Analysis of highbush blueberry (Vaccinium corymbosum L.) fruit growth with exponential mixed models

AbstractUltraviolet-B (UV-B) light (280–315 nm) is an important environmental signal that regulates plant development and photomorphogenesis, while also affecting the flavonoid pathway, including anthocyanin biosynthesis. Regarding the effects of UV-B radiation on fruits, the effects of a short-term or postharvest irradiation on fruit quality have been well-documented, but the effects of a long-term preharvest UV-B irradiation on fruit growth and coloration remain unclear. Thus, in this study, we investigated the effects of a long-term treatment involving an environmentally relevant UV-B dose on highbush blueberry (Vaccinium corymbosum) fruit. The preharvest UV-B treatment quickly promoted fruit growth and sugar accumulation, which is not commonly observed in other fruit tree species. The UV-B exposure also accelerated fruit ripening and coloration. The dual-luciferase assay proved that in blueberries, expression of VcUFGT encoding anthocyanin biosynthesis key enzyme, is positively and negatively regulated by VcMYBA1 and VcMYBC2, respectively. Throughout the fruit development stage, the UV-B treatment up-regulated VcMYBPA1 expression, which increased VcUFGT expression via VcMYBA1. In the green fruit stage, the UV-B treatment increased HY5 encoding UV receptor, which up-regulates VcMYBPA1 and down-regulates VcMYBC2, thereby promotes the accumulation of anthocyanins. On the other hand, excessive anthocyanin synthesis was inhibited by increased VcMYBC2 levels in mature fruits when exposed to UV-B light through HY5-independent pathway. In conclusion, anthocyanin-related MYB activators and repressor may coordinately balance the accumulation of anthocyanins in blueberry fruits, with UV-B treatments possibly influencing their effects in a stage-specific manner. The potential utility of preharvest UV-B treatments for improving blueberry fruit quality is discussed herein.

Ovule abortion occurred between 5 and 10 days after pollination (DAP) in self- and cross-pollinated `Sharpblue' blueberry (Vaccinium corymbosum L.) fruit. More ovule abortion occurred with self-pollination (35%) than with cross-pollination (22% for `Sharpblue' × `O'Neal' and 29% for `Sharpblue' × `Gulfcoast'), and there were more poorly developed ovules with self-pollination (88.1%) than with cross-pollination (× `O'Neal', 33.6%; × `Gulfcoast' 50.8%). The increase in ovule area correlated exponentially with fruit growth during early developmental stages, regardless of pollination treatment. However, cross-pollination resulted in significantly greater ovule area and fruit mass during early fruit development as well as at ripening. Ovule area was maximum at 25 to 30 DAP for both pollination treatments, followed by exponential fruit growth (stage III). Cross-pollination resulted in greater fruit growth and a shorter stage III. At 10 DAP, ovules from cross-pollination were larger than those from self-pollination, suggesting that cross-pollination initiated ovule growth immediately after fertilization. This research suggests that southern highbush blueberry fruit growth and development is intimately associated with ovule growth and development, which is affected by pollen sources.

A study was made of barriers to hybridization between tetraploid V. corymbosum L., the cultivated highbush blueberry, and V. elliottii Chapm., a wild diploid blueberry which may be useful in breeding cultivars adapted to the southeastern United States. Both prefertilization and postfertilization barriers were detected. Prefertilization barriers were mainly observed when V. elliottii was the male parent and were manifested as an arrest of pollen tube growth at the base of the style. In cases where fertilization did take place, regardless of which species was used as the female, a strong postfertilization barrier usually prevented development of hybrid zygotes. Zygotes remained in a resting state after fertilization and usually aborted before dividing. Meanwhile, the endosperm underwent four to six cycles of cell division before it started to degenerate. A causal relationship between embryo abortion and endosperm malfunction was not clearly established. Endosperm malfunction and embryo abortion were delayed when V. corymbosum was the female parent.
