Pressurized water (PW) extraction of galanthamine from Narcissus pseudonarcissus bulbs was performed. The obtained yield was compared with the yield from conventional acidified water extraction and methanolic Soxhlet extraction. Both PW and conventional acidified water extraction were followed by a subsequent purification step for the alkaloids. The PW extraction (70 °C, 150 bar, 45 min) yielded as much galanthamine as methanolic-Soxhlet extraction (ca. 3.50 mg/g). Meanwhile, acid-base extraction with 1% of HBr (v/v) at 65 °C for 3 h gave a lower yield (ca. 2.65 mg/g). A higher PW temperature did not significantly increase the galanthamine yield. Pressure increase is not necessary since more water-soluble compounds such as proteins and polysaccharides are co-extracted, resulting in high viscosity of the water extract solution, which hampers the filtration process. Hence, the acidity of the solution is highly important both in the case of PW extraction and acidified water extraction. Besides galanthamine, the total alkaloid profile following Narcissus alkaloids was also obtained. Lycoramine, O-methyloduline, norgalanthamine, epi-norgalanthamine, narwedine, oduline, haemanthamine, O-methyllycorenine, and a haemanthamine derivate were identified. Although a high yield was obtained from PW extraction, the further purification needs to be improved to obtain an economically feasible industrial extraction process.

In a pot experiment, an early-flowering Narcissus pseudonarcissus cv. ‘Dutch Master’ (DM) and late-flowering N. poeticus cultural form (PO) were examined. The photosynthetic rate (A), transpiration rate (E), stomatal conductance (gs), photosynthetic water use efficiency (WUE), relative chlorophyll content (CCI) and chlorophyll fluorescence (F) were measured regularly. Leaf length, scape length and weight of the plant organs were also measured. The DM cultivar had higher gs and lower E values than the PO on most measuring dates (season average: gs: DM: 165.34, PO: 123.63; E: DM: 1.39, PO: 1.78 mmol H2O m−2s−1). The A curve was similar for the two taxa, except for the first measuring dates. The basic F values (F0, Fm, Fv) for DM were lower and CCI values were higher than for PO (season average of CCI: DM: 94.82, PO: 60.34). The Fm/F0, Fv/F0 and CCI curves were well described by a second order equation. The seasonal change of F and CCI values was the greatest for both taxa near the leaf tip. Bulb growth occurred in the two taxa in approximately the same calendar period, regardless of flowering time. A significant part of the leaf growth in DM occurred after flowering, while the leaves of PO reached their mature size by flowering.

Two new minor Amaryllidaceae alkaloids were isolated from Hippeastrum × hybridum cv. Ferrari and Narcissus pseudonarcissus cv. Carlton. The chemical structures were identified by various spectroscopic (one- and two-dimensional (1D and 2D) NMR, circular dichroism (CD), high-resolution mass spectrometry (HRMS) and by comparison with literature data of similar compounds. Both isolated alkaloids were screened for their human acetylcholinesterase (hAChE) and butyrylcholinesterase (hBuChE) inhibition activity. One of the new compounds, a heterodimer alkaloid of narcikachnine-type, named narciabduliine (2), showed balanced inhibition potency for both studied enzymes, with IC50 values of 3.29 ± 0.73 µM for hAChE and 3.44 ± 0.02 µM for hBuChE. The accommodation of 2 into the active sites of respective enzymes was predicted using molecular modeling simulation.

AbstractAlzheimer’s disease (AD) is a disorder associated with progressive degeneration of memory and cognitive function. Galantamine is a licenced treatment for AD but supplies of the plant alkaloid that it is produced from, galanthamine, are limited. This three-year system study tested the potential to combine Narcissus-derived galanthamine production with grassland-based ruminant production. Replicate plots of permanent pasture were prepared with and without bulbs of Narcissus pseudonarcissus sown as lines into the sward. Two different fertiliser regimes were imposed. The above-ground green biomass of N. pseudonarcissus was harvested in early spring and the galanthamine yield determined. In the second harvest year a split-plot design was implemented with lines of N. pseudonarcissus cut annually and biennially. All plots were subsequently grazed by ewes and lambs and animal performance recorded. Incorporation of N. pseudonarcissus into grazed permanent pasture had no detrimental effects on the health or performance of the sheep which subsequently grazed the pasture. There was no consistency to the effects of fertiliser rates on galanthamine yields. There was no difference in overall galanthamine yield if N. pseudonarcissus was cut biennially (1.64 vs. 1.75 kg galanthamine/ha for annual combined vs biennial cuts respectively; s.e.d = 0.117 kg galanthamine/ha; ns). This study verified the feasibility of a dual cropping approach to producing plant-derived galanthamine.

Abstract Aims In species with morphophysiological seed dormancy (MPD), little is known about the effects of desiccation of imbibed seeds on embryo growth and germination. We studied seed responses to dehydration in nine species with different levels of MPD. Methods For each species, a control test was conducted by keeping seeds permanently hydrated and exposed to the optimal stratification-incubation sequence to promote embryo growth. Simultaneously, tests were run in which seed stratification was interrupted for 1 month by desiccation at room temperature. Important Findings In Clematis vitalba and Ribes alpinum, with nondeep simple MPD, desiccation affected neither embryo growth nor seed viability, but the desiccation led to a decrease of germinative ability in R. alpinum by 16%. The seeds of Narcissus pseudonarcissus subsp. munozii-garmendiae, with deep simple epicotyl MPD, tolerated desiccation in different embryo growth stages, but their germinative ability decreased slightly. The response of species with complex levels of MPD to desiccation was more variable: Delphinium fissum subsp. sordidum, with intermediate complex MPD, and Anthriscus sylvestris and Meum athamanticum, both with deep complex MPD, tolerated desiccation. In contrast, Ribes uva-crispa with nondeep complex MPD, Lonicera pyrenaica with intermediate complex MPD, and Chaerophyllum aureum with deep complex MPD, had diminished germination ability by desiccation. Although seeds of the species with simple levels of MPD tolerated desiccation, those of some species with complex levels were also highly tolerant. Thus, desiccation did not induce secondary dormancy in late embryo growth stages. The desiccation tolerance of imbibed seeds of most of the nine species may show their adaptability to climate change in the Mediterranean region.

The daffodil Narcissus pseudonarcissus L. contains alkaloids of pharmaceutical interest. Wild daffodil populations have diverse genetic backgrounds and various genetic traits of possible importance. Developing protocols for plant production from seeds may ensure the availability of a large reservoir of individuals as well as being important for species with bulbs that are difficult to acquire. The closely related Narcissus pseudonarcissus subsp. munozii-garmendiae and subsp. nevadensis were investigated in this study because the alkaloids isolated from both are of high pharmacological interest. At the dispersal time, the seeds of both were dormant with underdeveloped embryos, i.e., morphophysiological dormancy (MPD). Experiments were conducted outdoors and under controlled laboratory conditions. Embryo growth and the percentages of radicle and seedling emergence were calculated under different temperature–light stratifications. In N. munozii-garmendiae, embryo growth occurred during warm stratification (28/14 °C or 25/10 °C) and the radicle then emerged when the temperature decreased, but the shoot was dormant. In N. nevadensis, the seeds germinated when cold stratified (5 °C) and then incubated at cool temperatures. Thus, N. munozii-garmendiae and N. nevadensis exhibit different levels of MPD, i.e., deep simple epicotyl and intermediate complex, respectively. Plant production protocols from seeds were established for both taxa in this study.
