated from the in-depth research, the application of physcion and COS combination is more effective in down-regulated the malondialdehyde (MDA) content by facilitating the activities of the antioxidative enzymes (i.e., superoxide dismutase (SOD), catalase (CAT) and guaiacol peroxidase (G-POD)). Such results indicated that the combined use of physcion and COS neither affected the normal growth of maize seedlings, but also synergistically improved the antioxidant potential of the maize plants, resulting in plants with high stress resistance. Thus, the combined use of physcion and COS by seed coating in maize production has great potential to ensure yield and sustainable production of maize.

Canola is an important temperate oil crop that can be severely affected by drought. Understanding the physiological and molecular mechanisms involved in canola tolerance to water deficit is essential to obtain drought-tolerant productive cultivars. To investigate the role of antioxidant response and the possible involvement of calcium-dependent protein kinases (CDPKs) in canola tolerance to drought, we analyzed four genotypes with different sensitivity to water stress. Leaf relative water content, canopy temperature, PSII efficiency, electrolyte leakage index and lipid peroxidation were used as indicators to classify the cultivars as drought-tolerant or drought-sensitive. Antioxidant enzymes superoxide dismutase, guaiacol peroxidase and catalase displayed significantly higher activities in drought-tolerant than in drought-sensitive cultivars subjected to water deficit, suggesting that the efficiency of the antioxidant response is essential in canola drought tolerance. The increased expression of genes BnaCDPK6 and BnaCDPK14 under drought conditions, their differential expression in drought-tolerant and drought-sensitive genotypes, and the presence of multiple cis-acting stress-related elements in their promoter regions suggest that CDPKs are part of the signaling pathways that regulate drought response in canola. We propose the BnaCDPK genes and their regulator elements as potential molecular targets to obtain drought-tolerant productive canola cultivars through breeding or genetic transformation.

ABSTRACT: This study identified physiological and biochemical changes in ‘Fuyu’ persimmon buds during dormancy. Branches were collected between March and August 2015. Dormancy was evaluated by biological testing of isolated node cuttings at 25 °C and a photoperiod of 16 h. The variables analyzed were water content; reducing sugar content; respiratory activity; activity of superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), guaiacol peroxidase (POD) and polyphenol oxidase (PPO) enzymes; hydrogen peroxide content (H2O2) and lipid peroxidation. At the end of March 2015, the plants were already dormant, and the leaves and fruits present indicated a paradormancy effect. Induction of endodormancy may have occurred in June 2015, when chilling hours (CH) below 7.2 °C and higher CH below 12 °C began to accumulate, which coincided with the period in which there was a decrease in water content and respiratory activity, an increase in reducing sugars, a decrease in SOD, CAT, APX and PPO and an increase in H2O2. After an accumulation of 553 CH below 12 °C, the budburst capacity increased, and the buds presented increased water content, decreased reducing sugars content, increased respiratory activity, low activity in SOD, CAT, APX and POD and high levels of H2O2.

Abstract The vascular wilting caused by Fusarium oxysporum f. sp. dianthi (Fod) is the most relevant disease for carnation cultivation. Understanding the biochemical mechanisms involved in resistance to Fod will allow the development of new disease control strategies. In this research, the levels of some phytohormones such as salicylic acid (SA), methyl salicylate (MeSA), and methyl jasmonate (MeJA) were evaluated in symplast of carnation roots infected with this pathogen. The accumulation of these hormones was then correlated with the expression levels of symplastic peroxidases, enzymes involved in the plant resistance against pathogen during interaction. Our results suggested that pathogen infection causes a differential accumulation of SA, MeSA, and MeJA in a resistant cultivar (i.e. ‘Golem’), being earlier and higher than that observed in a susceptible one (i.e. ‘Solex’). Simultaneously, an increase of guaiacol peroxidase enzymatic activity (GPX) and transcriptional levels of a gene coding for a symplastic peroxidase were presented as part of the resistance response. The positive statistical correlation between the accumulation of SA and MeJA and the expression of peroxidases (GPX activity and mRNA levels) indicates the possible cellular relationship of these phenomena during the activation of the resistance to Fod. Our findings suggested some hormonal signaling mechanisms acting at the roots during the regulation of the biochemical response associated with resistance against Fod.

Somatic embryogenesis is being piloted for the commercial production of genetically improved Norway spruce (Picea abies L. Karst) forest regeneration material in Finland. The main challenge to making the process commercially relevant is the dependence on time-consuming and highly skilled manual labor. Automation and scaling up are needed to improve cost-effectiveness. Moving from the proliferation of embryogenic tissue on semisolid media to suspension cultures could improve process scalability. In a series of four experiments (overall, with 20 cell lines, 4–9 per experiment), the suitability of proliferation in suspension culture for Norway spruce somatic embryogenesis was evaluated based on the growth rate, indicators of stress conditions, good-quality cotyledonary embryo yield, and embling survival in a greenhouse. The proliferation rate in suspension was found equal to on semisolid media, but with a remarkable genotypic variation. Embryogenic tissue matured directly without pre-treatments from suspension onto semisolid media produced lower numbers of good-quality embryos than tissue matured from semisolid media. Rinsing the suspension-grown tissue with hormone-free liquid media before maturation improved embryo yield, bringing it closer to that of semisolid-grown tissue. Decreasing 6-benzylaminopurine and 2,4-dichlorophenoxyacetic acid concentrations in suspension proliferation media to 0.5 or 0.1 times those in semisolid media did not affect tissue growth and did not improve embryo production. The hydrogen peroxide (H2O2) content and guaiacol peroxidase activity were elevated in suspension cultures compared with semisolid medium, which had the same plant growth regulator content. In one experiment out of four, the greenhouse survival of germinants was lower when proliferation was carried out in full strength suspension than on semisolid media; in other experiments the survival rates were equal.

The effects of mycorrhiza, Thiobacillus and Nitroxin (Azotobacter and Azospirillum sp.) biofertilizers under drought stress conditions with four levels of field capacity (FC) (control(100%), 85%, 70%, and 50%) on the antioxidant enzyme activities of medicinal pumpkin (Cucurbita pepo convar. pepo var. Styriaca) were evaluated during the years 2018–2019. Irrigation levels exhibited significant effects on all studied variables, except for the catalase (CAT) enzyme. A significant correlation was observed between the effects of irrigation levels and biofertilizers on antioxidant enzymes, soluble protein content, and grain yield. The highest activity of catalase and ascorbate peroxidase (APX) enzymes was achieved using mycorrhiza in 50% FC. Increasing drought intensity and mycorrhiza stimulated glutathione reductase (GR) and guaiacol peroxidase (GPX) activities by 32% and 66%, while Nitroxin increased them by 16% and 43%, respectively. Under severe drought stress conditions, only mycorrhiza exhibited a positive effect on GR and GPX enzymes. Under moderate and severe drought stress conditions, Nitroxin increased grain yield by 13% and 12.6%, respectively. The irrigation regimes and bio-fertilizers had a significant effect on β-sitosterol percentage. The highest amount was observed at the highest level of drought stress. Among the various bio-fertilizers treatments, the application of Thiobacillus yielded the highest percentage of β-sitosterol. The results of the present study demonstrate that the application of biofertilizers is beneficial in coping with drought stress.
