This study aimed to determine the effectiveness and the best concentration of Moringa leaf extract (MLE) in the citrate-egg yolk (C-EY) to maintain the motility and viability of spermatozoa kacang goat. Semen was collected from 3 goats aged two years; by using the artificial vagina method. The semen was evaluated macroscopically and microscopically. The semen that had >70% sperm motility and >250x106/ml sperm concentration was divided into 4 equal tubes, each diluted with100% C-EY (P1), 10% MLE+ 90%C-EY (P2), 20% MLE +80% C-EY (P3), and 30% MLE+70% C-EY (P4). The diluted samples were then stored in a refrigerator (3-5?C) and evaluated for motility and viability every 24 hours. The study was designed using a completely randomized design (CRD) consisting of four treatments and five replications. The results showed that the addition of MLE in C-EY significantly affected goat spermatozoa's progressive motility and viability. The data showed that the spermatozoa kept during four days in a diluent of P2 had higher (P<0.05) motility 44.67±4.80% and viability 74.24±4.46%than the other three diluents of P1(36.00±4.70%; 70.10±3.6%), P3(33.67±0.42%; 66.85±4.99%) and P4 (29.67±3.99%; 63.96±5.44%). This study concluded that adding 10% MLE was the best concentration as source energy in 90% C-EY diluents, which effectively maintained the motility and viability of kacang goat spermatozoa for four days of storage at a temperature of 3-5oC.

Pharmacologically induced ex copula ejaculation is a method used for collection of semen when the traditional methods of semen collection are not feasible. Common indications for this method include health issues that either preclude the physical act of mating or result in impaired erection and ejaculation. The method also offers an alternative when there is a lack of equipment and facilities required for semen collection using the conventional artificial vagina method. A variety of pharmacological protocols for ex copula ejaculation in stallions have been evaluated in both research and clinical settings with no serious side-effects reported. In general, these protocols included tricyclic antidepressants, alpha-2 adrenergic receptor agonists, and smooth muscle stimulators, either individually or in combination. Although there is a lot of variation in the ejaculatory rates among studies, a combination of imipramine and xylazine appears to be an effective option for inducing ejaculation in stallions. In cases where this protocol is not successful, collections should be reattempted using a combination of imipramine, detomidine, and oxytocin. Regardless of the protocol used, a quiet environment with minimal disturbance is associated with a better outcome. In contrast to the body of literature available on pharmacologically induced ex copula ejaculation in horses, only a few studies have been published so far on this topic in donkeys. Further studies are warranted to investigate whether pharmacologically induced ex copula ejaculation is an effective method of semen collection in jacks and to compare different pharmacological protocols for inducing ejaculation in jacks.

This research was conducted at the Teaching Farm of the Faculty of Veterinary Medicine, Airlangga University, to know the effect of age, temperature, and humidity on the quality of semen of bulls which are turned into frozen semen. This research uses fresh semen samples of bulls of various ages grouped into 3 groups, young, middle age, and old groups and measures temperature and humidity during the storage process. Fresh semen is collected using an artificial vagina and then examined macroscopic and microscopic tests which is then checked for post thawing motility. Data analysis uses Analysis of Variants (ANOVA) followed by Duncan. The results showed that the percentage of spermatozoa motility of fresh semen and post thawing differed and age affected the quality of semen produced. There was no effect of temperature and humidity when processing of collecting semen. Bull’s semen at Teaching Farm Faculty of Veterinary Medicine Airlangga University has good quality and is suitable for Artificial Insemination.

Research on the evaluation of the spermatozoa quality of Brahma, Cochin and Bangkok chickens as the basis for the application of artificial insemination technology in native chickens has been carried out. The purpose of this study was to evaluate the spermatozoa quality of Brahma, Cochin and Bangkok rooster as the basis for determining the spermatozoa dose for artificial insemination in local chickens. method research was used a completely randomized design (CRD) with three treatments, that is: Brahma, Cochin and Bangkok rooster. Each treatment was repeated five times. Cement storage is carried out by inserting an artificial vagina into the cloaca of the rooster and stimulating it to climb the hen. The collected sperm were analyzed at the Animal Reproduction Laboratory, Faculty of Animal Science, Mataram University. The variables observed included motility, viability and morphology of spermatozoa. The results of studied that the spermatozoa motility of Brahma, Chicin and Bangkok rooster were 72 ± 2.74%. 74 ± 2.24% and 73 ± 2.74% (P> 0.05) respectively; The spermatozoan viability of Brahma rooster and Bangkok rooster was 99.2 ± 1.30%, 99.4 ± 0.55% and 99.2 ± 1.10% (P>0.05)respectively. The spermatozoan morphology of brahma rooster, chicin rooster and Bangkok rooster was 94.6 ± 2.30%, 94.4 ± 2.70% and 97.4 ± 2.07% (P> 0.05), respectively. Cochin rooster and Bangkok rooster respectively, 62.6 ± 7.92 x 107, 57 ± 5.83 x107 , and 65.2 ± 12.28 x107, respectively. The results of the study concluded that Brahma, Cochin and Bangkok rooster had good sperm quality and could be able to used in artificial insemination of Kampung hens.

Abstract This study aimed to determine the effects of different semen diluent on the quality and storage time of liquid semen of Donggala bull. Semen was obtained from four selected bulls which collected using artificial vagina. The semen diluent is based on Tris aminomethane-citric acid with different concentration of glucose namely P1=0.00g, P2=0.25g, P3=0.50g, and P4=0.75g. Another group of treatment also prepared based on Tris aminomethane-glucose with different concentration of egg yolk namely P5=0%, P6=15%, P7=20%, and P8=25%. The data obtained were then analyzed descriptively. Results showed tris aminomethane-citric acid-glucose diluents had sperm progressive motile at >40% until the day 4 of storage, and tris aminomethane-glucose-egg yolk only able to support sperm life for 2 days, and in P8 group shows 41.02% motile on the day 3. In this study, we found the pattern of the increasing of shelf life affect to the decrease rate of sperm viability and normality.

The current study was aimed to evaluate the antioxidative effect of L-Carnitine at post thawing following cryopreservation of Nili-Ravi buffalo sperm. For this purpose, semen from three buffalo bulls were collected for 3 weeks using an artificial vagina (N=18; replicates). The qualified ejaculates were diluted employing tris-citric acid extender i.e., control did not receive any L-Carnitine and experimental groups having 0.5, 1.0, and 1.5 ng/mL of L-carnitine at 37 C with approximately 50 x 106 sperm/mL. The semen was cooled at 4 C and then equilibrated (4 hours), filled in straws (0.5 mL) at4 C, placed on LN2 vapours for 10 min, and kept into an LN2 container. The thawed semen was evaluated for post-thaw quality. The integrity of the sperm plasma membrane and motility (P?0.05) was highest in the extenders having 1.0 ng/mL of L-carnitine as compared to the control(received no L-Carnitine). However, sperm chromatin integrity and viability(live sperm with intact acrosome) remained similar. It was concluded that supplementing 1.0 ng/mL L-Carnitine of extender can improve the post-thaw quality of cryopreserved sperm. Based on the results of the current experiments it is recommended to include L-carnitine extender to improve post-thaw quality of buffalo sperm in terms of its motility and integrity ofits plasma membrane. <p class="MDPI17abstract"><strong>Objective: </strong>The purpose of this research was to find out the rise of spermatozoa motility of Waringin sheep which was given additional amount of PGF2α in commercial semen diluents (Andromed).</p><p class="MDPI17abstract"><strong>Methods: </strong>A total of 3 Waringin sheep aged 2-3 years were used in this study. Semen samples were collected using an artificial vagina and evaluation of the quality of the spermatozoa was carried out macroscopically and microscopically. This study used a unidirectional completely randomized design. After evaluated, semen samples were added with Andromed diluents afterwards divided into three groups treatment, K1; K2; and K3 where each group was then added with physiologic NaCl, 37,5 µg PGF2α, and 75 µg PGF2α. All of the samples were equilibrated in refrigerator for 4 hours and spermatozoa motility was examined, then after the freezing process was carried out and stored for 24 hours, the post thawing spermatozoa motility was examined. The data obtained was analyzed using one-way analysis of variance (ANOVA) and followed by Duncan test.<strong></strong></p><p class="MDPI17abstract"><strong>Results: </strong>The result of the research showed that motility of post equilibration spermatozoa (%) of Waringin sheeps at K1; K2; and K3 were 43,67±1,20; 75,00±4,00; and 82,33 ±3,18 (P&lt;0,05); and motility of post thawing spermatozoa (%) of Waringin sheeps at K1; K2; and K3 were 25,33±1,85; 49,33±3,17; dan 54,00 ±5,50 (P&lt;0,05).<strong></strong></p><strong>Conclusions: </strong>In conclusion that was the rise in motility of prefreezing spermatozoa and motility of post thawing spermatozoa of Waringin sheep which given additional amount of PGF2α on Andromed diluents.

Objective: To assess the testicular and semen characteristics in Criollo LecheroTropical (CLT) bulls in the subhumid Mexican tropics.Design/methodology/approach: Eight bulls were included in the study and distributedin two groups: G1 (n=5): young bulls and G2 (n=3): adult bulls. All bulls were managedunder grazing and were evaluated throughout one year. From each bull the followingmeasurements were taken once a month: live weight, body condition score, scrotalcircumference, and testicular width, length and volume. Semen was obtained every 3months via artificial vagina. Semen variables evaluated in each collection were: aspect,volume, mass and individual motility, and sperm concentration and morphology. Results: Live weight and testicular measurements increased linearly during the study inbulls from both groups. All bulls had scrotal circumference larger than the minimumthreshold value for cattle and semen of high quality.Study limitations/implications: The low availability of CLT bulls prevented theinclusion of a larger number of animals in the study. This low availability of animalsmakes it necessary to establish standard values for testicular measurements and semencharacteristics in the CLT breed in order to select the best individuals as sires andcontribute to its conservation.Findings/conclusions: The CLT bulls had good scrotal circumference and semenquality since a young age and into adulthood. These traits make the CLT bulls animportant alternative for livestock breeding in the tropics.
