Characterization of Cryoglobulins by Immunoblotting

Abstract We adapted an immunoblotting technique for the immunochemical characterization of cryoglobulins. We first compared the results from 157 samples with results obtained by immunofixation and immunoelectrophoresis and then with clinical observations in 125 cases. Full identification was possible in 98% of the cases by immunoblotting, in 54% by immunofixation, and in 28% by immunoelectrophoresis. Novel microheterogeneity aspects were observed by immunoblotting and immunofixation in 13% and 6% of the cases studied, respectively. Our results confirm the frequency of autoimmune and infectious diseases associated with mixed cryoglobulinemias, whereas a lymphoproliferative syndrome was observed only in cases of cryoglobulinemia with a monoclonal constituent or a microheterogeneity aspect, which was not always revealed by immunofixation or immunoelectrophoresis. The pathophysiological importance and classification of microheterogeneity aspects is not clear. These observations justify using a sensitive and specific method for identifying cryoglobulins, even when present at low concentrations, and call for long-term studies of these patients.

Long-term studies of the North Caucasian geochemical province allowed to establish regional abundances and calculate accumulation (dispersion) factors for chemical elements in rocks, soils, and plants. Certain natural regional patterns characterize the province. Associations of elements in high and low concentrations are often determined by the predominant composition of rocks: carbonate-terrigenous, terrigenous, and igneous. The study of the average contents of several chemical elements in the soils of the province showed that the association of accumulated elements includes metals with different migration characteristics. Thus, despite the rather close values of the ionic radii, Pb, Zn, Cu, and Li (judging by the ionic potential) are characterized by the formation of cations, while Mn, Mo, and Zr form complex ions. Such elements as Zn, Cu, and Pb are mainly accumulated on hydrosulfuric barriers, while Mo, Co, and Mn are stopped by oxygenous barriers. For Cu, Zn, Mo, and Co, biogenic accumulation plays a significant role, while for Pb and Ni it is practically absent. The absolute dispersion of the elements did not reach environmentally hazardous values, although it indicates a fairly intensive migration. In woody plants, Ba, Nb, Sc, Sr, and Zn are accumulated most intensively.

The observed sedimentation data from some Iranian large reservoirs were used to characterize sediment distribution patterns within these reservoirs and a modified approach was developed. This new idea is of an empirical nature and is based on some parameters obtained from the minimum stream power principle. The main advantage of this new characterization is that it eliminates the need for classification of the dam based on any extra data and solely depends on the characteristics of the reservoir geometry. It also rectifies some practical difficulties for application of the existing method. The developed technique achieved by this study is recommended for preliminary design stages to interpret long-term cumulative distribution of sediment and is applicable for reservoirs that have sediment particles that are mainly of fine-grained material.

The results of veterinary meat inspection classification of 4 000 372 bovine carcasses reflect long-term aspects of health status in cattle herds and the quality of transport and handling of animals at slaughterhouses. Veterinary inspectors recorded the data obtained from meat inspection classification of bovine carcasses at slaughterhouses in the CzechRepublicduring the period of 1995–2002 together with the reasons for classification. The trends were evaluated by a comparison of two periods (Period I, 1995–1998, and Period II, 1999–2002) by means of calculating the indexes of values from Period II compared to those of Period I. Bovine carcasses classified as capable for human consumption (edible) were found in 87.87% of cases (88.83% during Period I and 86.58% during Period II, index 0.97), while those classified as capable for processing (conditionally edible) were found in 7.53% of cases (7.38% during Period I and 7.71% during Period II, index 1.04), and those condemned in 4.60% of cases (3.79% during Period I and 5.71% during Period II, index 1.51). The most important reason for classifying the carcasses as condemned was the finding of sensorial changes in meat, which occurred in 2.56% of cases (2.23% during Period I and 3.00% during Period II, index 1.35), followed by lesions due to non-infectious diseases – 1.00% (0.81% during Period I and 1.25% during Period II, index 1.53), added deleterious substances – 0.88% (0.60% during Period I and 1.27% during Period II, index 2.11), lesions due to respiratory infections – 0.03% (0.02% during Period I and 0.04% during Period II, index 1.74), and lesions due to miscellaneous infectious diseases – 0.10% (0.10% during Period I and 0.10% during Period II, index 1.05). Other reasons to condemn the carcasses included improper identification, lesions due to digestive infections, lesions due to tuberculosis, lesions due to paratuberculosis, lesions due to salmonellosis, leucosis and parasitic diseases. The occurrence of these conditions was on the level of mere hundredths of per cent. According to the results of meat inspection classification, the risk of food-borne diseases originating from bovine carcasses tends to be greater in the lesions due to non-infectious conditions with a long-term increasing trend. A considerable increase in the numbers of bovine carcasses condemned because of lesions due to paratuberculosis (index 4.62) represents an alarming finding with regard to potential food safety hazards.

Physical or chemical procedures could efficiently remove contaminants including pesticides such as carbamates from high concentrations of toxicants. Bioremediation, on the other hand, is frequently a less expensive option in the long term when used at low concentrations. Isolation of multiple toxicants removing microorganisms is the goal of bioremediation. In this paper we report on the molybdenum reduction of the bacterium and its ability to grow on the carbamates carbofuran and carbaryl as carbon sources. Both the carbamates carbofuran and carbaryl cannot support molybdenum reduction when used as the sole carbon sources. Between pH 6.0 and 6.8 and between 30 and 34 oC, the bacterium is most efficient in converting molybdate to Mo-blue. For molybdate reduction, glucose was shown to be the strongest electron donor, with maltose and sucrose coming in second and third, respectively, and d-mannitol and d-adonitol coming in last. Phosphate concentrations of 2.5 to 7.5 mM and molybdate concentrations of 20 to 30 mM are also needed. Identical to that of a decreased phosphomolybdate, the Mo-blue produced by the new Mo-reducing bacteria has an absorption spectrum similar to prior Mo-reducing bacteria. Inhibition of molybdenum reduction was 73.3, 50.1, 50.1 and 20.7 percent, respectively, by mercury, copper, silver and lead at 2 ppm. The bacterium was tentatively identified as Serratia sp. strain Amr-4 after biochemical investigation. This bacterium's ability to detoxify a variety of toxicants is highly sought after, making it a significant bioremediation agent.

Long-term ground-based measurements of aerosol optical properties in Athens, Greece, for the period 2008–2018 performed by the National Observatory of Athens are used in order to investigate the aerosol climatology of the area. In this study, we utilize quality-assured measurements of the aerosol optical depth (AOD), Single Scattering Albedo (SSA) and Ångström exponent obtained by CIMEL photometers in the framework of the Aerosol Robotic Network (AERONET) to extract the seasonality and the trends of aerosols in the region. Higher aerosol loads are found during spring and summer months. A 1.1% per year decrease for AOD at 440 nm and 0.4% decrease per year for SSA during the studied period are recorded. Collocated and synchronous PM10 values, for a five-year period, are used in order to study ground-level conditions. Also, the Planetary Boundary Layer Height from ERA-5 is used to investigate the stratification of the particles. The classification of aerosols using AERONET data is performed to separate dust, biomass burning, polluted urban, marine and continental dominant aerosol mixtures. Also, the characterization of AOD provided by Copernicus Atmosphere Monitoring Service (CAMS) is investigated. Finally, seasonal AOD trends recorded from AERONET from satellite sensors (MODIS-Aqua/MODIS-Terra) and estimated by CAMS are examined, and significant differences have been found.

The classification of medically unexplained symptoms (MUS) could have negative consequences for patients with functional somatic syndromes (FSS). By grouping related but distinct syndromes into one label, the MUS classification fails to inform clinicians about their patients’ health condition. In research settings, the MUS classification makes patient samples more heterogeneous, obstructing research into the underlying pathology of FSS. Long-term studies have shown that MUS are often appraised as medically explained symptoms at follow-up and vice versa, raising doubts about the reliability of this distinction.

AbstractEnergy restriction (ER; also known as caloric restriction) is the only nutritional intervention that has repeatedly been shown to increase lifespan in model organisms and may delay ageing in humans. In the present review we discuss current scientific literature on ER and its molecular, metabolic and hormonal effects. Moreover, criteria for the classification of substances that might induce positive ER-like changes without having to reduce energy intake are summarised. Additionally, the putative ER mimetics (ERM) 2-deoxy-d-glucose, metformin, rapamycin, resveratrol, spermidine and lipoic acid and their suggested molecular targets are discussed. While there are reports on these ERM candidates that describe lifespan extension in model organisms, data on longevity-inducing effects in higher organisms such as mice remain controversial or are missing. Furthermore, some of these candidates produce detrimental side effects such as immunosuppression or lactic acidosis, or have not been tested for safety in long-term studies. Up to now, there are no known ERM that could be recommended without limitations for use in humans.

SUMMARYWe investigated the genetic structure of 99 isolates of serotype 23FStreptococcus pneumoniaefrom children with acute respiratory infections collected over two periods from 1997 to 2006, and 2010. All isolates were susceptible to vancomycin and amoxicillin–clavulanic acid; 97 were resistant to erythromycin, 95 of which carried theermBgene and two carried bothmefA/EandermBgenes. Multidrug resistance to three or more classes of antibiotics was exhibited by 90 isolates. Sequence types ST342 and ST81 were the most frequent in 1997–2006 and 2010, respectively. All CC81 isolates were non-susceptible to β-lactam antibiotics and had higher minimum inhibitory concentration values for penicillin than other clone complexes and sequence types. The increased β-lactam antibiotic resistance may have resulted from the replacement of multidrug-resistant clones related to ST81. Long-term studies onS. pneumoniaeserotype 23F, especially the ST81 clone, should be conducted to better understand the epidemiological picture of this pathogen in China.
