8 EFFECT OF CIRCULATING LEPTIN LEVELS AT SYNCHRONIZATION ON SUBSEQUENT FIXED-TIMED ARTIFICIAL INSEMINATION PREGNANCY RATES IN CROSSBRED BEEF HEIFERS

It is generally accepted that reproductive efficiency is the limiting factor in the profitability of most livestock operations. The heritabilities of reproductive traits are typically low, so most advances in reproductive efficiency must be achieved through changes in management practices. Recent research indicates that leptin may play a role in the reproductive processes for many domesticated livestock species. Leptin has been shown to affect nutrient intake, metabolic efficiency, and animal adiposity, and may be associated with age at puberty onset and the length of the postpartum interval in cattle. Therefore, the objective of this study was to determine whether circulating leptin concentrations during estrous synchronization would affect subsequent pregnancy rates after fixed-timed AI (FTAI) in crossbred beef heifers. Before the initiation of the spring breeding season, 54 crossbred (Red Angus × Simmental) long yearling heifers, averaging 326 ± 6.3 kg with a mean body condition score (BCS) of 5.2 ± 0.33, a mean reproductive tract score of 2.1 ± 0.57, and a mean ovary score of 2.7 ± 0.76, were synchronized with a controlled internal drug-release (CIDR) vaginal implant and 2 mg of estradiol benzoate (EB) on Day 0, a 25-mg injection of prostaglandin and CIDR removal on Day 7, and 2 mg of EB on Day 8, with FTAI occurring 52 hours post-CIDR removal. Blood samples were collected via jugular venipuncture 4 times: twice during the synchronization protocol starting 2 days post-CIDR insertion, once at insemination, and once 4 days after insemination. All females were inseminated with frozen–thawed semen from the same fertile bull. Pregnancy was determined 45 days postinsemination via transrectal ultrasonography. Twenty females (37%) were confirmed pregnant to FTAI. There was no difference in average weights (324 ± 6.6 v. 323 ± 3.7 kg), BCS (5.1 ± 0.1 v. 5.2 ± 0.07), reproductive tract scores (2.2 ± 0.5 v. 2.1 ± 0.5) or ovary scores (2.7 ± 0.2 v. 2.7 ± 0.1) for the pregnant and nonpregnant females, respectively. However, those females that were pregnant from FTAI had higher (P = 0.02) mean plasma leptin concentrations (3.33 ± 0.22 ng mL–1) compared with heifers that did not conceive to FTAI (2.66 ± 0.19 ng mL–1) across all sampling days. In addition, as early as 2 days post-CIDR insertion, females that became pregnant from FTAI tended to have higher (P = 0.13) plasma leptin concentrations (2.76 ± 0.18 ng mL–1) compared with those heifers that did not conceive to FTAI (2.19 ± 0.15 ng mL–1). These results indicate that circulating plasma leptin levels in beef cattle during synchronization may be associated with subsequent fertility and might be a useful tool to select females to increase the efficacy of FTAI protocols in beef cattle.

AbstractThe objective was to evaluate different oestrous synchronization schemes in beef cattle under range conditions. In experiment 1, 202 heifers averaging 26 months of age were assigned randomly to three treatments: (a) GnRH-PG. (no. = 44) day 0 (D0): injection of GnRH; D7: injection of PGF2α; D0 to D25 oestrous detection (OD) and artificial insemination (AI); (b) OD-PG. (no. = 45) D0 to D4; OD + AI; D5: injection of PGF2α; D5 to D25 OD + AI; and (c)Ovsynch. (no. = 113) D0: injection of GnRH; D7: injection of PGF2α; D9: injection of GnRH and 16 h later AI at fixed time. In experiment 2, 318 non-suckling cows were assigned randomly to three treatments: (a) GnRH-PG. (no. = 106) D0: injection of GnRH; D7: injection of PGF2α; D0 to D25 OD + AI; (b) GnRH-MAP-PG. (no. = 106) D0: injection of a GnRH analogue and insertion of an intravaginal sponge impregnated with 250 mg of medroxyprogesterone acetate; D7: injection of PGF2α and sponge withdrawal; D0 to D25 OD + AI; (c) controls. (no. = 106) OD and AI. The experimental period lasted 25 days and, with exception of the Ovsynch treatment, oestrous detection was carried out twice a day (a.m./p.m.) and inseminations performed 12 h later. The pregnancy rates assessed by ultrasonography 30 days after AI were: experiment 1: (a) 30·0%; (b) 28·6% and (c) 62·3% (P < 0·05); experiment 2: (a) 60·2%; (b) 57·8% and (c) 45·5% (P < 0·05). It is concluded that in heifers and non-suckling cows, oestrous synchronization treatments result in higher pregnancy rates. In non-suckling cows, the addition of a progestagen did not improve the response.

This study evaluated the use of cooled semen in a fixed-time artificial insemination (FTAI) program compared with frozen–thawed semen to improve pregnancy rates in beef cattle. Ejaculates of three bulls were collected and divided into two treatments: (1) frozen–thawed semen and (2) cooled semen. Egg-yolk extender without glycerol was used for the cooled semen treatment. Straws (25 × 106 spermatozoa) were submitted to cooling for preservation at 5°C for 24 h, after which FTAI was performed. Nelore cows (n = 838) submitted to FTAI were randomly inseminated using frozen–thawed semen or cooled semen. There was a 20% increase in the pregnancy per AI (P AI–1) using cooled semen compared with frozen–thawed semen (59.9 ± 4.7 vs 49.4 ± 5.0%; P < 0.005). There was no difference in P AI–1 among the bulls (P = 0.40). The frozen–thawed semen had fewer functional spermatozoa than did the cooled semen when evaluated by sperm motility (61.7 vs 81.0%), slow thermoresistance test (41.7 vs 66.7%) and hypoosmotic swelling test (38.3 vs 53.7%; P < 0.05). The percentage of sperm abnormalities did not differ between the freeze–thawing and cooling processes (18.6 vs 22.1%; P > 0.05). Because there was less damage to spermatozoa and improvement in P AI–1, the use of cooled semen instead of frozen–thawed semen is an interesting approach to increase reproductive efficiency in cattle submitted to a FTAI protocol.

An important proportion of embryonic deaths in cattle occur during the critical period of embryo development and maternal recognition of pregnancy. High serum progesterone concentrations during this period are associated to higher embryo survival, and the use of eCG could enhance luteal activity. Two experiments were carried out to evaluate the effect of administering eCG at the time of removal of a progesterone-releasing device and 14 days after fixed-time artificial insemination (FTAI) in beef cattle. Multiparous suckled cows between 60 and 90 days postpartum (n = 297; Exp. 1), and two-year old prepuberal heifers (n = 689; Exp. 2) with a body condition score (BCS) between 3.5 and 5 (1 to 8 scale) were used. At the beginning of the experiments, none of females used had a corpus luteum detected by ultrasonography. Females received an intravaginal device containing 0.5 g progesterone (DIB 0.5, Syntex, Buenos Aires, Argentina) and 2 mg oestradiol benzoate i.m. (Benzoato de Oestradiol, Syntex). Eight days later, 500 μg of cloprostenol (Ciclase DL, Syntex) and 0.5 mg oestradiol cypionate (Cipiosyn, Syntex) were given i.m. and devices were removed; FTAI was performed 52 to 56 h after DIB removal. At the time of device removal, the animals were randomly allocated to 4 groups, to receive 400 IU of eCG i.m. (Novormon, Syntex) or placebo at device removal and/or 14 days after FTAI, in a 2 × 2 factorial design. Control group did not receive eCG. In Exp. 1, nose plates were placed on calves on the day of DIB insertion and left for 10 days in order to prevent suckling. Pregnancy diagnosis was performed by ultrasonography at 30 and 60 days after FTAI. Data were analysed by logistic regression. Pregnancy rate at 30 days after FTAI was affected by treatment (P < 0.05; Table 1). Late embryonic/fetal loss rates (non-pregnant females at 60 days after FTAI over those pregnant at 30 days) were 1.6% (3/182) for cows in Exp. 1 and 4.8% (14/292) for heifers in Exp. 2; no differences were detected among groups. In conclusion, administration of 400 IU of eCG at device removal increases pregnancy rate, both in anestrus cows and heifers. However, the administration of a second dose of eCG 14 days after FTAI suggests a possible effect on embryo survival within the first 30 days of pregnancy in cows but not in heifers. Two experiments were designed to evaluate the effect of the timing of fixed-time AI (FTAI) in relation to the removal of an intravaginal progesterone-releasing device (1 g of progesterone, DIB, Syntex SA, Buenos Aires, Argentina) on pregnancy rates in Bos indicus × Bos taurus cross-bred heifers. In experiment 1, 285 Bonsmara × zebu cross-bred heifers, between 18 and 24 months of age and with a body condition score (BCS) between 3.0 and 3.5 (1-5 scale) were used. On the day of initiation of treatment (Day 0), the heifers’ ovaries were palpated (92% of them had a CL) and they received a new DIB plus 2 mg of estradiol benzoate (EB; Syntex SA) and 250 μg of cloprostenol (Ciclase DL, Syntex SA). On Day 8, DIB devices were removed and all heifers received 250 μg of Ciclase plus 0.5 mg of estradiol cypionate (ECP; Cipiosyn, Syntex SA). At that time the heifers were randomly divided to receive FTAI between 48 to 49 h, 53 to 54 h, or 58 to 59 h after DIB removal. The heifers underwent FTAI with semen from 4 bulls by 2 inseminators. In experiment 2, 260 heifers from the same group as those used in experiment 1 (87% with a CL) were treated exactly as those in experiment 1, except that previously used DIB was inserted on Day 0. Pregnancy diagnosis was performed 30 days post-fixed-time AI by ultrasonography. The data were analyzed by logistic regression, taking into account the effect of time of FTAI, semen, and inseminator on pregnancy rates. In experiment 1, pregnancy rates were lower (P = 0.04) in the heifers undergoing FTAI between 48 and 49 h after DIB removal (46/95, 48.4%) than those undergoing FTAI 53 to 54 h (61/99, 61.6%) or 58 to 60 h (57/91, 62.6%) after DIB removal. However, no differences in pregnancy rates were found (P = 0.72) in experiment 2 between the 3 treatment groups, with 39/91 (42.9%) for the 48 to 49 h group, 45/89 (50.6%) for the 53 to 54 h group, and 35/89 (43.8%) for the 58 to 59 h group. There was no effect of the semen or inseminator (P > 0.2) in either experiment. We conclude that when Bos indicus × Bos taurus beef heifers are synchronized with new DIB devices and ECP, higher pregnancy rates are obtained in heifers undergoing FTAI late (between 53 to 60 h after DIB removal) than in those undergoing FTAI early (48 to 49 h after DIB removal). However, time of insemination does not apparently affect pregnancy rates when Bos indicus × Bos taurus beef heifers are synchronized with previously used DIB devices and ECP.

We have previously shown that expression of oestrus significantly influences the time of ovulation and pregnancy rates to timed-AI (P/TAI) in beef cattle synchronized with progesterone devices and oestradiol (Bó et al. 2016 Theriogenology 86, 388–396). Furthermore, delaying TAI from 48 h to 54–58 h after device removal improved P/TAI of those animals not showing oestrus by 48 h. The objective of the present study was to determine whether the administration of GnRH to those animals not showing oestrus by 48 h after device removal had an impact on P/TAI. Non-lactating and suckled beef cows and heifers (Bonsmara, Brangus, and Braford; n = 868), with a corpus luteum (CL) or a follicle ≥8 mm in diameter detected by ultrasonography (Mindray DP 30 Vet, 7.5 MHz, China) and body condition score 2 to 4 (1 to 5 scale) were synchronized in 3 replicates. On Day 0, animals received a progesterone device (DIB 0.5 g, Zoetis, Argentina) and 2 mg of oestradiol benzoate (Von Franken, Argentina). On Day 8, DIB were removed and cows received 0.150 mg of D+cloprostenol (Río de Janeiro, Argentina) plus 0.5 mg oestradiol cypionate (Cipiosyn, Zoetis). In addition, all cows were tail-painted on the sacrococcygeal area (CeloTest, Biotay, Argentina) at DIB removal and were observed to determine the expression of oestrus, according to the percentage of paint loss (PL) 48 h later. The animals that showed oestrus (PL > 30%) were recorded and TAI at that time while those with PL ≤ 30% (not in oestrus) were randomly allocated to receive 100 μg of gonadorelin (GnRH; Gonasyn, Zoetis) or no treatment and TAI between 54 to 58 h after DIB removal. Pregnancy was diagnosed by ultrasonography at 45 days after TAI, and data were analysed by logistic regression. There were no significant differences among replicates, inseminators, semen source or animal class (dry cows, suckled cows or heifers; P > 0.11). However, animals showing oestrus by 48 h after DIB removal had higher P/TAI (371/666; 55.7; P < 0.04) than those that did not show oestrus (96/202; 47.5%). Within animals not showing oestrus at 48 h, those that were in oestrus by the TAI at 54 to 58 h had higher P/TAI (71/124, 57.3%; P < 0.01) than those that did not show oestrus within that period (27/78; 34.6%). In addition, P/TAI was higher (P < 0.01) in animals that received GnRH (66/113; 58.4%) than in those that did not receive GnRH (30/89; 33.7%). In conclusion, the expression of oestrus significantly affected P/TAI in beef cattle synchronized with progesterone devices and oestradiol. Furthermore, treatment of animals not showing oestrus by 48 h after DIB removal with GnRH resulted in improved P/TAI performed 6 to 10 h later.

Abstract Apparent ruminal digestibility of forage soybean-based silages, with and without pearl millet, was determined along with evaluation of silages on heifer performance and reproductive function. Fermenters were utilized in a Latin square design and randomly assigned to 1 of the following treatments: 1) control diet of alfalfa haylage (CON), 2) soybean silage (SB) or 3) soybean and pearl millet silage (SB×PM). All diets were formulated to meet or exceed nutrient requirements of replacement beef heifers targeted to gain 0.79 kg/d. These same diets were fed to 90 Angus-Simmental beef replacement heifers [body weight (BW) = 366 kg; body condition score (BCS) = 5.53; age = 377 ± 11 d] 65 d prior to timed artificial insemination (TAI). Heifers were randomly allotted by breed, BCS and BW to 1 of the 3 treatments, with 3 reps/treatment. Diets were terminated 21 d post-TAI and heifers were commingled and placed on a common diet. Pubertal status was determined by progesterone concentrations of 2 blood samples taken 10 d apart prior to both trial initiation as well as initiation of estrous synchronization. Ovulatory follicle diameter was determined at time of breeding by ultrasonography. Pregnancy diagnosis was accomplished 35 and 66 d post-TAI, respectively, to calculate TAI and end of season pregnancy rates. Neither SB nor SB×PM had an effect (P &gt; 0.37) on apparent ruminal digestion of nutrients compared to the CON. Final BW (414 kg; P ≥ 0.10) and BCS (5.28; P ≥ 0.26) for the heifers were similar among treatments. Likewise, there were no differences in TAI (48%; P &gt; 0.43) or overall breeding season (93%; P &gt; 0.99) pregnancy rates. Ovulatory follicle diameters (11.7 mm) was not different (P &gt; 0.19) among treatments. In summary, forage soybean-based silages, with and without pearl millet, was an acceptable alternative forage for developing replacement beef heifers.

Abstract Estrous response and pregnancy rates resulting from fixed-time (FTAI) or split-time (STAI) artificial insemination were compared among heifers following treatment with the 7-d CO-Synch + CIDR® (controlled internal drug release;1.38g progesterone) protocol. Heifers (n = 456) were assigned to balanced treatments based on weight and reproductive tract score (RTS; Scale 1–5). Gonadotropin-releasing hormone (GnRH; 100 µg gonadorelin acetate) was administered coincident with CIDR® insertion. Progesterone inserts were removed after 7 d, and PG (250 mg im cloprostenol sodium) was administered at CIDR® removal. Estrus detection aids (Estrotect®) were applied at the time PG was administered. Estrous status was recorded at FTAI or STAI, and estrus was defined as removal of ≥ 50% of the grey coating from the Estrotect® patch. Heifers assigned to the FTAI treatment received GnRH and were artificially inseminated 54 h after PG administration. In the STAI treatment, only heifers that expressed estrus prior to 54 h were artificially inseminated at that time. For heifers failing to express estrus, AI was postponed 24 h and only those heifers that failed to exhibit estrus by the delayed time (78 h) received GnRH concurrent with AI. Estrous response prior to the standard time of FTAI did not differ between treatments (P = 0.3). Total estrous response was increased (P &lt; 0.001) among heifers assigned to STAI (74%, STAI; 47%, FTAI); however, pregnancy rates resulting from AI were similar between treatments (48%, STAI; 46%, FTAI; P = 0.4), as were pregnancy rates at the end of a 60 d breeding season (P = 0.6). In summary, STAI resulted in a greater estrous response following treatment with the 7-d CO-Synch + CIDR® protocol; however, this did not result in an increase in pregnancy rate compared to heifers that received FTAI.

Reproductive efficiency is closely tied to the profitability of dairy herds, and therefore successful dairy operations seek to achieve high 21-day pregnancy rates in order to reduce the calving interval and days in milk of the herd. There are various factors that impact reproductive performance, including the specific reproductive management program, body condition score loss and nutritional management, genetics of the cows, and the cow comfort provided by the facilities and management programs. To achieve high 21-day pregnancy rates, the service rate and pregnancy per artificial insemination (P/AI) should be increased. Currently, there are adjustments in timed artificial insemination (TAI) protocols and use of presynchronization programs that can increase P/AI, even to the point that fertility is higher with some TAI programs as compared with AI after standing estrus. Implementation of a systematic reproductive management program that utilizes efficient TAI programs with optimized management strategies can produce high reproductive indexes combined with healthy cows having high milk production termed “the high fertility cycle”. The scientific results that underlie these concepts are presented in this manuscript along with how these ideas can be practically implemented to improve reproductive efficiency on commercial dairy operations.

This study aimed to compare the effects of 2 inducers of new follicular wave [oestradiol benzoate (EB) or hCG] administrated at onset of resynchronization protocols initiated 22 days after the first timed AI (TAI) of beef heifers. We evaluated the effect of treatments on corpus luteum (CL) development, serum progesterone (P4) concentrations, and pregnancy rates. A total of 467 grazing Brangus heifers, with 24- to 26-months, 320 ± 10 kg of body weight, and 3.2 ± 0.5 body condition score, were utilised. The resynchronization protocol began 22 days after the first TAI (Day 22). Heifers were randomly allocated into 1 of 3 groups: EB (n = 199), hCG (n = 101), or control untreated (n = 167). The EB- and hCG-treated heifers received an intravaginal P4 device (Procliclar, 0.75 g of P4; Hertape, Brazil) and an IM treatment with 1 mg of EB (Benzoato HC, Hertape) or 1000 IU of hCG (Vetecor 5000, Hertape), respectively. On Day 30, P4 device was removed, and the pregnancy diagnosis was performed. At this time heifers had the CL area (cm2) determined by ultrasound and seric P4 concentration evaluated by chemiluminescence assay. The assay sensitivity was 0.1 ng mL–1, and the intra- and interassay coefficients of variation were 7.9 and 10.6%, respectively. Nonpregnant heifers after the first TAI (EB = 97, hCG = 36) received intramuscularly 150 μg of sodium cloprostenol (Veteglan Luteolítico, Hertape) and 1 mg of oestradiol cypionate (Cipionato HC, Hertape). The second TAI was performed 48 to 52 h after the P4 device removal. On Day 62, heifers were submitted to a second pregnancy diagnosis. No effect of the body condition score (P = 0.28) was observed on the first and second pregnancy per AI. Pregnancy rates after the first TAI were higher (P = 0.03) on hCG-treated heifers (64.4%) than EB (51.3%) or the control group (58.7%). The EB group determined greater (P = 0.04) pregnancy rate after the second TAI (49.5%) than the hCG group (22.2%). The cumulative pregnancy rates were similar (P = 0.46) between groups [EB (72.4%) and hCG (68.3%)]; however, both were greater (P < 0.05) than the control group (58.6%). Heifers treated with hCG (3.42 ± 0.76) presented greater CL area at Day 30 (P < 0.05) than the EB (2.44 ± 0.57) and control (2.61 ± 0.61) groups. The treatment with hCG 22 days after TAI determined greater (12.43 ± 3.48; P < 0.05) serum P4 concentrations on Day 30 than the EB (6.92 ± 3.04) and control (7.29 ± 2.45) groups. Therefore, the use of hCG at the beginning of resynchronization protocol 22 days after the TAI increased pregnancy rate following the first TAI. However, the use of 1 mg of EB determined greater pregnancy rate after the second TAI, and no detrimental effects on CL development, P4 concentration, and on first TAI pre-established pregnancy.
