Recent results suggest that people hold a notion of the true self, distinct from the self. Here, we seek to further elucidate the “true me”—whether it is good or bad, material or immaterial. Critically, we ask whether the true self is unitary. To address these questions, we invited participants to reason about John—a character who simultaneously exhibits both positive and negative moral behaviors. John’s character was gauged via two tests--a brain scan and a behavioral test, whose results invariably diverged (i.e., one test indicated that John’s moral core is positive and another negative). Participants assessed John’s true self along two questions: (a) Did John commit his acts (positive and negative) freely? and (b) What is John’s essence really? Responses to the two questions diverged. When asked to evaluate John’s moral core explicitly (by reasoning about his free will), people invariably descried John’s true self as good. But when John’s moral core was assessed implicitly (by considering his essence), people sided with the outcomes of the brain test. These results demonstrate that people hold conflicting notions of the true self. We formally support this proposal by presenting a grammar of the true self, couched within Optimality Theory. We show that the constraint ranking necessary to capture explicit and implicit view of the true self are distinct. Our intuitive belief in a true unitary “me” is thus illusory.

Abstract The aim of this study was to evaluate the possible difference in seminal characteristics of Dorper rams of either high, medium or low social ranks in northern Mexico (26º N). The male’s social status was quantified through a competitive behavioral test in a herd of 36 adult males, the aggressive interactions and their consequences were registered to calculate the success index (SI), where low hierarchy (LH) rams had a SI of 0 to 0.33, medium hierarchy (MH) rams had a SI of 0.34 to 0.66 and high hierarchy (HH) had a SI of 0.67 to 1. Afterwards, the rams were subjected to an estrogenized female (2 mg of estradiol cypionate) for teasing in order to collect a semen sample with an artificial vagina. The semen was evaluated for volume, motility and concentration. The seminal characteristics differences were analyzed using a one-way ANOVA tests and stablishing a p-value of 0.10 as significance level in the SPSS statistical package. Results are shown in table 1. In conclusion, low hierarchy Dorper rams show a statistical tendency to have a lower sperm concentration.

IntroductionAlterations on the innate inflammatory response may underlie the pathophysiology of psychiatric diseases, but the mechanisms implicated remain elusive. Current antipsychotics modulate pro/anti-inflammatory pathways, but the specific mechanisms involved remain elusive. One attractive possibility is the regulation of the intracellular signalling pathways of the innate immune receptors Toll-like 3 (TLR3), which triggers antiviral and inflammatory responses.AimsTo elucidate the regulatory role of paliperidone on maternal immune activation (MIA) induced alterations on TLR3 pathway and on the two emerging endogenous antiinflammatory/antioxidant mechanisms NRF2/antioxidant enzymes pathway and the cytokine milieu regulating M1/M2 polarization in microglia.MethodsPregnant mice were treated with the synthetic Toll-like Receptor 3 (TLR3) agonist Poly(I:C) in gestational day 9 and chronically treated with paliperidone (0,05 mg/kg i.p.) in adult offspring. Animals were sacrificed one day after treatment and behavioral test. Inflammation oxidative stress-related mediators were analysed at mRNA and protein level in prefrontal cortex samples. In addition, behavioral test t-maze was conducted.ResultsPaliperidone prevented TLR3 pathway activation and the subsequent MIA-induced neuroinflammatory response. Also, paliperidone induced an increment in the activity and protein expression of nuclear NRF2, as well as increased mRNA levels of the antioxidant enzymes HO1, SOD and catalase in the MIA model. Otherwise, paliperidone increases the antiinflammatory cytokines levels TGFβ and IL-10 in favour of a M2 microglia profile and increased the levels of the M2 cellular markers ArgI and FOLR2.ConclusionsThe modulation of neuroinflammation and enhancement of endogenous antioxidant/anti-inflammatory pathways by current and new antipsychotics could represent an interesting therapeutic strategy for the future.Disclosure of interestThe authors have not supplied their declaration of competing interest.

AbstractMemory consolidation requires astrocytic microdomains for protein recycling; but whether this lays a mechanistic foundation for long-term information storage remains enigmatic. Here we demonstrate that persistent synaptic strengthening invited astrocytic microdomains to convert initially internalized (pro)-brain-derived neurotrophic factor (proBDNF) into active prodomain (BDNFpro) and mature BDNF (mBDNF) for synaptic re-use. While mBDNF activates TrkB, we uncovered a previously unsuspected function for the cleaved BDNFpro, which increases TrkB/SorCS2 receptor complex at post-synaptic sites. Astrocytic BDNFpro release reinforced TrkB phosphorylation to sustain long-term synaptic potentiation and to retain memory in the novel object recognition behavioral test. Thus, the switch from one inactive state to a multi-functional one of the proBDNF provides post-synaptic changes that survive the initial activation. This molecular asset confines local information storage in astrocytic microdomains to selectively support memory circuits.
