Background: Holothuria scabra is one of the most commercially important species found in the Pacific region. The sea cucumber extracts have been widely reported to have beneficial health effects. The aim of this study was to determine the nutritional compositions of H. scabra, and compare its important nutritional contents with that of other species.Methods: The sea cucumbers were dissected, sliced into small pieces, and then freeze-dried. The nutritional compositions, including proximate composition, amino acids, fatty acids, collagen, GABA, Vitamin A, C, and E of the whole body and body wall of H. scabra, were analyzed.Results: H. scabra contained a high quantity of protein (22.50% in whole body and 55.18% in body wall) and very low lipids (1.55% in whole body and 1.02% in body wall). The three most abundant amino acids found in both the whole body and body wall were glycine, glutamic acid, and proline. The main fatty acids found in the whole body were stearic acid and nervonic acid, and in the body wall were arachidonic acid and stearic acid. The whole body and body wall also contained high levels of essential amino acids, essential fatty acids, and collagen, in addition to moderate amounts of vitamin E and low amounts of GABA and vitamin C.Conclusions: The sea cucumber, H. scabra, contained high quantity of protein and very low lipid. It contained high essential amino acids, essential fatty acids, nervonic and arachidonic acids, and collagen, which also contained GABA, vitamin C, and vitamin E.Keywords: sea cucumber; Holothuria scabra; nutrition components; functional food

This research is conducted in the coastal waters of Bahoi Village, West Likupang Sub-district, North Minahasa District with the aims to know the types of sea-related environmental factors in the research site and know the community structure of the sea cucumber including the density, Index of diversity, dominance index, and distribution patterns. Data collection was done by using the line transect method, and sampling technique by using quadrat. The results of the study found 5 species of sea cucumber i.e. Holothuria atra,  Holothuria scabra,  Bohadschia marmorata, Actinopyga echinites, and  Bohadschia vitiensis. The individual density of all species is 0.12 ind/m2, and the diversity index (H ') 0.950. The value of the similarity index/compatibility obtained is e = 0.918. The result of the calculated dominance index of sea cucumber species is 1.0 and the pattern of spreading species of sea cucumber in the research site shows a random spread pattern because the value of dispersion index  (I) approaches 1 (one). Keywords: community; Sea cucumber; Bahoi; Dominance; Diversity.AbstrakPenelitian ini dilakukan di perairan pantai desa Bahoi, Kecamatan Likupang Barat, Kabupaten Minahasa Utara dengan tujuan untuk mengetahui jenis-jenis teripang yang ada di lokasi penelitian dan mengetahui struktur komunitas teripang yang meliputi kepadatan, indeks keanekaragaman, indeks dominasi, dan pola penyebaran. Pengambilan data dilakukan dengan metode line transek, dan teknik pengambilan sampel dengan mengunakan kuadrat. Hasil penelitian ditemukan 5 spesies teripang yaitu Holothuria atra, Holothuria scabra, Bohadschia marmorata, Actinopyga echinites dan Bohadschia vitiensis. Kepadatan individu dari seluruh spesies yaitu 0,12 ind/m2, indeks keanekaragaman (H’) 0,950. Nilai indeks kemerataan/keserasian diperoleh nilai indeks e = 0,918. nilai indeks dominasi spesies teripang diperoleh 1,0 dan pola penyebaran spesies teripang dilokasi penelitian menunjukkan pola sebaran acak (random) dikarenakan nilai indeks dispersi (I) mendekati 1 (satu).Kata Kunci : Komunitas; Teripang; Perairan Bahoi; Dominasi; Keanekaragaman

Teripang pasir, Holothuria scabra merupakan salah satu komoditas perikanan yang mempunyai nilai ekonomi tinggi di Asia. Populasinya di alam semakin menurun karena aktivitas penangkapan secara berlebihan. Untuk mengantisipasi menurunnya stok teripang di alam, maka perlu segera dilakukan pengembangannya melalui kegiatan budidaya. Di samping ketersediaan benih, pakan merupakan salah satu faktor yang sangat menentukan keberhasilan budidaya teripang. Penelitian ini bertujuan mendapatkan formulasi pakan buatan yang tepat untuk mendukung pertumbuhan teripang pasir. Empat pakan percobaan dengan formula berbeda, sebagai perlakuan, dibuat dalam bentuk pelet kering. Pakan diformulasi menggunakan tepung Sargassum sp., tepung Ulva sp., tepung Gracilaria sp., yang dikombinasi dengan tepung kedelai dan tepung beras dengan proporsi berbeda. Pada percobaan-1, benih teripang (hasil pembenihan) ukuran 14,4 ± 6,5 g ditebar dalam delapan buah bak berukuran 2 m x 1 m x 0,6 m dengan kepadatan 100 ekor/bak. Teripang diberi pakan percobaan dengan frekuensi satu kali sehari selama lima bulan. Pada percobaan-2, benih teripang ukuran 7,0 ± 1,6 g ditebar dalam delapan buah keramba jaring apung berukuran 1 m x 1 m x 1 m yang diletakkan di tambak dengan kepadatan 50 ekor per jaring. Teripang diberi pakan percobaan dengan frekuensi satu kali sehari selama empat bulan. Hasil percobaan menunjukkan pakan buatan yang diformulasi menggunakan tepung Sargassum sp., tepung Ulva sp., tepung kedelai, dan tepung beras dapat mendukung pertumbuhan dan sintasan teripang pasir, baik yang dipelihara dalam bak maupun dalam jaring apung di tambak. Sintasan teripang tidak dipengaruhi oleh pakan percobaan (P>0,05). Pakan buatan dengan komposisi bahan 30% tepung Sargassum sp., 35% tepung Ulva sp., 4% tepung kedelai, dan 18% tepung beras menghasilkan pertumbuhan terbaik dan dapat diaplikasikan pada pemeliharaan teripang pasir.Sandfish, Holothuria scabra is a highly valued sea cucumber product in Asian markets. Current exploitation has reduced its wild stock to an alarming level. In order to alleviate the over-exploitation to its wild population and provide a reliable supply of market demands, the aquaculture technology of sandfish has to be developed and perfected. Feed is one of the important factors for a successful sea cucumber aquaculture, besides seed supply. The aim of this experiment was to obtain an appropriate feed formulation to support the growth of sandfish. Four experimental diets (dry pellet) were formulated using seaweed meal of Sargassum, Ulva, and Gracilaria, combined with soybean meal and rice flour, each with different proportions. In experiment-1, cultured sandfish juveniles with an initial weight of 14.4 ± 6.5 g were stocked into eight concrete tanks (2 m x 1 m x 0.6 m) with a density of 100 juveniles/tank. In experiment-2, sandfish juveniles with an initial weight of 7.0 ± 1.6 g were stocked into eight floating net cages (1 m x 1 m x 1 m) erected in a pond with a density of 50 juveniles/cage. The sandfish juveniles were fed once daily with the experimental diets for five and four months for experiment-1 and experiment-2, respectively. Results of the experiment showed that diet formulated with Sargassum, Ulva, and soybean meals and rice flour produced good growth and survival of sandfish, both reared in concrete tanks and in floating net cages. The experimental diets did not affect the survival of sandfish (P>0.05). Formulated diet containing 30% Sargassum meal, 35% Ulva meal, 4% soybean meal, 18% rice flour, and 6% ‘lap lap’ flour gave the best growth and could be applied for grow-out of sandfish.

AbstractAquacultivated sea cucumbers often suffer from SKin Ulceration Diseases (SKUDs). SKUDs have been observed in six holothuroid species from nine countries. All SKUDs present a similar symptom—the skin ulceration—and can be induced by bacteria, viruses, or abiotic factors. We here provide an update on SKUDs in holothuroids and analyse the case of the SKUD observed in Holothuria scabra in Madagascar. Field observations revealed a seasonality of the disease (i.e. wintertime maximum peak). Morphological analyses of integument ulcers showed that sea cucumbers react by forming a collagen fibre plug. Metagenomic analyses revealed a higher proportion of Vibrionaceae (Gammaproteobacteria) in ulcers in comparison to the healthy integument of the same individuals. Experimental infection assays were performed with ulcer crude extracts and bacteria isolated from these extracts (e.g. Vibrio parahaemolyticus) but did not significantly induce skin ulceration. Our results suggest that the disease is not induced by a pathogen or, at the very least, that the pathogen is not found within the ulcers as the disease is not transmissible by contact. An initial cause of the SKUD in Madagascar might be the repeated and prolonged exposures to cold temperatures. Opportunistic bacteria could settle in the dermis of ulcerated individuals and promote the ulcer extension. We propose a general nomenclature for SKUDs based on the acronym of the disease, the affected sea cucumber species (e.g. Hs for Holothuria scabra), the concerned region using an ISO code 3166-2 (e.g. MG for Madagascar), the description date (e.g. 20 for the year 2020), and, when known, the inducing agent (first letter of the general taxon, b for bacteria, v for virus in currently known cases; a a if it is an abiotic inducing parameter; nothing if the inducing cause has not been precisely identified). The disease described in this work will be designated under the name SKUD Hs-MG-20.
