Changes in Abscisic Acid Levels During Imbibition and Germination of Non-Dormant and Thermodormant Lettuce Seeds

Non-dormant lettuce seeds (Lactuca sativa L. cv. Great Lakes) have an optimum temperature for germination of about 25°C. When the seeds were imbibed for 20 h at this temperature their content of abscisic acid fell from 98.0 ng per g fresh weight to less than 6 ng per g original fresh weight. When seeds were imbibed at 10 and 32°C, they exhibited dormancy. An inverse relationship was found between the abscisic acid content of the seeds and their percentage germination.

Three alfalfa (Medicago sativa L.) cultivars differing in fall dormancy were examined for endogenous abscisic acid content in relation to fall dormancy. The alfalfa cultivars studied were 'Beaver' (dormant type), 'Lahonton' (semidormant type), and 'Moapa 69' (nondormant type). Alfalfa seedlings were grown in controlled-environment chambers set at a 16-h photoperiod (16 h light: 8 h dark) and 20:15 °C (light:dark) for 55 days. They were cut and then regrown under environmental conditions simulating summer and fall in western Canada. In leaf samples, the ABA level increased from about 200 in 3-week-old plants to more than 1200 ng/g fresh weight in 8-week-old plants. After cutting, abscisic acid levels dropped abruptly and then increased gradually, reaching a peak level at the stage of the appearance of flower buds. The peak of abscisic acid content was highest in 'Beaver', next highest in 'Lahontan', and lowest in the nondormant type, 'Moapa 69'. At the 6th week of regrowth, abscisic acid content in 'Beaver' was found to be 2200 ng/g fresh weight, which is the highest reported level in nonstressed plants of any species. The analysis of variance suggests photoperiod, temperature, and photoperiod × temperature interaction all have a significant effect on alfalfa regrowth and on abscisic acid content. Under long days and cool temperatures, the amount of foliage regrowth and the characteristic of fall dormancy are manifestly correlated. There is no apparent link between endogenous abscisic acid content and fall dormancy.

Thirty-month-old 'Pêra' orange trees grafted on 'Rangpur' lemon trees grown in 100 L pots were submitted to water stress by the suspension of irrigation. CO2 assimilation (A), transpiration (E) and stomatal conductance (g s) values declined from the seventh day of stress, although the leaf water potential at 6:00 a.m. (psipd) and at 2:00 p.m. (psi2) began to decline from the fifth day of water deficiency. The CO2 intercellular concentration (Ci) of water-stressed plants increased from the seventh day, reaching a maximum concentration on the day of most severe stress. The carboxylation efficiency, as revealed by the ratio A/Ci was low on this day and did not show the same values of non-stressed plants even after ten days of rewatering. After five days of rewatering only psi pd and psi2 were similar to control plants while A, E and g s were still different. When psi2 decreases, there was a trend for increasing abscisic acid (ABA) concentration in the leaves. Similarly, stomatal conductance was found to decrease as a function of decreasing psi2. ABA accumulation and stomatal closure occurred when psi2 was lower than -1.0 MPa. Water stress in 'Pera´ orange trees increased abscisic acid content with consequent stomatal closure and decreased psi2 values.
