Abstract A method was developed for simultaneous determination of oxytetracycline (OTC), chlortetracycline, tetracycline, and doxycycline levels in cow milk, serum, and urine by capillary electrophoresis (CE). The tetracyclines (TCs) were extracted specifically with a metal-chelating affinity column. Extracts were then applied to C18 cartridges treated with dimethyldichlorosilane, and the TCs were eluted with ethanol. Salt-free sample residues were run on CE with a diode array detector at 370 nm. A 50 cm × 75 μm uncoated capillary at 15 kV and 23°C was used to separate the TCs. The run buffer contained 10 mM sodium dodecyl sulfate, 50 mM borate, and 50 mM phosphate, pH 8.5. TC peaks were identified by migration times, coinjection of standards, and diode array spectra. Limits of detection for TCs were 1.3-5.3 ng/mL, and limits of quantitation were 1.7-8.7 ng/mL in cow fluid samples. OTC appeared consistently at 13.3-13.4 min in milk, serum, and urine samples from a cow treated with OTC. OTC was further identified by coinjection of OTC standard and OTC diode array spectra. The CE method can be used to simultaneously determine 4 TC residues at low parts-per-billion levels.

Introduction. Recently, there has been a growing trend in the number of obese and overweight patients. To date, sibutramine is the most effective drug for treating obesity and overweight. The drug is an inhibitor of the reuptake of serotonin and norepinephrine, which leads to a decrease in hunger, and therefore, to weight loss.Aim. To develop and validate a methodology for the determination of sibutramine in drugs by capillary electrophoresis (CE) using an ultraviolet diode array detector.Materials and methods. Quantitative determination of sibutramine in drugs was carried out using the CE method with an ultraviolet diode array detector. A solution of phosphate buffer 50 mmol pH = 7.0 was used as a solvent and working electrolyte; to separate the peaks – quartz capillary 56 cm, 50 μm.Results and discussion. The developed method was validated according to the following parameters: specificity, linearity, correctness, precision, limit of detection and limit of quantification.Conclusion. A method for the quantitative determination of sibutramine in drugs by the CE method using an ultraviolet diode array detector has been developed and validated. This method meets all the requirements of General Pharmacopoeia Monograph 1.1.0012.15 «Validation of the analytical method» and can be used to control the quality of drugs, the active pharmaceutical substance of which is sibutramine.

This study presents the validation of a high-performance liquid chromatography diode array detector (HPLC-DAD) method for the determination of different mycosporine-like amino acids (MAAs) in the red alga Bostrychia scorpioides. The investigated MAAs, named bostrychines, have only been found in this specific species so far. The developed HPLC-DAD method was successfully applied for the quantification of the major MAAs in Bostrychia scorpioides extracts, collected from four different countries in Europe showing only minor differences between the investigated samples. In the past, several Bostrychia spp. have been reported to include cryptic species, and in some cases such as B. calliptera, B. simpliciuscula, and B. moritziana, the polyphyly was supported by differences in their MAA composition. The uniformity in the MAA composition of the investigated B. scorpioides samples is in agreement with the reported monophyly of this Bostrychia sp.
