Saponins are plant and marine animal specific metabolites that are commonly considered as molecular vectors for chemical defenses against unicellular and pluricellular organisms. Their toxicity is attributed to their membranolytic properties. Modifying the molecular structures of saponins by quantitative and selective chemical reactions is increasingly considered to tune the biological properties of these molecules (i) to prepare congeners with specific activities for biomedical applications and (ii) to afford experimental data related to their structure–activity relationship. In the present study, we focused on the sulfated saponins contained in the viscera of Holothuria scabra, a sea cucumber present in the Indian Ocean and abundantly consumed on the Asian food market. Using mass spectrometry, we first qualitatively and quantitatively assessed the saponin content within the viscera of H. scabra. We detected 26 sulfated saponins presenting 5 different elemental compositions. Microwave activation under alkaline conditions in aqueous solutions was developed and optimized to quantitatively and specifically induce the desulfation of the natural saponins, by a specific loss of H2SO4. By comparing the hemolytic activities of the natural and desulfated extracts, we clearly identified the sulfate function as highly responsible for the saponin toxicity.

ABSTRACTThe purpose of this study was to determine the effect of the addition of sea cucumber flour on the organoleptic and proximate characteristics of wet noodles. This study consisted of four treatments, namely without the addition of sand sea cucumber flour (T0), the addition of 5% sea cucumber flour (T1), the addition of 10% sea cucumber flour (T2), and the addition of 15% sea cucumber flour (T3). The analyzed parameters were organoleptic and physicochemical values. This study used a completely randomized design (CRD). Data were analyzed using Analysis of Variance (ANOVA) and continued with Duncan's Multiple Range Test (DMRT) at a 95% confidence level. The results show that the effect of adding sand sea cucumber flour had a very significant effect on organoleptic assessment in T1 treatment (95% wheat flour: 5% sea cucumber flour) with preference scores of color, aroma, taste, and texture reached 4.03 (like), 4.06 (like), 3.91 (like), and 4.06 (like). Proximate analysis of the best treatment shows that it contained 50.12% water, 1.18% ash, 0.20% fat, 7.23% protein, and 41.75% carbohydrate. The results show that the product met the standard of SNI 01-2987-1992 for ash, fat, and protein contents. Penelitian ini terdiri dari empat perlakuan yaitu tanpa penambahan tepung teripang pasir (T0), penambahan tepung teripang pasir 5% (T1), penambahan tepung teripang pasir 10% (T2), dan penambahan tepung teripang pasir 15% (T3), lalu dianalisis terhadap nilai organoleptik dan fisikokimia. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL). Data di analisis menggunakan Analysis of Varian (ANOVA) dan dilanjutkan dengan uji Duncan’s Multiple Range Test (DMRT) pada taraf kepercayaan 95%. Hasil penelitian menunjukkan bahwa pengaruh penambahan tepung teripang pasir berpengaruh sangat nyata terhadap penilaian organoleptik pada perlakuan T1 (tepung terigu 95% : tepung teripang pasir 5%) sebesar warna 4.03 (suka), aroma 4.06 (suka), rasa 3.91 (suka), dan tekstur 4.06 (suka). Analisis proksimat perlakuan terbaik diperoleh nilai kadar air 50.12%, kadar abu 1.18%, kadar lemak 0.20%, kadar protein 7.23% dan kadar karbohidrat 41.75%. Berdasarkan hasil penelitian nilai kadar abu, kadar lemak, dan kadar protein telah memenuhi SNI 01-2987-1992.Kata kunci: Teripang Pasir, Mie Basah

<p class="Papertext"><strong>The Preference of The Sandfish Holothuria scabra on Macroalgae-Based Feed. </strong>The sandfish, <em>Holothuria scabra</em>, is one ofthe marine species that has the potential to be developed as a source of high-value functional food. However, the growth of sandfish is still constrained by the availability of suitable feed. This study aimed to determine the level of preference of sea cucumbers for artificial feed made from macroalgae. This study used six main ingredients: control (sea-sand),  FF-1 (commercial shrimp feed), FF-2 (<em>Padina</em> sp.), FF-3 (<em>Ulva</em> sp.), FF-4 (<em>Sargassum</em> sp.), FF-5 (combination). A completely randomized design with three replications was implemented. Nutritional values of feed and the sandfish body, feed consumption, specific growth rate (SGR), survival, and water quality were investigated. The results revealed that all treatments had a 100 percent survival rate, even though the SGR for all treatmentswere negative. Artificial feed made from <em>Padina</em> sp. (FF-2) had the highest average feed consumption of 0.51 gram/day. This value was significantly greater than the other treatments (p&lt;0.05). The SGR reached a positive value in the last week of the maintenance phase. The nutritional quality of the sandfish body generally decreased compared to the initial rearing condition. On the other hand, water quality was optimal for sandfish growth throughout the experiment.It can be concluded that the feed derived from <em>Padina</em> sp. is preferable for <em>Holothuria scabra </em>and has the potential to be further developed.</p>

Echinoderms are one of organisms that inhabit intertidal areas which have important ecological roles in marine ecosystems, also have high economic values. The community structure and distribution pattern of Echinoderms in the intertidal area can provide an overview of the condition of the Echinoderm community. This research aims to determine the community structure of Echinoderms in various habitat types in the intertidal areas of Lakeba Beach, Baubau Town. The community structure of Echinoderms were observed using quadratic transec method. Echinoderms found were 777 individuals consisting of 18 species from 5 classes. Echinoderms were distributed in four habitat types, i. e. sandy, sand covered with seagrass, rocky, and rock covered with algae habitats. The highest abundance of Echinoderm was Ophiocoma scolopendrina (Ophiuroidea) and the lowest were Maretia planulata, Arachnoides placenta (Echinoidea), Holothuria scabra, H. leucospilota, Stichopus horrens (Holothuroidea), and Isocrinidae (Crinoidea). The species diversity of Echinoderms was categorized as moderate (H’ = 1,42180), species evenness was moderate (E = 0,49191), and no dominant species of Echinoderm community (D = 0,36601). Echinoderm community living in the intertidal area was influenced by antropogenic factors.  Echinodermata merupakan salah satu organisme yang mendiami daerah intertidal yang memiliki peran ekologis penting bagi ekosistem laut, juga memiliki nilai ekonomis yang tinggi. Struktur komunitas dan pola distribusi Echinodermata pada daerah intertidal dapat memberikan gambaran mengenai kondisi komunitas Echinodermata. Penelitian ini bertujuan untuk mengetahui struktur komunitas dan pola distribusi Echinodermata di daerah intertidal Pantai Lakeba, Kota Baubau. Struktur komunitas dan pola distribusi Echinodermata diamati menggunakan metode transek kuadrat. Echinodermata yang ditemukan berjumlah 777 individu yang terdiri atas 18 jenis dari 5 kelas. Echinodermata terdistribusi pada empat tipe habitat yaitu habitat pasir, pasir yang ditutupi lamun, batu, dan batu yang ditutupi alga. Echinodermata yang memiliki kelimpahan tertinggi adalah Ophiocoma scolopendrina (Ophiuroidea) dan kelimpahan terendah adalah Maretia planulata, Arachnoides placenta (Echinoidea), Holothuria scabra, H. leucospilota, Stichopus horrens (Holothuroidea), dan Isocrinidae (Crinoidea). Keanekaragaman jenis Echinodermata dikategorikan sedang (H’ = 1,42180), kemerataan jenis sedang (E = 0,49191) dan tidak ada jenis yang dominan dalam komunitas Echinodermata (D = 0,36601). Komunitas Echinodermata yang hidup di daerah intertidal pantai dipengaruhi oleh faktor antropogenik.
