Abstract High variability in somatic embryo (SE)-forming capacity has previously been observed in several spinach cultivars. Such variability frequently accounted for more variation in embryogenic response of the explants than the factor being investigated. Hence, the variability in embryogenic capacity was examined in the present study at both the population and the single-seedling level, using seeds of spinach cultivar Matador obtained from nine European seed companies. Seed population obtained from Slovenia (Sl) was superior to others, with the highest regeneration frequency (100%) and the highest mean SE number (14.4). A total of 82% of these seedlings had 80–100% of regenerating explants, while in populations with intermediate embryogenic capacity approximately 40% of seedlings had 20–60% of regenerating explants. The explants from the majority of seedlings (52–100%) in the least responsive populations were irresponsive. Furthermore, the explants from Sl seedlings regenerated from 10–20 (43.5%) up to > 20 (27.6%) SEs on average, while the explants from the majority of seedlings belonging to other populations regenerated 1–10 SEs. The present study strongly indicates that the variability of plant material must not be overlooked, because choosing more responsive individuals for one treatment and less responsive ones for another may lead to misinterpretation of the data.

AbstractIn vitro cultures of Leucojum aestivum are considered as an alternative for the production of galanthamine, which is used for the symptomatic treatment of Alzheimer’s disease. We studied the effects of auxins 2,4-dichlorophenoxyacetic acid (2,4-D), 4-amino-3,5,6-trichloropicolinic acid (picloram), 3,6-dichloro-o-anisic acid (dicamba) at concentrations of 25 and 50 µM on the induction of embryogenic callus and its capacity to induce somatic embryogenesis and alkaloid accumulation. The embryogenic response of the explants was from 30% for 25 µM of dicamba to 100% for picloram (for both 25 and 50 µM). 2,4-D (50 µM) stimulated greater callus proliferation and somatic embryo induction as compared to the other auxins. Polyethylene glycol (PEG) stimulated somatic embryo maturation. Callus grown on media containing 50 µM of auxins produced fewer phenolic compounds as compared with callus grown on media containing 25 µM of auxins. GC-MS analyses showed seven alkaloids in the in vivo bulbs and two to four in callus culture. Galanthamine was detected in callus cultivated with 2,4-D (25, 50 µM), picloram (25 µM), and dicamba (50 µM). Other alkaloids, trisphaeridine, tazettine, and 11-hydroxyvittatine were accumulated only in callus growing on medium with picloram (50 µM).

DNA barcoding is a relatively new method of identifying plant species using short sequences of chloroplast DNA. Although there is a large number of studies using barcoding on various plant species, there are no such studies in the genus Secale. In this study the plant material consisted of 10 cultivated and non-cultivated species and subspecies of rye genus. Three chloroplast DNA regions (rbcL, matK, trnH-psbA) were tested for their suitability as DNA barcoding regions. Universal primers were used, and sequenced products were analyzed using Neighbor Joining and the Maximum Likelihood in the MEGA 7.1 program. We did not observe high variability in nucleotide sequences within the matK and rbcL regions. Only 2.2% of the sequences showed polymorphism in the rbcL region, while 6.5% in the matK region. The most variable trnH-psbA (15.6%) intergenic region was the most useful for rye barcoding. Individual application of the studied regions did not provide the expected results. None of the regions used in the study allowed the division of rye species and subspecies according to the adopted classification of the genus Secale. The results confirm that the use of matK and rbcL is insufficient for DNA barcoding in rye species, and better discrimination within the genus Secale can be obtained only in combination with the non-coding trnH-psbA sequence. Our results also indicate the necessity of using a different region. All of the new sequences have been deposited in Genbank.

Aim of study: Nickel (Ni) and Copper (Cu) are essential metals for the growth and development of plants. In view of the above, the aim of this work was to quantify and correlate Ni and Cu concentrations in the leaf and the parts of the fruit [pod husk, pulp, tegument (seed coating) and cotyledons] of clonal cacao genotype PH 16.Area of study: Cacao genotypes were collected from adult plants grown on farms located in three different climatic regions of southern Bahia, Brazil.Material and methods: Plant material was collected in four plots of twenty farms, located under different edaphic and topographic conditions. They were subjected to chemical analysis and later to statistical analyses.Main results: There was high variability of Ni and Cu concentrations in all evaluated plant materials. Leaf, pulp, and tegument were the plant materials that accumulated more Ni. On the other hand, the greatest accumulation of Cu occurred in the tegument and in the pod husk, while in the cotyledons there was little accumulation of these metals. The concentrations of Ni were influenced by the three climatic regions, a fact not observed for Cu, except at the leaf level. There was interdependence between the accumulation of Ni in the leaves and in the different parts of the fruit, a fact not observed for Cu.Research highlights: Since Ni and Cu accumulated in low concentrations in the cacao beans cotyledons, raw material for the manufacture of chocolate and other food products, these metallic elements do not affect the consumers' health.

The Bio-efficacy of eleven plant extracts namely viz.Neem Kernel; Rhizome of Ginger; Leaves of Datura, Gajarghas, Harsingar, Oak and Latjeera; Bulb of Garlic and Onion; Flowers of Chrysenthemum and Fruits of Chilli in the concentration of 5 percent and imidacloprid @ 40 g ai/ha was tested against mustard aphid, Lipaphiserysimi and their effect on D. rapae and Coccinellid beetle were tested in the Department of Entomology, College of Agriculture, Gwalior (M.P.). All the tested plant materials and imidacloprid @ 40 g ai/ha were effective significanty in reducing the aphid population over control.The aphid population in treated plots ranged from 7.2 to 40.0 as against 85.4 aphid/twig in untreated control. Among the plant material, three sprays of Neem Kernel were found most effective followed by three sprays of chilli fruits.All the plant extracts were found significantly safer to D. rapae and coccinellid bettle in comparision to insecticide (imidacloprid).

In evaluations of clover performances on summer-dry Himatangi sandy soil, it was found that none could match lucerne over summer. Emphasis was therefore placed on production in autumn-winter- early spring when lucerne growth was slow. Evaluations of some winter annual clover species suggested that Trifolium spumosum, T. pallidum, T. resupinatum, and T. vesiculosum would justify further investigation, along with T. subterraneum which is already used in pastures on this soil type. Among the perennial clover species, Kenya white clover (7'. semipilosum) showed outstanding recovery from drought and was the only species to produce significantly in autumn. However, it failed to grow in winter-early spring. Within red clover, materials of New Zealand x Moroccan origin substantially outproduced the commercial cultivars. Within white clover, material from Israel, Italy and Lebanon, as well as progeny of a selected New Zealand plant, showed more rapid recovery from drought stress and subsequently better winter growth than New Zealand commercial material ('Grasslands Huia'). The wider use of plant material of Mediterranean origin and of plants collected in New Zealand dryland pastures is advocated in development of clover cultivars for New Zealand dryland situations.

The article provides analysis of existing structures of lock supports for fixation of plug-in rod-type downhole pumps. Based on the analysis, the connection of the lock support elements and the pump is evaluated. Description is given of the new structure of the lock support equipped with a set of spring-loaded and radially extendable sliders. The latter have conical surfaces interacting in forward and reverse directions with pump armature. The proposed design has high variability, which allows to perform modeling of the product both at the stage of design and at the stage of implementation.

Genetic variability of domestic dogs is a source for effective process of breed formation and creating unique gene complexes. In the world, for preservation of genetic resources of dogs, there are dog training associations which have great confidence: American Club Dog Breeders (AKC), the British Kennel Club (KC) and the Federation Cynologique Internationale (FCI), aimed at protecting breeding dogs, standards creation, registration of a breed, and issuance of accurate pedigrees. Evaluation of the genetic diversity of dog breeds is able to significantly complement and improve their breeding programs. Since breeds of dog differ in morphological and economic characteristics, the problem of finding of the breed features in the genome of animals is becoming more topical. From this point of view, French Bulldog is an interesting breed of dog (FRANC.BULLDOGGE, FCI standard number 101) which belongs by the classification of breeds, adopted in FCI, to the group IX – a dog-companion for health and fun, but to a subgroup of fighting dogs of a small format. French Bulldog breed has been researched slightly not only in Ukraine and also abroad, as the main work of all dog association is focused on solving theoretical and practical issues of breeding, keeping, feeding, veterinary protection and others. The study was conducted at Research Department of Molecular Diagnostic Tests of Ukrainian Laboratory of Quality and Safety of Agricultural Products. 33 animals of French Bulldog breed, admitted to use in dog breeding of Ukrainian Kennel Union (UKU), were involved for the genetic analysis using DNA markers. The materials of the research were buccal epithelial cells, selected before the morning feeding of animals by scraping mucous membrane of oral cavity with disposable, dry, sterile cotton swab. Genomic DNA was extracted using KIT-set of reagents for DNA isolation according to the manufacturer's instructions. PEZ1, PEZ3, PEZ6, PEZ8, FHC 2010, FHC 2054 markers, recommended by International Society for Animal Genetics (ISAG), ACN, КC and FCI, were used for research. As a result of research 25 alleles for all the loci were detected in the experimental sample of dogs. The average number of alleles at the locus Na, obtained by direct counting, was 4.16. The most polymorphic loci for this breed were PEZ6 and PEZ3 with 8 and 6 allelic variants. Monomorphic loci were PEZ8 and FHC 2054 which had 4 and 3 alleles and the lowest level of polymorphism was observed for PEZ 1 and FHC 2010 loci in which only 2 alleles were identified. On analyzing the molecular genetic characteristics of dogs of French Bulldog breed, we found a high variability of genotype on rare alleles, which included alleles: M, C, D, E, J, K, L, O, N and representing 60% of the total number of the identified alleles. C, D, E alleles for PEZ3 locus and O allele at PEZ6 locus are unique to the sampling of dogs because they are not repeated in other loci. Typical alleles: N, F, R, I, P, K, M are 40% of the total. But F, R alleles for PEZ3 locus and P allele for locus PEZ6 are not repeated either in standard allelic variants or in rare one, indicating a high information content of these alleles and loci to be used for further monitoring of allele pool, genetic certification and identification of dogs. Microsatellite DNA loci were analyzed as a result of investigations of French Bulldogs and the most informative: PEZ3, PEZ6 and PEZ8 were found, which have high efficiency in individual and breed certification of dogs due to high variability. These data allow further monitoring of the state of genetic diversity of the breed and the development of measures for improvement of breeding to preserve the structure of breeding material. The study of individual and population genetic variability is advisable to continue for breeding of French Bulldogs "in purity" and preserving valuable gene complexes. The results are the basis for further monitoring of the proposed informative panels of microsatellite DNA markers for genotyping dog of French Bulldog breed and their complex evaluation.
