Accumulation of 8-hydroxydeoxyguanosine and its relationship with antioxidant parameters in patients with Eales’ disease: Implications for antioxidant therapy

Azadirachtin (Aza) used as insecticide due to inhibiting growth of insects and preventing them from feeding on plants. To understand the effects of contamination of this insecticide on phototrophs, and to determine the responses of these organisms against these insecticides are extremely important in understanding how the ecosystem is affected. In this study, chlorophyll-a amount, OD 560 and antioxidant parameters (total SOD, APX, GR, Proline, MDA and H2O2) were determined in order to understand the effect of Aza on Arthrospira platensis Gomont. Aza was applied between 0–20 μg mL−1 concentrations for 7 days in the study. Enzyme analysis was conducted at the end of the 7th day. There was a statistically significant decrease in the absorbance of OD560 and the chlorophyll-a content in A. platensis cultures exposed to the Aza (0–20 μg mL−1) during 7 days due to the increase in pesticide levels. SOD activity decreased at 8, 16 and 20 μg mL−1 concentrations; GR enzyme activity showed a significant decrease compared to the control at a concentration of 20 μg mL−1. APX activity did not change significantly compared to control. The MDA content increased significantly at 16 and 20 μg mL−1 concentrations. The H2O2 content significantly increased at 12, 16 and 20 μg mL−1 concentrations (p < 0.05) while the free proline content decreased at 4 μg mL−1 concentration (p < 0.05). As a result, regarding the Aza concentrations used in this study may be a step to prevent pesticide pollution in the environment.

Objective: To evaluate the effect of cefepime on hematological changes, immunological disorders and hepatic oxidative damage in rats experimentally infected with E.coli ATCC 25922. Design: Randomized controlled experimental study. Animals: Thirty-two adult male albino rats weighting150-200 g. Procedures: Rats used for this study were randomly assigned into 4 equal groups: the control one, E.coli infected group (1×108CFU/I/P/once), the cefepime treated group (45 mg/kg bw/I/M/day) for 5 days and the E.coli infected group that treated with cefepime 24h after bacterial inoculation as previously described. Hematological and immunological parameters, liver function biomarkers and hepatic oxidative stress and antioxidant markers were determined. Results: Our result revealed that E.coli infection induced a significant elevation in the erythrocytes count, hemoglobin concentration, PCV% and total leukocytic count (TLC) (P < 0.05). In the same respect, liver function biomarkers, serum glucose, total cholesterol, and triglyceride levels as well hepatic malondialdehyde (MDA), nitric oxide (NO), TNF-α, IL-10, and lysozyme activity were significantly increased compared to the control rats (P < 0.05). In contrast, hepatic reduced glutathione (GSH), superoxide dismutase (SOD) and catalase (CAT) were decreased significantly (P < 0.05). Cefepime treatment in E.coli + CFPM group reduced the elevated eythrogram, TLC and liver function biomarkers. Cefepime also ameliorated the oxidative damage and inflammatory response induced by E.coli infection. Conclusion and clinical relevance: Cefepime is safe when administered in a fixed-dose and possess antioxidant that contributes to improve efficacy against adverse effect induced by E.coli ATCC 25922 infection.

AbstractExtracts of the fruiting bodies of the Oyster mushroom (Pleurotus ostreatus) grown on wood substrates (beech, oak, linden, walnut, poplar) and extracts of the fruiting bodies of the Oyster mushroom (Pleurotus pulmonarius) grown in nature on aspen wood were used to determine the total phenols, total flavonoids, lycopene and β-carotene. The content of individual antioxidants varies considerably depending, not only on the substrate, but also on the extracting agents. The highest content of total phenols and total flavonoids was found in methanol and water extracts of the fruiting bodies of the Oyster mushrooms grown on oak and linden substrates. The maximum content of lycopene and β-carotene was determined in acetone and n-hexane (ratio 4 : 6) extracts of the fruiting bodies of the Oyster mushroom grown on an oak block. The results obtained in this study demonstrated that the quantitative and also probably the qualitative composition of the antioxidants in the fruiting bodies of Oyster mushrooms depended considerably on the substrate composition.
