Abstract Pawn mutants of Paramecium tetraurelia lack a depolarization-activated Ca2+ current and do not swim backward. Using the method of microinjection and sorting a genomic library, we have cloned a DNA fragment that complements pawn-B (pwB/pwB). The minimal complementing fragment is a 798-bp open reading frame (ORF) that restores the Ca2+ current and the backward swimming when expressed. This ORF contains a 29-bp intron and is transcribed and translated. The translated product has two putative transmembrane domains but no clear matches in current databases. Mutations in the available pwB alleles were found within this ORF. The d4-95 and d4-96 alleles are single base substitutions, while d4-662 (previously pawn-D) harbors a 44-bp insertion that matches an internal eliminated sequence (IES) found in the wild-type germline DNA except for a single C-to-T transition. Northern hybridizations and RT-PCR indicate that d4-662 transcripts are rapidly degraded or not produced. A second 155-bp IES in the wild-type germline ORF excises at two alternative sites spanning three asparagine codons. The pwB ORF appears to be separated from a 5′ neighboring ORF by only 36 bp. The close proximity of the two ORFs and the location of the pwB protein as indicated by GFP-fusion constructs are discussed.

AbstractWidespread and repeated use of glyphosate resulted in an increase in glyphosate-resistant (GR) weeds. This led to an urgent need for diversification of weed control programs and use of PRE herbicides with alternative sites of action. Field experiments were conducted over a 4-yr period (2015 to 2018) across three locations in Nebraska to evaluate the effects of PRE-applied herbicides on critical time for weed removal (CTWR) in GR soybean. The studies were laid out in a split-plot arrangement with herbicide regime as the main plot and weed removal timing as the subplot. The herbicide regimes used were either no PRE or premix of either sulfentrazone plus imazethapyr (350 + 70 g ai ha−1) or saflufenacil plus imazethapyr plus pyroxasulfone (26 + 70 + 120 g ai ha−1). The weed removal timings were at V1, V3, V6, R2, and R5 soybean stages, with weed-free and weedy season-long checks. Weeds were removed by application of glyphosate (1,400 g ae ha−1) or by hoeing. The results across all years and locations suggested that the use of PRE herbicides delayed CTWR in soybean. In particular, the CTWR without PRE herbicides was determined to be around the V1 to V2 (14 to 21 d after emergence [DAE]) growth stage, depending on the location and weed pressure. The use of PRE-applied herbicides delayed CTWR from about the V4 (28 DAE) stage up to the R5 (66 DAE) stage. These results suggest that the use of PRE herbicides in GR soybean could delay the need for POST application of glyphosate by 2 to 5 wk, thereby reducing the need for multiple applications of glyphosate during the growing season. Additionally, the use of PRE herbicides could provide additional modes of action needed to manage GR weeds in GR soybean.

Abstract Literary Critic and Sinologist, Takeuchi Yoshimi, provides post-colonial and decolonial studies a logic of resistance that seeks to destabilize the colonialist projects of Western modernity without repeating its structural logic. In this regard, Takeuchi's logic of resistance functions as a dialectical lens into the “emancipatory traps” of Western modernity that frame the victim–victimizer paradox by turning negativity into a method of generating heuristic possibilities. But in this pursuit to look for alternative sites for mining theoretical possibilities, Takeuchi returns to the origins of Chinese modernity for imagining a proper logic of Asian resistance, that which could be deployed as a resource for negating the imperial gestures of modernist thought while affirming the positive kernel of the Enlightenment with the hope of bringing forth a global world that is continuously transformed by the cultural particulars themselves. The goal of this article is to further elucidate Takeuchi's logic of Asian resistance and to discuss how this logic can be read as having the potential to correct Nishida Kitarō's and the Kyoto School's failed attempt to overcome modernity.

The Three Gorges Dam Project (TGDP) on the Yangtze River in Hubei Province is touted as the long term solution to China’s increasing energy and agricultural needs. TGDP, with a 1084 km2 reservoir of 39.3 billion m3 capacity, will be the largest multipurpose water conservancy project ever built in the world. The main structures of the project are dam, hydropower stations and navigation facilities. After studying fifteen alternative sites and approval of Environmental Impact Statement (EIS) in February 1992, the construction of the dam began in 1994 with an expected duration of 17 years. Annual reduction of 40-50 million tons of coal consumption by providing 85 billion kilowatt hours of electricity, protection of 1.6 million hectares of agricultural land from floods and considerable improvement of navigation and tourism sector are the major anticipated benefits of TGDP. However, the proposed benefits put significant pressure on ecological and socio-cultural environment of the TGDP area. Relocation and resettlement of over 1.3 million local populations, inundation of several sites of historical importance, deforestation and loss of biodiversity including several endangered species of flora and fauna across a region of 58,000 km2, increased sedimentation, water pollution and potential earthquake risks are the major adverse consequences of TGDP. In large-scale gradient studies, selection of the best research sites is critical but time-consuming and costly. Multivariate methods can be used to quickly identify suitable sites from existing data bases. Based on a study of acid deposition in the Great Lakes region (the Michigan Gradient Study), we illustrate the use of multivariate methods in screening potential research sites for similarity. Sites were examined using cluster analysis, principal coordinates analysis, and correspondence analysis. The graphical displays generated by the multivariate methods were used to identify similar sites across the gradient. A list of 31 potential sites was reduced to 5 similar research sites and several alternative sites. The results of the multivariate methods compared well with more traditional methods of research site selection but allowed for multiple comparisons of many potential sites using a variety of data from existing data bases. By eliminating sites that are unacceptable with respect to available data, the multivariate methods reduce the number of sites that require field visitation prior to final site verification. This process represents a substantial savings in time and effort when dealing with a long list of potential research sites.

INTRODUCTION The use of laparoscopy as a diagnostic and therapeutic tool is being used increasingly in the emergency setting with many of these procedures being performed by trainees. While the incidence of iatrogenic injuries is reported to be low, we present six emergency or expedited cases in which the bladder was perforated by the suprapubic trocar. CASES Three cases were related to the management of appendicitis, two to negative diagnostic laparoscopies for lower abdominal pain and one to an ectopic pregnancy. Management of the bladder injuries varied from a urinary catheter alone to laparotomy with debridement of the abdominal wall due to sepsis and later reconstruction. Four of the six cases were performed by registrars. CONCLUSIONS Although the incidence of bladder injury is low, its importance is highlighted by the large number of laparoscopies being performed. In addition to catheterisation of the patient, care must be taken with the insertion of low suprapubic ports and consideration should be made regarding alternative sites. Adequate laparoscopic supervision and training in port site planning is required for surgical trainees.

AbstractThe transport activity of the calcium ATPase SERCA is modulated by an inhibitory interaction with a 52-residue transmembrane peptide, phospholamban (PLB). Biochemical and structural studies have revealed the primary inhibitory site on SERCA, but PLB has been hypothesized to interact with alternative sites on SERCA that are distinct from the inhibitory site. The present study was undertaken to test these hypotheses and explore structural determinants of SERCA regulation by PLB. Steered molecular dynamics (SMD) and membrane protein-protein docking experiments were performed to investigate the apparent affinity of PLB interactions with candidate sites on SERCA. We modeled the relative binding of PLB to several different conformations of SERCA, representing different enzymatic states sampled during the calcium transport catalytic cycle. Overall, the SMD and docking experiments suggest that the canonical binding site is preferred, but also provide evidence for alternative sites that are favorable for certain conformational states of SERCA.
