We have previously shown that donation of blood into anticoagulants containing half the normal amount of citrate results in a dramatic improvement in the stability of coagulation factor VIII and has no adverse effect on the in vitro qualities of red cells or platelets during storage. To confirm the viability of stored cellular components we are now performing autologous survival studies in healthy volunteers using radiolabelled cells from red cells and platelets stored for 35 and 5 days respectively. Results to date indicate a 24 hour survival of 80% for red cells stored at a haematocrit of 0.70 for 35 days. Infusion of Ill-In oxine labelled platelets after storage for 5 days in full or half-strength citrate gave recoveries of 40% and survivals of 7 days. These encouraging results suggest use of halfstrength citrate may be a route to increasing factor VIII supply without any additional donor recruitment. Further in vitro studies have also been performed on cellular components and reveal adequate in vitro quality for half-strength citrate blood held at room temperature for 20 hours prior to component preparation.

Abstract 1. Saline suspensions of human red cells, as well as those of several animal species, were agglutinated by normal saline extracts of the Fava bean. 2. This agglutination was potentiated in titer 100-fold in a medium of 10 per cent acacia, as a diluent. 3. The inhibition of the hemagglutination action of the Fava bean extract by human serum was apparently attributable to the gamma globulin fraction. 4. The Fava bean principle could be transferred from cell to cell, as shown by heat-elution and acacia technics. 5. Fava-sensitized red cells did not exhibit increased susceptibility in the test tube to complement, hemolysin, or osmotic or mechanical fragility. 6. The mechanism of in vivo red cell destruction in Favism is as yet unknown, but a special immunologic susceptibility to the action of the bean’s principle is suspected in certain persons. 7. It is suggested that the relation of acacia to Fava-sensitized red cells may form the basis of a diagnostic test for Favism in the early, acute stages of the disease.

Young adult chimpanzees immunized with human blood products produced circulating antibodies which reacted with human red cells of a certain proportion of chimpanzees. In addition, agglutinins were formed which reacted with the animals' own erythrocytes. That these agglutinins were true autoantibodies was demonstrated by: (a) their ability to sensitize the animals' own erythrocytes at 37°C both in vivo and in intro; (b) the iso-specificity which they displayed toward other chimpanzee red cells; and (c) the fact that they belonged to the γG-class of immunoglobulins. Complement appeared to be bound to the in vivo sensitized cells but no evidence of increased cell destruction was observed. It seemed most likely that these autoagglutinins were produced as a result of active immunization with closely related antigens.
