Near-infrared spectroscopy and multivariate analysis techniques were employed to nondestructively evaluate the rancidity of perilla seed oil by developing prediction models for the acid and peroxide values. The acid, peroxide value, and transmittance spectra of perilla seed oil stored in two different environments for 96 and 144 h were obtained and used to develop prediction models for different storage conditions and time periods. Preprocessing methods were applied to the transmittance spectra of perilla seed oil, and multivariate analysis techniques, such as principal component regression (PCR), partial least squares regression (PLSR), and artificial neural network (ANN) modeling, were employed to develop the models. Titration analysis shows that the free fatty acids in an oil oxidation process were more affected by relative humidity than temperature, whereas peroxides in an oil oxidation process were more significantly affected by temperature than relative humidity for the two different environments in this study. Also, the prediction results of ANN models for both acid and peroxide values were the highest among the developed models. These results suggest that the proposed near-infrared spectroscopy technique with multivariate analysis can be used for the nondestructive evaluation of the rancidity of perilla seed oil, especially the acid and peroxide values.

Introduction: Reactivation of Epstein-Barr virus (EBV) is common in allogeneic hematopoietic cell transplantation (HCT). EBV infection leads to post-transplantation lymphoproliferative disease (PTLD), a life-threatening complication in this setting. Frequent molecular monitoring of viral load, especially in high risk patients and pre-emptive use of Rituximab has improved the outcome of EBV infection. However, the expansion of alternative transplants, leads to higher incidence and effective measures are warranted. Methods: We have retrospectively studied the clinical characteristics of EBV reactivation in consecutive patients that underwent HCT between 2007-2019, when pre-emptive administration of Rituximab was a standard of care in our Unit and possible correlations were sought. EBV reactivation was considered when viral load >8500 viral genomic copies (VGC)/ml in whole blood was documented during regular molecular monitoring with RQ-PCR. Patients received treatment with Rituximab, at a scheme according to physician's decision. We considered undetectable levels as resolution of infection. Patients with PTLD proven by lymph node biopsy were treated as previously described by our group. Results: Among 546 HCT recipients, EBV reactivation was detected in 100 patients, that suffered from hematologic malignancy (98) or aplastic anemia (2) and received grafts from matched sibling (23), unrelated (70) or haploidentical donors (12). Haploidentical donors were significantly higher in patients with EBV reactivation compared to our transplant population (12% versus 6%, p<0.001). Eighty-eight patients received myeloablative and 12/100 reduced intensity conditioning. Overall, EBV reactivation was detected at median 65 (20-2970) post-transplant days (median load: 26100, range 8690-2670000 VGC/ml). Rituximab was administered in 74 patients at median 4 (3-158) days post EBV reactivation. Most patients (63/74) received one cycle of Rituximab until undetectable EBV load. Rituximab cycles (median 1, range 0-3) were not associated with outcomes. Relapse of EBV reactivation was noted in 13/100 patients, with greater incidence among patients with later resolution of infection (27 vs 14 days in non relapsed, p<0.01). Late onset neutropenia related to Rituximab was noted in 16/74 patients and significantly correlated with increased EBV loads. Significantly higher viral loads were also noticed among patients who received ATG (44550 vs 20000 VGC/ml) or had haploidentical donors (60800 vs 22750 VGC/ml). Multivariate analysis confirmed that all above factors were independently associated to increased viral load. CMV concurrent reactivation was noted in 47 patients. Patients that received preemptive anti-CMV treatment presented with significantly delayed resolution of EBV infection, probably corresponding to greater immunosuppression. Five patients (two with haploidentical and three with unrelated donors) presented PTLD at 41 days post transplantation. ROC curve analysis identified a cut off of 67150 VGC/ml that predicts PLTD with 80% sensitivity and specificity (green line in Figure). Relapse free survival (RFS), overall survival (OS) and treatment-related mortality (TRM) in the entire cohort were similar regardless the EBV viral load or PTLD [4-year RFS 32.2%, 4-year OS 48.1% in a median follow up 29 months (4-216)]. ATG and chronic GVHD were independently associated with OS in the multivariate analysis (p<0.001, p<0.05 respectively). Similarly, ATG, chronic GVHD and age at transplant were independently associated to higher TRM in multivariate analysis (HR: 0.1, 1.16, 1.03, 95%CI: 0.15-0.5, 0.008-1.16, 1.007-1.05, respectively p<0.05). A trend for higher TRM was also noted among patients with EBV loads higher than 50000VGC/ml. Conclusion: Our study indicates that regular monitoring and use of preemptive therapy is an effective strategy for prevention of EBV related complications. RFS and OS were not associated to severity of EBV reactivation. A useful cut off of EBV load for PTLD prevention was identified (67150 VGC/ml, specificity and sensitivity: 80%). However, expanding use of alternative transplants warrants a more effective treatment strategy. In this setting, use of specific antiviral cytotoxic cell lines could enhance viral specific cell mediated immunity and provide a better outcome in immunocompromised HCT recipients. Для идентификации кофе используют методы газовой и жидкостной хроматографии, которые дают точную и подробную информацию о его химическом составе, однако трудоемки, сложны по пробоподготовке и непригодны для оперативного мониторинга качества. Цель настоящего исследования – разработка и апробация метода идентификации кофе по ботаническому виду, географическому месту произрастания и обжарке с применением Фурье-ИК-спектроскопии и многомерного анализа. В качестве объектов исследования были образцы кофе в зернах, различающиеся по ботаническому виду (арабика/робуста), географическому месту произрастания (Азия/Америка/Африка) и обжарке (жареный/нежареный). Для разработки моделей идентификации кофе в зернах была сформирована база спектральных данных и применены методы многомерного анализа – метод главных компонент (МГК) и дискриминантный анализ (ДА). ИК-спектры образцов кофе регистрировали с помощью Фурье-ИК-спектрометра Bruker ALPHA с алмазным модулем НПВО в диапазоне 4000–400 см–1 при разрешающей способности спектрометра 2 см–1. Спектральные данные были экспортированы из встроенного программного обеспечения OPUS 7.3.5.0 в Excel. При анализе матрицы спектральных данных выявлены наиболее интенсивные полосы поглощения ИК-спектра, приписываемые наличию функциональных групп воды, липидов, полисахаридов, кофеина и хлорогеновой кислоты в кофе. При сравнении ИК-спектров образцов кофеина, декофеинизированного кофе и кофе в зернах выявлены полосы поглощения спектра, которые можно использовать для построения калибровочной модели содержания кофеина в составе кофе в зернах. По спектральным данным МГК построена многомерная модель градации образцов кофе в зависимости от ботанического вида и наличия обжарки. По матрице факторных нагрузок выявлены полосы поглощения спектра, объясняющие различия образцов по ботаническому виду и обжарке и вносящие наибольший вклад в разделение образцов кофе на группы. Методом ДА по 19 переменным – коэффициентам поглощения на волновых числах спектра разработана система классификационных функций градации образцов кофе по географическому месту произрастания. Доказано, что сочетание Фурье-ИК-спектроскопии с методами многомерного анализа можно использовать как быстрый и неразрушающий инструмент для идентификации кофе в зернах. Gas and liquid chromatography methods are used to identify coffee. They provide accurate and detailed information about its chemical composition; however they are time-consuming, complex in sample preparation and unsuitable for operational quality monitoring. The purpose of this study is to develop and test a method for identifying coffee by botanical species, geographical place of growth and roasting using FTIR-spectroscopy and multivariate analysis. Samples of coffee beans were selected as objects of research, differing in botanical type (Arabica/Robusta), geographical place of growth (Asia/America/Africa) and roasting (roasted/not roasted). To develop models for the identification of grain coffee, a spectral database was formed and the methods of multivariate analysis were applied: principal components analysis (PCA), discriminant analysis. The IR-spectra of coffee samples were recorded using a Bruker ALPHA FTIR-spectrometer with a diamond module in the range of 4000–400 cm–1 with a resolution of the spectrometer of 2 cm–1. Spectral data were exported from the OPUS 7.3.5.0 embedded software to Excel. During analysis the matrix of spectral data, the most intense absorption bands of the IR-spectrum were revealed, attributed to the presence of functional groups of water, lipids, polysaccharides, caffeine and chlorogenic acid in grain coffee. By comparison the IR spectra of the samples: caffeine, decaffeinated coffee and grain coffee, absorption bands of the spectrum were revealed, which can be used to build a calibration model of the caffeine content in the composition of coffee beans. Using PCA based on the spectral data, a multivariate model of the gradation of coffee by botanical type and depending on the roast was build. According to the matrix of factor loadings, absorption bands of the spectrum were revealed, explaining the differences between the samples in botanical type and roasting and making the greatest contribution to the division of coffee samples into groups. By the method of discriminant analysis using 19 variables – absorption coefficients at the wave numbers of the spectrum – a system of classification functions for the gradation of grain coffee samples according to the geographical place of growth has been developed. It is proved that the combination of FTIR-spectroscopy with multivariate analysis methods can be used as a fast and non-destructive tool for identifying coffee beans.

The present work analyzes the statistical effectiveness of different acoustic features in the automatic identification of hypernasality. Acoustic features reflect part of information contained in perceptual analysis; in part, due to their estimation is derived directly or indirectly from the vocal cords behavior. Consequently, it is convenient to apply multivariate analysis techniques in determining the effectiveness of voice features. The effectiveness is studied by using multivariate analysis techniques that are meant for feature extraction and feature selection, as well (latent variable models, heuristic search algorithms).

Our approach can be viewed as a combination of representation theorems in partial differential equations and harmonic analysis techniques.
