ABSTRACT Objective To evaluate the incidence of Y-chromosome microdeletions in individuals born from vasectomized fathers who underwent vasectomy reversal or in vitro fertilization with sperm retrieval by epididymal aspiration (percutaneous epididymal sperm aspiration). Methods A case-control study comprising male children of couples in which the man had been previously vasectomized and chose vasectomy reversal (n=31) or in vitro fertilization with sperm retrieval by percutaneous epididymal sperm aspiration (n=30) to conceive new children, and a Control Group of male children of fertile men who had programmed vasectomies (n=60). Y-chromosome microdeletions research was performed by polymerase chain reaction on fathers and children, evaluating 20 regions of the chromosome. Results The results showed no Y-chromosome microdeletions in any of the studied subjects. The incidence of Y-chromosome microdeletions in individuals born from vasectomized fathers who underwent vasectomy reversal or in vitro fertilization with spermatozoa recovered by percutaneous epididymal sperm aspiration did not differ between the groups, and there was no difference between control subjects born from natural pregnancies or population incidence in fertile men. Conclusion We found no association considering microdeletions in the azoospermia factor region of the Y chromosome and assisted reproduction. We also found no correlation between these Y-chromosome microdeletions and vasectomies, which suggests that the assisted reproduction techniques do not increase the incidence of Y-chromosome microdeletions.

The aim of the study was to compare the ability of various media for cryopreservation of sperm to ensure their viability after thawing and to assess the possibility of using Narcissus pseudonarcissus lectin to determine the viability of native and cryopreserved human sperm by flow cytometry. Materials and methods. Used ejaculate 54 men aged 26 to 47 years, undergoing treatment for infertility. The control was a native ejaculate, which was also used for the in vitro fertilization procedure. Four parallel samples were frozen using various commercial media. After storage and thawing, spermatozoa viability was assessed by flow cytometry using three dyes and Narcissus pseudonarcissus lectin. Results. All assays showed that cryopreservation led to a twofold decrease of sperm viability, dye to the changes in the composition and properties of cell membrane, decrease in mitochondrial membrane potential, as well as the damages of acrosomal complex and nucleus. The lowest decrease in sperm viability was shown for Quinns advantage sperm freezing medium for cryopreservation. Conclusion. Flow cytometry makes it possible to evaluate with high efficiency sperm viability as the part of in vitro fertilization. The results of viability assessment using daffodil lectin make the prediction of in vitro fertilization outcome more accurate.

Приводятся данные обследования мужчин с азооспермией и олигозооспермией тяжелой степени, имеющих и не имеющих микроделеции длинного плеча Y-хромосомы, а также результаты биопсии тестикулярной ткани. Отсутствие делеций региона AZF, отсутствие гипоплазии яичек, а также нормальные показатели ФСГ, ЛГ и ингибина В являются прогностически благоприятными критериями в отношении успешности получения сперматозоидов при биопсии ткани яичка для проведения экстракорпорального оплодотворения (ЭКО). The article presents the results of a survey of men with azoospermia and severe oligozoospermia, with and without microdeletion of the long arm of the Y chromosome, as well as the results of a testicular biopsy. The absence of deletions of the AZF region, the absence of testicular hypoplasia, as well as normal levels of FSH, LH and inhibin B are prognostically favorable criteria for the success of obtaining spermatozoa with a biopsy of testicular tissue for in vitro fertilization (IVF).

Работа посвящена анализу возрастных изменений сперматозоидов человека и их функциональных свойств у мужчин 26-47 лет. В работе использовали эякулят, полученный от 54 мужчин в рамках проведения процедуры экстракорпорального оплодотворения (ЭКО). Пациенты были разделены на три группы (26-29 лет, 30-34 года и старше 35 лет). В ходе работы собирали параметры спермограммы и результаты оценки жизнеспособности сперматозоидов, полученные с помощью метода проточной цитометрии, а также проводили ретроспективный анализ эффективности экстракорпорального оплодотворения. Установлено, что средние значения параметров спермограммы по группам находились в пределах физиологической нормы, однако около 59 % пациентов имели индивидуальные отклонения по 1-3 показателям. Цитометрический анализ выявил быстрое нарастание с возрастом функциональных нарушений сперматозоидов, затрагивающих аппараты распознавания, проникновения (акросома), энергетический аппарат клетки (его митохондрия) и плотность упаковки хроматина в его ядре. Результатом становится снижение вероятности оплодотворения с 88 % в 26-29 лет до 61 % после 35 лет при проведении ЭКО. Обосновывается значение полученных результатов для анализа возрастных изменений мужской репродуктивной системы и практики лечения мужского бесплодия. The work is devoted to the analysis of age-related changes in human spermatozoa and their functional properties in men aged 26-47 years. The work used ejaculate obtained from 54 men as part of the in vitro fertilization procedure. The patients were divided into three groups (26-29, 30-34 and over 35 years old). In the course of the work, the parameters of the spermogram were collected, as well as the results of assessing the viability of spermatozoa, obtained using the method of flow cytometry, and also a retrospective analysis of the effectiveness of in vitro fertilization was carried out. It was found that the average values of spermogram parameters in groups were within the physiological norm, however, about 59 % of patients had individual deviations in terms of 1-3 indicators. Cytometric analysis revealed a rapid increase with age in functional disorders of spermatozoa, affecting the recognition and penetration apparatus (acrosome), the energy apparatus of the cell (its mitochondrion) and the density of chromatin in its nucleus. The result is a decrease in the probability of fertilization from 88 % at 26-29 years old to 61 % after 35 years, even with in vitro fertilization. The significance of the results obtained for the analysis of age-related changes in the male reproductive system and the practice of treating male infertility is substantiated.
