Genetic purity testing of F1 hybrid seed with molecular markers in cabbage (Brassica oleracea var. capitata)

Cabbage (Brassica oleracea) is an important vegetable crop that is cultivated worldwide. Previously, we reported the identification of two dominant complementary hybrid lethality (HL) genes in cabbage that could result in the death of hybrids. To avoid such losses in the breeding process, we attempted to develop molecular markers to identify HL lines. Among 54 previous mapping markers closely linked to BoHL1 or BoHL2, only six markers for BoHL2 were available in eight cabbage lines (two BoHL1 lines; three BoHL2 lines; three lines without BoHL); however, they were neither universal nor user-friendly in more inbred lines. To develop more accurate markers, these cabbage lines were resequenced at an ~20× depth to obtain more nucleotide variations in the mapping regions. Then, an InDel in BoHL1 and a single-nucleotide polymorphism (SNP) in BoHL2 were identified, and the corresponding InDel marker MBoHL1 and the competitive allele-specific PCR (KASP) marker KBoHL2 were developed and showed 100% accuracy in eight inbred lines. Moreover, we identified 138 cabbage lines using the two markers, among which one inbred line carried BoHL1 and 11 inbred lines carried BoHL2. All of the lethal line genotypes obtained with the two markers matched the phenotype. Two markers were highly reliable for the rapid identification of HL genes in cabbage.

In sustainable agricultural systems, intercropping using living mulches (LM) provides many beneficial ecosystem services. The objective of these two-year field experiments was to study the suitability of different LM options of burr medic (<em>Medicago polymorpha</em> L. var. <em>anglona</em>) for organic cauliflower (<em>Brassica oleracea</em> L.) cultivation in two sites under Mediterranean conditions. In central Italy (Experiment 1) contemporary and delayed (to crop) sowings of LM were compared with a no-cover crop treatment, contrasting two local cauliflower cultivars and a F1 Hybrid. In southern Italy (Experiment 2) the sustainability of systems combining LM (anticipated and contemporary sowing compared with no-cover) and organic fertilisation strategies was assessed. The aboveground biomasses dry weights of cauliflower crop (heads and residues), burr medic and weeds were separately determined. Results suggested that in Experiment 1 the LM was not able to smother weeds establishment and growth, as a consequence of early sowing, while cauliflower yield was reduced. Moreover, the genotypes behaviour was greatly influenced by the LM sowing times. In Experiment 2, irrespective of the agronomic practices applied, climatic conditions notably influenced cauliflower cultivation and also reduced the mean yield. Therefore, the recorded differences between the two experimental sites highlighted the need to tailor the LM strategies to the different environmental conditions.

ABSTRACTSeed Purity Testing of F3 Progeny of Rice Lines Derived from a Cross between Pandanwangi x PTB-33 Estimated by Simple Sequence Repeat MarkersSeeds with high purity is required to produce maximum crop yield. Genetic purity of selected F3rice seed progenies derived from a cross between Pandanwangi x PTB33 was estimated by SSR(simple sequence repeats) molecular markers. Objective of current experiment was to obtain riceseed with high genetic purity (100%) in terms of homogeneity of alleles. The experiment wasconducted at Laboratory of Plant Analysis and Biotechnology, meanwhile field experiment wasperformed at Ciparanje Experimental Station, Faculty of Agriculture, Universitas Padjadjaran.Based on primer designed by Bradbury (aromatic trait) and primers RM589 and RM586 (supposedto correlate with brown planthopper resistance gene), seeds of F3 selected had 100% geneticpurity. Percobaan ini bertujuan untuk mendapatkan generasiF3 yang memiliki kemurnian genetik 100%. Penelitian dilaksanakan di Kebun PercobaanCiparanje, Fakultas Pertanian Universitas Padjadjaran dan Laboratorium Analisis dan BioteknologiTanaman. Hasil analisis menggunakan primer Bradbury menunjukkan 100% benih murniberdasarkan karakter aromatik, begitupun berdasarkan karakter ketahanan terhadap wereng(primer RM589 dan RM586) menunjukkan 100% benih murni. Marka molekuler SSR yangdigunakan untuk verifikasi kemurnian mampu menunjukkan kemurnian genetik benih padi yangtinggi. Genotip PP dapat dilanjutkan untuk pengujian dan atau perbanyakan benih sumber.Kata kunci: benih padi, F3, marka SSR, uji kemurnian genetik
