Abstract Background Spatial interactions and insulation of chromatin regions are associated with transcriptional regulation. Domains of frequent chromatin contacts are proposed as functional units, favoring and delimiting gene regulatory interactions. However, contrasting evidence supports the association between chromatin domains and transcription. Result Here, we assess gene co-regulation in chromatin domains across multiple human cancers, which exhibit great transcriptional heterogeneity. Across all datasets, gene co-regulation is observed only within a small yet significant number of chromatin domains. We design an algorithmic approach to identify differentially active domains (DADo) between two conditions and show that these provide complementary information to differentially expressed genes. Domains comprising co-regulated genes are enriched in the less active B sub-compartments and for genes with similar function. Notably, differential activation of chromatin domains is not associated with major changes of domain boundaries, but rather with changes of sub-compartments and intra-domain contacts. Conclusion Overall, gene co-regulation is observed only in a minority of chromatin domains, whose systematic identification will help unravel the relationship between chromatin structure and transcription.

AbstractThe demand for lightweight construction is constantly increasing. One approach to meet this challenge is the development of hybrid components made of dissimilar materials. The use of the hybrid construction method for bulk components has a high potential for weight reduction and increased functionality. However, forming workpieces consisting of dissimilar materials requires specific temperature profiles for achieving sufficient formability. This paper deals with the development of a specific heating and cooling strategy to generate an inhomogeneous temperature distribution in hybrid workpieces. Firstly, the heating process boundaries with regard to temperature parameters required for a successful forming are experimentally defined. Secondly, a design based on the obtained cooling strategy is developed. Next a modelling embedded within an electro-thermal framework provides the basis for a numerical determination of admissible cooling rates to fulfil the temperature constraint. Here, the authors illustrate an algorithmic approach for the optimisation of cooling parameters towards an effective minimum, required for applicable forming processes of tailored forming.

The presented paper is devoted to an innovative way of teaching mathematics, specifically the subject combinatorics in high schools. This is because combinatorics is closely connected with the beginnings of informatics and several other scientific disciplines such as graph theory and complexity theory. It is important in solving many practical tasks that require the compilation of an object with certain properties, proves the existence or non-existence of some properties, or specifies the number of objects of certain properties. This paper examines the basic combinatorial structures and presents their use and learning using relations through the Placemat method in teaching process. The effectiveness of the presented innovative way of teaching combinatorics was also verified experimentally at a selected high school in the Slovak Republic. Our experiment has confirmed that teaching combinatorics through relationships among talented children in mathematics is more effective than teaching by a standard algorithmic approach.
