To date, the association of an imbalance of the intestinal microbiota with various human diseases, including both diseases of the gastrointestinal tract and disorders of the immune system, has been shown. However, despite the huge amount of accumulated data, many key questions still remain unanswered. Given limited data on the composition of the gut microbiota in patients with ulcerative colitis (UC) and irritable bowel syndrome (IBS) from different parts of Siberia, as well as the lack of data on the gut microbiota of patients with bronchial asthma (BA), the aim of the study was to assess the biodiversity of the gut microbiota of patients with IBS, UC and BA in comparison with those of healthy volunteers (HV). In this study, a comparative assessment of the biodiversity and taxonomic structure of gut microbiome was conducted based on the sequencing of 16S rRNA genes obtained from fecal samples of patients with IBS, UC, BA and volunteers. Sequences of the Firmicutes and Bacteroidetes types dominated in all samples studied. The third most common in all samples were sequences of the Proteobacteria type, which contains pathogenic and opportunistic bacteria. Sequences of the Actinobacteria type were, on average, the fourth most common. The results showed the presence of dysbiosis in the samples from patients compared to the sample from HVs. The ratio of Firmicutes/Bacteroidetes was lower in the IBS and UC samples than in HV and higher the BA samples. In the samples from patients with intestinal diseases (IBS and UC), an increase in the proportion of sequences of the Bacteroidetes type and a decrease in the proportion of sequences of the Clostridia class, as well as the Ruminococcaceae, but not Erysipelotrichaceae family, were found. The IBS, UC, and BA samples had signif icantly more Proteobacteria sequences, including Methylobacterium, Sphingomonas, Parasutterella, Halomonas, Vibrio, as well as Escherichia spp. and Shigella spp. In the gut microbiota of adults with BA, a decrease in the proportion of Roseburia, Lachnospira, Veillonella sequences was detected, but the share of Faecalibacterium and Lactobacillus sequences was the same as in healthy individuals. A signif icant increase in the proportion of Halomonas and Vibrio sequences in the gut microbiota in patients with BA has been described for the f irst time.

The addition of silver ions or nanoparticles to impart antimicrobial properties to polymeric or other materials is a widely used method. However, it should be noted that the antiviral and antimicrobial effect of silver nanoparticles that come into contact with the environment, associated with their size, with a decrease in the size of nanoparticles, their effectiveness increases sharply. In the present work, we used a biodegradable polymer polylactide (PLA), which is obtained by condensation of lactic acid or ring-opening polymerization of lactide. These studies will further contribute to the development of new safe materials, in particular for food packaging, which is undoubtedly an urgent problem. The work aims is to obtain the silver-containing polymer composites based on polylactide by thermochemical reduction of Ag+ ions using natural (chitosan) and synthetic (polyethyleneimine (PEI)) polymers and to study the structure, morphology, thermomechanical and antimicrobial properties of the obtained nanocomposites. Thermochemical reduction of Ag+ ions in the bulk of polymer films, containing PLA, silver palmitate as a precursor of Ag NP and reducing agent (PEI or chitosan), has been performed by keeping them at 100–170 °C within 5 minutes. The polymeric film composites, cast from chloroform solution, were heated in an oven using precise thermal regulator VRT-3. As a result of reduction, the films get attained silver color, the Ag content in the bulk of films varied from 1 to 4 wt. %, and the thickness of the films was 110 μm. Structure, morphology, thermomechanical and antimicrobial properties of two types of nanocomposites – PLA-Ag-PEI and PLA-Ag-chitosan, formed by the thermochemical reduction of Ag+ in polymer films have been studied using wide-angle X-ray scattering (WAXS) (a DRON-4-07 diffractometer), a transmission electron microscope (TEM) (JEM-1230 JEOL, Japan), and thermomechanical analysis (a UIP-70M device). Antimicrobial activity of the obtained nanocomposites was investigated applying reference strains of opportunistic bacteria Staphylococcus aureus and Escherichia coli. It has been found that thermochemical reduction of Ag+ ions in the bulk of polymer films, when using synthetic or natural polymers (PEI or chitosan) as a reducing and stabilizing agent of silver nanoparticles occurs at 160 °C during 5 minutes. It has been found that the average size of Ag nanoparticles in the polymeric matrix is equal to ~ 7 and ~ 4 nm for PEI and chitosan, respectively. It has been shown that PLA-Ag-chitosan nanocomposites have much higher antimicrobial activity against S. aureus and E. coli strains as compared to PLA-Ag-PEI nanocomposites.

Research objective: to determine the characteristics of nutritional status (NS) in women of reproductive age with typical clinical signs of lichen sclerosus of the vulva (LSV) using key NS parameters. Materials and methods. The study included 75 women with average age 31.3 ± 1.3 years. 50 women were diagnosed with LSV and NS disorders (main group), and 25 women were practically healthy (control group). Information about all patients was collected through direct interviews, clinical examination and study of medical records. The presence of anogenital pruritus, soreness or burning, dryness, dyspareunia, urinary disturbances, perianal and/or intestinal symptoms, inflammatory bowel disease, thyroid dysfunction and tenderness, and other comorbidities were recorded. NS evaluated using anthropometric, laboratory and clinical studies. Body mass index, index of nutritional risk (nutritional deficiency), vitamins, micro- and macroelements value, scatological parameters were determined in all patients, and the intestinal microflora was examined.Results. The results of the study showed a disorder of NS in most patients with LSV by all studied parameters in comparison with the control group. Thus, it was found that 60% of patients with LSV had abnormal body mass index as well as NS was observed in 72% of patients. In addition, patients with LSV also had a higher deficiency of vitamins D and B12, and some important micro- and macronutrients. The results of the coprological study showed the neutral fats in feces, unchanged muscle fibers, extracellular and intracellular starch, decreased levels of bifidobacteria and lactobacilli in 68–74% of patients and opportunistic bacteria and fungi in 30–46% of women in the main group.Conclusions. Thus, the results of this study indicate the association between the abnormal NS and LSV. However, further research is needed to assess the course of the underlying disease and to analyze the metabolic pathways that lead to disorders of lipid, carbohydrate and protein metabolism in target patients, to select of the correct nutrients and alternative treatments.

The roles of host-associated bacteria have gained attention lately, and we now recognise that the microbiota is essential in processes such as digestion, development of the immune system and gut function. In this study, Atlantic cod larvae were reared under germ-free, gnotobiotic and conventional conditions. Water and fish microbiota were characterised by 16S rRNA gene analyses. The cod larvae’s transcriptional responses to the different microbial conditions were analysed by a custom Agilent 44 k oligo microarray. Gut development was assessed by transmission electron microscopy (TEM). Water and fish microbiota differed significantly in the conventional treatment and were dominated by different fast-growing bacteria. Our study indicates that components of the innate immune system of cod larvae are downregulated by the presence of non-pathogenic bacteria, and thus may be turned on by default in the early larval stages. We see indications of decreased nutrient uptake in the absence of bacteria. The bacteria also influence the gut morphology, reflected in shorter microvilli with higher density in the conventional larvae than in the germ-free larvae. The fact that the microbiota alters innate immune responses and gut morphology demonstrates its important role in marine larval development.

Most bacteria found in ticks are not pathogenic to humans but coexist as endosymbionts and may have effects on tick fitness and pathogen transmission. In this study, we cultured and isolated 78 bacteria from 954 Ixodes ricinus ticks collected in 7 sites of a Belgian peri-urban forest. Most isolated species were non-pathogenic environmental microorganisms, and were from the Firmicutes (69.23%), Actinobacteria (17.95%) and Proteobacteria (3.84%) phyla. One bacterium isolate was particularly noteworthy, Cedecea davisae, a rare opportunistic bacterium, naturally resistant to various antibiotics. It has never been isolated from ticks before and this isolated strain was resistant to ampicillin, cefoxitin and colistin. Although cultivable bacteria do not represent the complete tick microbiota, the sites presented variable bacterial compositions and diversities. This study is a first attempt to describe the culturable microbiota of ticks collected in Belgium. Further collections and analyses of ticks of different species, from various areas and using other bacterial identification methods would strengthen these results. However, they highlight the importance of ticks as potential sentinel for opportunistic bacteria of public health importance.

Fish are widely exposed to higher microbial loads compared to land and air animals. It is known that the microbiome plays an essential role in the health and development of the host. The oral microbiome is vital in females of different organisms, including the maternal mouthbrooding species such as Nile tilapia (Oreochromis niloticus). The present study reports for the first time the microbial composition in the buccal cavity of female and male Nile tilapia reared in a recirculating aquaculture system. Mucus samples were collected from the buccal cavity of 58 adult fish (∼1 kg), and 16S rRNA gene amplicon sequencing was used to profile the microbial communities in females and males. The analysis revealed that opportunistic pathogens such as Streptococcus sp. were less abundant in the female buccal cavity. The power play of certain bacteria such as Acinetobacter, Acidobacteria (GP4 and GP6), and Saccharibacteria that have known metabolic advantages was evident in females compared to males. Association networks inferred from relative abundances showed few microbe–microbe interactions of opportunistic pathogens in female fish. The findings of opportunistic bacteria and their interactions with other microbes will be valuable for improving Nile tilapia rearing practices. The presence of bacteria with specific functions in the buccal cavity of female fish points to their ability to create a protective microbial ecosystem for the offspring.

Background: Loss of pregnancy in mares can have many different causes, including both infectious and non-infectious conditions. Extrapolation of findings from other studies is often uncertain as the significance of each cause varies across regions. Causes of pregnancy loss in mares have never been thoroughly studied in Denmark, so a prospective cross-sectional cohort study targeting the entire Danish population of pregnant mares was performed over a period of 13 months to obtain knowledge of the significance of individual causes. Fifty aborted or prematurely delivered stillborn fetuses were submitted for necropsy and examined by a panel of diagnostic laboratory methods.Results: Overall, a cause of fetal loss was established for 72% of the examined cases. Most cases (62%) were lost due to a non-infectious cause, of which obstruction of the feto-placental blood circulation due to severe torsion of the umbilical cord was most prevalent. Pregnancy loss due to a variety of opportunistic bacteria, including bacteria not previously associated with abortion in mares, accounted for 12%, while equid alphaherpesvirus (EHV) type 1 was the cause of pregnancy loss in 8% of the cases. EHV type 4 and Chlamydiaceae species were identified in some cases, but not regarded as the cause of fetal loss.Conclusion: Umbilical cord torsion was found to be the most prevalent cause of fetal loss in Danish mares, while infectious causes such as EHV type 1 and streptococci only accounted for a minor proportion of the losses. The study highlights the need for defined criteria for establishing an abortion diagnosis in mares, particularly in relation to EHV types 1 and 4.

Relevance. The activity of the digestive organs is largely determined by the feeding regime, in particular, the frequency, timeliness, and the order of feeding the feed. The digestibility of nutrients is influenced by the preparation of feed for feeding, which facilitates mechanical processing in the digestive tract and the action of digestive enzymes. Against the background of a decrease in the natural resistance of the animal body, an important aspect is the use of mineral supplements and the enrichment of diets with biologically active substances that increase the nutritional value and the degree of assimilation of feed by animals. The aim of the work was a comparative assessment of methods for studying the degree of assimilation of feed by animals.Methods. To determine the degree of food assimilation, 2 drops of isotonic (0.9%) sodium chloride solution and a small amount of intestinal contents were applied to a glass slide, a thin smear was prepared, dried in air for 30 min, and large particles were removed. The preparations were fixed for 15 min with 96% ethyl alcohol, then 3–5 drops of a 1.0% aqueous solution of methylene blue, Lugol, Sudan III were applied. Along with the indicated conventional methods, we tested a method for preparing preparations using fixation in vapors of a 25.0% solution of glutaraldehyde for 30–40 minutes, in vapors of a 1.0% aqueous solution of osmium tetroxide.Results. The kinetics of the formation of intestinal microbiocenoses is characterized by a decrease in the number of lactobacilli and an increase in the number of pathogenic and opportunistic bacteria. In a comparative assessment of the preparation methods, the advantage of microscopic research methods is the possibility of a detailed study of the contents, the degree of assimilation of feed components in the gastrointestinal tract, and a visual assessment of the state of the intestinal microflora. The advantage of the method for preparing preparations with the use of vapor fixation of a 25.0% solution of glutaraldehyde and osmium tetroxide is the possibility of visualizing the formation of monospecific and polyspecies biofilms by optical and scanning electron microscopy. The preparations were sufficiently contrasting, there were no artifacts, bacteria and fungi stained brown, which made it possible to count even pinpoint colonies using a magnifying glass or an optical microscope. The easily digestible feed results in a low fecal yield and a stable consistency. The use of functional food “Pristine Gold Vet” in the nutrition of dogs and fur-bearing animals made it possible to increase the absorption of nutrients by 18,1% and 15,3%.

The One Health approach emphasizes the importance of antimicrobial resistance (AMR) as a major concern both in public health and in food animal production systems. As a general classification, E. coli can be distinguished based on the ability to cause infection of the gastrointestinal system (IPEC) or outside of it (ExPEC). Among the different pathogens, E. coli are becoming of great importance, and it has been suggested that ExPEC may harbor resistance genes that may be transferred to pathogenic or opportunistic bacteria. ExPEC strains are versatile bacteria that can cause urinary tract, bloodstream, prostate, and other infections at non-intestinal sites. In this context of rapidly increasing multidrug-resistance worldwide and a diminishingly effective antimicrobial arsenal to tackle resistant strains. ExPEC infections are now a serious public health threat worldwide. However, the clinical and economic impact of these infections and their optimal management are challenging, and consequently, there is an increasing awareness of the importance of ExPECs amongst healthcare professionals and the general public alike. This review aims to describe pathotype characteristics of ExPEC to increase our knowledge of these bacteria and, consequently, to increase our chances to control them and reduce the risk for AMR, following a One Health approach.
