Effect of cefepime on hematological, immunological and oxidant/antioxidant parameters in rats experimentally infected with E. coli ATCC 25922

Objective: To evaluate the effect of cefepime on hematological changes, immunological disorders and hepatic oxidative damage in rats experimentally infected with E.coli ATCC 25922. Design: Randomized controlled experimental study. Animals: Thirty-two adult male albino rats weighting150-200 g. Procedures: Rats used for this study were randomly assigned into 4 equal groups: the control one, E.coli infected group (1×108CFU/I/P/once), the cefepime treated group (45 mg/kg bw/I/M/day) for 5 days and the E.coli infected group that treated with cefepime 24h after bacterial inoculation as previously described. Hematological and immunological parameters, liver function biomarkers and hepatic oxidative stress and antioxidant markers were determined. Results: Our result revealed that E.coli infection induced a significant elevation in the erythrocytes count, hemoglobin concentration, PCV% and total leukocytic count (TLC) (P < 0.05). In the same respect, liver function biomarkers, serum glucose, total cholesterol, and triglyceride levels as well hepatic malondialdehyde (MDA), nitric oxide (NO), TNF-α, IL-10, and lysozyme activity were significantly increased compared to the control rats (P < 0.05). In contrast, hepatic reduced glutathione (GSH), superoxide dismutase (SOD) and catalase (CAT) were decreased significantly (P < 0.05). Cefepime treatment in E.coli + CFPM group reduced the elevated eythrogram, TLC and liver function biomarkers. Cefepime also ameliorated the oxidative damage and inflammatory response induced by E.coli infection. Conclusion and clinical relevance: Cefepime is safe when administered in a fixed-dose and possess antioxidant that contributes to improve efficacy against adverse effect induced by E.coli ATCC 25922 infection.

The present study aimed to investigate the ameliorative effects of fenugreek seeds and curcumin on hematotoxicity induced by nicotine in male albino rats. 30 male F-344/NHsd Fischer rats, weighing from 180 to 200g were used in the present study. The animals were divided into five groups (6 rats for each); Group I (control group), Group II (nicotine treated group), Group III (nicotine/fenugreek seeds co-administered), Group IV (nicotine/curcumin co-administered), and Group V (nicotine/curcumin& fenugreek seeds co-administered). At the end of the experimentation and 24 hours after the last dose, all animals were anaesthetized with ether and blood samples were collected by heart puncture. The samples were collected in clean dry tubes containing the anticoagulant substance EDTA and used for the hematological studies. The results showed that the animals treated with nicotine for 4 weeks showed a significant decrease in RBCs count, hemoglobin concentration, hematocrit value, MCH, MCHC, and platelets count, and increased MCV and WBCs count as compared to the control group. Co-administration of nicotine with fenugreek and/or curcumin caused improvement in all hematological parameters when compared with nicotine group. It can be concluded that nicotine had a strong effect on the hematological parameters. The ingestion of fenugreek and/or curcumin prevent the hematoxicity induced by nicotine. The current study suggests that fenugreek and curcumin may be useful in combating free radical-induced hematotoxicity induced by nicotine.

AbstractIn this study, we attempt to verify the claim that the leaf-extract of Telfairia occidentalis can remedy oxidative damage condition as well as assess its phytochemical content. Fifteen male albino rats weighing 180 g to 240 g were randomly divided into three groups of five rats each. Group A was designated the control group while group B and C were both induced with 40 mg/kg body weight 2,4-dinitrophenyl hydrazine. Group C was subsequently treated with 200 mg/kg body weight of ethanol extract of T. occidentalis leaf for 21 days. At the end of the treatment, the animals were sacrificed, and serum of the samples were subjected to relevant tests. Result shows that the plant leaf contained saponin, tannins, alkaloids, flavonoids and phenols whereas, terpenes, steroids and anthraquinones were not detected. The serum enzymes alanine aminotransferase (ALT) and alkaline phosphatase (ALP) were significantly elevated from 17.43 u/L and 28.40 u/L to 21.60 u/L and 34.27 u/L respectively. These were significantly lowered in the group C to 18.37 u/L and 29.23 u/L respectively for ALT and ALP. Also, a significant lowering of superoxide dismutase (SOD) activity was observed in the treated group (54.33 u/mg) from 79.40 u/mg recorded in the intoxicated group. Similarly, a significant decrease in malondialdehyde was observed in the treated group (25.80 u/mg) relative to the intoxicated group (35.87 u/mg). Moreover, catalase activity in the treated group (7.43 u/mg) was significantly lower compared with the intoxicated group. Our observation confirmed that ethanolic leaf extract of T. occidentalis reversed the oxidative damage condition in albino rats. The result confirms the ethnomedicinal use of the plant in the management of oxidative stress related diseases.

Endosulfan is a worldwide used synthetic insecticide that has an important role on management of pests in agriculture. The present work was undertaken to determine the effect of endosulfan on the haematological and haemochemical parameters of albino rats. Rats were feed with 5 mg/kg body weight endosulfan in mixed food stuff for 42 days. The studies were conducted on sexually matured male rats covering five groups of animals with control. Total counts of erythrocytes and haemoglobin were decreased and leucocytes were increased in treated group. Differential counts of leucocytes showed significant increase in basophils and monocytes. The levels of serum glucose, urea, creatinine and bilirubin increased significantly, suggesting that the synthetic insecticide had remarkable toxic effects on the haematological and biochemical parameters in the experimental animals. The study was designed to demonstrate the antimicrobial and antioxidant effects of Oregano against C. freundii. The study consist of two parts, the first part (invitro) including study the sensitivity of C. freundii against safflower extract and antibiotics. where, the results showed very high inhibition efficacy of extract, which reached to 30 mm, while Neomycin inhibition zone reach to 14mm, Novobiocin inhibition zone reach to 9mm and Tetracycline inhibition zone reach to 9mm In the second part (in vivo) used 24 albino rats and divide to six groups (each group consist 4 rats). control group, group injected with (1x105 CFU/ml/25mg) bacteria, group injected with (1x105 CFU/ml/25mg) bacteria and treated with neomycin, group injected with (1x105 CFU/ml/25mg) bacteria and treated with novobiocin, group injected with (1x105 CFU/ml/25mg) bacteria and treated with tetramycine, group injected with (1x105 CFU/ml/25mg) bacteria with extract. The MDA levels in infected group and treated groups with antibiotics increased and GSH levels in infected group and treated groups with antibiotics show decreased (P < 0.05) compared control rats. In treated group with extract, MDA and GSH levels show on-significant change compare with control rats. It was concluded from present study that the Oregano has an antimicrobial and antioxidant activity against C. freundii.
