Studies of rotavirus particles in humans, monkeys and various non-primates with acute gastroenteritis have involved detection of virus in fecal material by electron microscopy. The EM techniques most commonly employed have been the conventional negative staining (Fig. 1) and immune aggregation (Fig. 2) procedures. Both methods are somewhat insensitive and can most reliably be applied to samples containing large quantities of virus either naturaLly or as a result of concentration by ultracentrifugation. The formation of immune complexes by specific antibody in the immune aggregation procedures confirms the rotavirus diagnosis, but the number of particles per given microscope field is effectively reduced by the aggregation process. In the present communication, we describe use of an on-grid immunoelectron microscopic technique in which rotavirus particles are mounted onto microscope grids that were pre-coated with specific antibody. The technique is a modification of a method originalLy introduced by Derrick (1) for studies of plant viruses.

Waste valorization represents one of the main social challenges when promoting a circular economy and environmental sustainability. Here, we evaluated the effect of the polyphenols extracted from apple peels, normally disposed of as waste, on the amyloid aggregation process of κ-casein from bovine milk, a well-used amyloidogenic model system. The effect of the apple peel extract on protein aggregation was examined using a thioflavin T fluorescence assay, Congo red binding assay, circular dichroism, light scattering, and atomic force microscopy. We found that the phenolic extract from the peel of apples of the cultivar “Fuji”, cultivated in Sicily (Caltavuturo, Italy), inhibited κ-casein fibril formation in a dose-dependent way. In particular, we found that the extract significantly reduced the protein aggregation rate and inhibited the secondary structure reorganization that accompanies κ-casein amyloid formation. Protein-aggregated species resulting from the incubation of κ-casein in the presence of polyphenols under amyloid aggregation conditions were reduced in number and different in morphology.

A model of the transition layer between the shell and the core of a ceramic matrix composite from coal waste and clay has been developed. The chemical, granulometric and mineral compositions of the beneficiation of carbonaceous mudstones and clay were studied. The technological and ceramic properties of raw materials for the samples manufacturing were determined. The method of manufacturing multilayer ceramic samples from coal waste, clay and their mixture is given. The number of transition layers in the contact zone between the clay shell and the core from coal wastes is determined. The deformation and swelling phenomena of model samples from coal wastes, clay, and their mixtures were revealed at the firing temperature of more than 1000 °C. The formation of a reducing ambient in the center of the sample with insufficient air flow is shown. The influence of the carbonaceous particles amount and the ferrous form iron oxide in the coal wastes on the processes of expansion of multilayer samples during firing has been established.

We have synthesized a set of nanostructured carbon samples including a variety of carbon nanotubes and carbonaceous particles, by catalytic thermal decomposition of CH4 on catalyst LaNi 5 powder with different reaction temperatures. Products obtained at reaction temperatures 550~900°C were characterized by means of HR-TEM, SEM and Raman Scattering. In addition, electrochemical charge–discharge cycling method was carried out at room temperature to measure the reversible hydrogen capacity in pressed electrodes containing mixture of catalyst, nanostructured carbon samples and carbonaceous particles. Results showed that the abundance ratio of well-crystallized graphite to amorphous carbon in each product increases with increasing reaction temperatures. This preliminary study showed also that the hydrogen storage capacity of synthesis products measured in an electrochemical half-cell at room temperature correlates with the nanostructure and morphology of the variety of nanostructured carbon samples. Additionally, the hydrogen adsorption capacity against specific surface area (BET) for synthesis products produced at temperatures higher than 670°C is ranging from 14 to 25 wt.%/(1000 m2/g).
