AbstractPotato invertase inhibitor (StInvInh2) positively regulates cold-induced sweetening (CIS) resistance by inhibiting the activity of vacuolar invertase. The distinct expression patterns of StInvInh2 have been thoroughly characterized in different potato genotypes, but the related CIS ability has not been characterized. The understanding of the regulatory mechanisms that control StInvInh2 transcription is unclear. In this study, we identified an ERF‐VII transcription factor, StRAP2.3, that directly regulates StInvInh2 to positively modulate CIS resistance. Acting as a nuclear-localized transcriptional activator, StRAP2.3 directly binds the ACCGAC cis-element in the promoter region of StInvInh2, enabling promoter activity. Overexpression of StRAP2.3 in CIS-sensitive potato tubers induced StInvInh2 mRNA abundance and increased CIS resistance. In contrast, silencing StRAP2.3 in CIS-resistant potato tubers repressed the expression of StInvInh2 and decreased CIS resistance. We conclude that cold-responsive StInvInh2 is due to the binding of StRAP2.3 to the ACCGAC cis-element in the promoter region of StInvInh2. Overall, these findings indicate that StRAP2.3 directly regulates StInvInh2 to positively modulate CIS resistance, which may provide a strategy to improve the processing quality of potatoes.

Работа посвящена разработке и тестированию универсальных праймеров для ПЦР-амплификации полноразмерных генов-ортологов β-фруктофуранозидазы (кислой вакуолярной инвертазы) у видов и сортов картофеля (Solanum tuberosum). Крахмал – основной источник энергии и резервный углевод, накапливающийся в амилопластах клубней. Образовавшаяся в результате фотосинтеза молекула глюкозы при реакции с фруктозой образует сахарозу – основную транспортную форму углеводов в растении. В клубни сахароза доставляется по флоэме (апопластный путь), где в межклеточном пространстве расщепляется до глюкозы и фруктозы, которые затем проникают в клетки паренхимы. Глюкоза служит в дальнейшем субстратом для синтеза крахмала в амилопластах. Однако при воздействии пониженных температур крахмал в клубнях картофеля разрушается до редуцирующих сахаров. Параллельно этому процессу идет ресинтез сахарозы до глюкозы и фруктозы за счет фермента кислой вакуолярной инвертазы (β-фруктофуранозидазы), кодируемой геном Pain-1. В совокупности эти процессы приводят к избыточному накоплению моносахаров в клубнях картофеля, так называемому холодовому осахариванию (cold-induced sweetening). При этом создаются условия для интенсивного образования меланоидинов, вызывающих потемнение мякоти картофеля, что значительно ухудшает товарное качество продукта. Таким образом, изучение гена Pain-1, кодирующего вакуолярную инвертазу, а именно, его идентификация и анализ структуры – важная задача, необходимая для поиска доноров, устойчивых к холодовому осахариванию. Первоочередная задача для этого – разработка и тестирование праймерных комбинаций, позволяющих амплифицировать полноразмерный ген у диких видов картофеля, а также сортов и линий культивируемого картофеля (S. tuberosum). В данной работе приведены результаты разработки и тестирования универсальных праймеров, с помощью которых можно амплифицировать как полноразмерные гены-ортологи, так и фрагменты гена Pain-1, а также подобраны оптимальные условия для проведения ПЦР реакции. Было разработано 6 праймерных комбинаций (PainF – PainR, PainF – Pain1exR, Pain1exF – Pain3exR, Pain2inF – Pain2inR, Pain3exF – Pain5exR, Pain5exF – PainR), среди которых комбинация PainF – PainR позволяла амплифицировать полноразмерный ген, остальные – внутренние и будут использованы в дальнейшем при секвенировании фрагментов исследуемого гена. Эти праймеры были успешно протестированы на 15 образцах, включающих представителей пяти дикорастущих видов картофеля (S. gourlay, S. chacoense, S. pinnatissectum, S. stoloniferum, S. vernei) и десяти сортов российской и зарубежной селекции (Гала, Ласунок, Ред Скарлетт, Рассет Бербанк, Мирас, Башкирский, Жуковский ранний, Матушка, Елизавета, Сударыня).The purpose of research is design and testing of universal primers for PCR amplification of full-length-fructofuranozidase orthologs genes (acid vacuolar invertase) in wild species and potato (Solanum tuberosum) varieties. In the tuber, sucrose is delivered via phloem (apoplast), where it splits into glucose and fructose, which then go to the parenchyma cells. Glucose is a further substrate for the starch synthesis in amyloplasts. However, low temperatures influence on potato tubers leads to starch break down to reducing sugars. In parallel to this process there is happens resynthesis of sucrose to glucose and fructose by acid vacuolar invertase enzyme (β-fructofuranosidase) encoded by Pain-1 gene. Together, these processes lead to an excessive accumulation of monosaccharides in potato tubers. This process also called as cold-induced sweetening. It creates conditions for the intensive formation of melanoidins, which cause a potato tubers darkening, which considerably impairs the commercial quality of the product. Thus, the study Pain-1 gene that encodes the vacuolar invertase (its identification and structure analysis) is an important task required for the search of donors resistant to cold-induced sweetening. The primary task for this is the design and testing of primer combinations that allow to amplify the full-length gene in wild potato species, varieties and lines of cultivated potato. In this work, we develop and test universal primers, that can amplify both full-length orthologs and fragments of the Pain-1 gene, and also select the optimal conditions for carrying out the PCR reaction. In total 6 primer combinations were designed (PainF - PainR, PainF - Pain1exR, Pain1exF - Pain3exR, Pain2inF - Pain2inR, Pain3exF - Pain5exR, Pain5exF - PainR), where PainF - PainR primer combination allowed to amplify a full-sized gene, the rest are internal and will be used in the further fragments sequencing of the β-fructofuranosidase gene. These primers were successfully tested on 15 samples, including five wild species of potato (S. gourlay, S. chacoense, S. pinnatissectum, S. stoloniferum, S. vernei) and ten varieties of Russian and foreign breeding (Gala, Lasunok, Red Scarlet , Rasset Burbank, Miras, Bashkirsky, Zhukovsky ranniy, Matushka, Elizaveta, Sudaryna).

The presence of awns on the ear is associated with a number of important plant properties, such as drought resistance, quality of the grain mass during processing, etc. The main manifestations of this trait are controlled by the B1 gene, which has recently been identified and encodes the C2H2 zinc finger transcription factor. Based on the previously identified SNPs in the promoter region of this gene, we constructed markers for dominant and recessive alleles which determine awnless and awned phenotypes, respectively. The markers were successful for use in targeting the respective alleles of the B1 gene in 176 varieties of common wheat, accessions of T. spelta L., as well as on F2/F3 hybrids from crosses between awned and awnless forms of T. aestivum. We first identified a new allele, b1mite, which has both an insert of a miniature Stowaway-like transposon, 261 bp in length, and 33 novel SNPs in the promoter region. Despite these changes, this allele had no effect on the awned phenotype. The possible mechanisms of the influence of the analyzed gene on phenotype are discussed.

Research Highlights: Sequence phylogeny, genome organisation, gene structure, conserved motifs, promoter cis-element and expression profiling of poplar NACs related to salt stress were detected. In addition, expression of two salt-induced NACs was analysed. Background and Objectives: NAC transcription factor (TF) proteins are involved in a wide range of functions during plant development and stress-related endurance processes. To understand the function of Populus NAC TFs in salt stress tolerance, we characterised the structure and expression profile of a total of 289 NAC members. Materials and Methods: Sequence phylogeny, genome organisation, gene structure, motif composition and promoter cis-element were detected using bioinformatics. The expression pattern of Populus NAC TFs under salt stress was also detected using RNA-Seq and RT-qPCR. Results: Synteny analysis showed that 46 and 37 Populus NAC genes were involved in whole-genome duplication and tandem duplication events, respectively. The expression pattern of Populus NAC TFs under salt stress showed the expression of the 289 PtNACs of 84K poplar was induced. Similar expression trends of NACs were found in Populus simonii × P. nigra T. S. Hwang et Liang and Arabidopsis thaliana (L.) Heynh. Conclusions: The correlation analysis showed that the expression of two differentially expressed NAC genes PtNAC024 and PtNAC182 was significantly associated with most of the 63 differentially expressed genes tested. The expression of PtNAC024 and PtNAC182 in different tissues was also analysed in silico and different expression patterns were found. Together, this study provides a solid basis to explore stress-related NAC TF functions in Populus salt tolerance and development.

AbstractControl of potato sprouting is important to ensure constant supply of high-quality potato to the industry. Efficient control of sprouting can be achieved by chemical treatment or cold temperature. Recent bans on anti-sprouting molecules are prompting the use of cold storage in the potato value chain. Unfortunately, storage of potato at low temperatures is associated with cold induced sweetening (CIS) due to the induction of the vacuolar invertase gene under low temperatures. Because CIS is associated with the production of the potentially carcinogenic acrylamide in processed potatoes, concise knowledge on the regulatory mechanisms controlling the CIS-phenotype in potatoes is expected to help pave the way for the production of CIS-resistant potato varieties. Here, we dissect the promoters of the Vacuolar invertase (Vinv) genes from CIS-susceptible and CIS-resistant varieties to investigate their implication in CIS-phenotype determination. Using bisulfite sequencing and CRISPR-dCas9-DRM2-mediated de novo DNA methylation, we show that the CIS-resistant phenotype of Verdi, is in part due to hypermethylation of its Vinv promoter, more specifically in the 1.0-1.7kb region. Those findings open new perspectives to engineer CIS-resistant potatoes by genome and epigenome modifications.

Пахотные угодья Республики Коми в основном представлены дерново-подзолистыми почвами с низким естественным плодородием, которые без применения удобрений быстро подвергаются деградационным процессам, что резко снижает их продуктивность. Цель исследований: оценка эффективности долговременного применения органических и минеральных удобрений при выращивании картофеля в кормовом севообороте в условиях Республики Коми. Методической основой выполнения работ были комплексные исследования дерново-подзолистых почв, клубней картофеля на участках многолетнего полевого эксперимента, заложенного на землях Института агробиотехнологий ФИЦ Коми НЦ УрО РАН в соответствии с «Методическими указаниями географической сети опытов с удобрениями». Длительный период исследований (более 40 лет) характеризовался различными климатическими условиями, которые отразились на количестве урожая и качестве клубней картофеля. Было проведено 7 ротаций (14 лет) с выращиванием картофеля. Вносили органические удобрения – торфонавозный компост (ТНК) в дозах 40 и 80 т/га (1 и 2 фон, соответственно) под картофель и минеральные удобрения – NPК в дозах 1/3; 1/2 и 1 для восполнения выноса растениями элементов питания. Установлено положительное влияние комплексного применения органических и минеральных удобрений на урожайность и качество картофеля, а также на плодородие дерново-подзолистой легкосуглинистой среднеокультуренной почвы в кормовом севообороте в почвенно-климатических условиях Республики Коми. В результате применения шестипольного севооборота и комплексного внесения удобрений с 1978 года к 2019 году в почвах снизилась обменная и гидролитическая кислотность; повысилось содержание гумуса на 0,5% (в контроле); на 0,2–0,5% (с тремя дозами минеральных удобрений); на 0,3–1,1% (с ТНК); на 0,2–1,6% (при совместном применении органических и минеральных удобрений). Наибольшие урожаи картофеля получены при совместном применении органических (80 т/га) и минеральных удобрений (1 NPК) и составили 37,1 т/га клубней (8,5 тыс/га кормовых единиц). При таком соотношении удобрений показано высокое качество клубней картофеля: содержание крахмала 15,5%, витамина С – 19,4 мг%, сырого протеина – 14,1%. Количество нитратного азота не превышало ПДК (ПДК 250 мг.с.м.). Экономические расчеты показали, что при внесении ТНК 40 т/га + 1 NPК в дерново-подзолистые почвы с.-х. использования в среднетаежной зоне Республики Коми при выращивании картофеля, условный чистый доход составит 68,4 тыс. р. с 1 га, себестоимость 1 т картофеля – 2,8 тыс. р., рентабельность 188,7%. The arable land of the Republic of Komi is mainly represented by dern-sub-ground soils with low natural fertility, which without the use of fertilizers are quickly subjected to degradation processes, which dramatically reduces their productivity. The aim of the research is to evaluate the efficiency of long-term use of organic and mineral fertilizers in the growing of potatoes in feed crop rotation in the conditions of the Komi Republic. The methodological basis for carrying out the works was comprehensive studies of dern-subhead soils, potato tubers on the sites of a multi-year field experiment laid on the lands of the Institute of Agrobiotechnology of the FRC Komi SC UB RAS in accordance with the «Methodological Instructions of the Geographical Network of Experiments with Fertilizers». A long period of research (more than 40 years) was characterized by different climatic conditions, which affected the number of crops and the quality of potato tubers. There were 7 rotations (14 years) with potato cultivation. Organic fertilizers – peat-avous compost (TNK) in doses of 40 and 80 t/ha (1 and 2 background, respectively) for potatoes and mineral fertilizers – NPK in doses of 1/3; 1/2 and 1 to replenish the carry-out of food elements by plants. The positive effect of the integrated use of organic and mineral fertilizers on the yield and quality of potatoes, as well as on the fertility of soddy-podzolic light loamy medium cultivated soil in fodder crop rotation in the soil and climatic conditions of the Komi Republic, has been established. As a result of the use of six-bed crop rotation and the integrated application of fertilizers, metabolic and hydrolytic acidity decreased in soils from 1978 to 2019; humus content increased by 0.5% (in control); 0.2–0.5% (with three doses of mineral fertilizers); 0.3–1.1% (with TNCs); 0.2–1.6% (with combined use of organic and mineral fertilizers). The largest potato yields were obtained from the combined use of organic (80 t/ha) and mineral fertilizers (1 NPK) and amounted to 37.1 t/ha tubers (8.5 thousand/ha feed units). With this ratio of fertilizers, the high quality of potato tubers was also determined: the starch content was 15.5%, vitamin C – 19.4 mg%, crude protein – 14.1%. The amount of nitrate nitrogen did not exceed the MPC (MPC 250 mg.s.m.). Economic calculations showed that if TNK is added 40 t/ha 1 NPK to the dern-sub-ground soils of agricultural use in the medium-sized zone of the Komi Republic when growing potatoes, the conditional net income will be 68.4 thousand rubles from 1 ha, the cost of 1 t of potatoes – 2.8 thousand rubles, profitability 188.7%.
