A starch-hydrolysing and extremely halophilic archaeon (strain BD-3T), isolated from Aibi salt lake in Xin-Jiang, China, was characterized phenotypically and genotypically in order to determine its taxonomic status. On the basis of its polar lipid composition, nucleotide sequences of its 16S rRNA genes, genomic DNA G+C content (62.4 mol%) and growth characteristics, the organism could be assigned to the genus Haloarcula. Phenotypic differences and low DNA–DNA hybridization values to related Haloarcula species distinguished strain BD-3T from recognized Haloarcula species. It is therefore concluded that strain BD-3T represents a novel species, for which the name Haloarcula amylolytica sp. nov. is proposed. The type strain is BD-3T (=CGMCC 1.5335T=JCM 13557T).

An extremely halophilic archaeon belonging to the order Halobacteriales was isolated from Fuente de Piedra salt lake, Spain. This strain, designated FP1T, was a pleomorphic coccoid, neutrophilic and required at least 15 % (w/v) NaCl for growth. Strain FP1T grew at 37–60 °C, with optimal growth at 50 °C. Mg2+ was not required, but growth was observed with up to 10 % (w/v) MgSO4. Polar lipid analysis revealed the presence of mannose-6-sulfate(1-2)-glucose glycerol diether as a major glycolipid. Both C20C20 and C20C25 core lipids were present. The genomic DNA G+C content was 62.0 mol%. Phylogenetic analysis based on comparison of 16S rRNA gene sequences demonstrated that the isolate was most closely related to species of the genus Haloterrigena. DNA–DNA reassociation values between strain FP1T and the most closely related species of the genus Haloterrigena (Haloterrigena thermotolerans, Haloterrigena saccharevitans and Haloterrigena limicola) were lower than 29 %. It is therefore considered that strain FP1T represents a novel species of the genus Haloterrigena, for which the name Haloterrigena hispanica sp. nov. is proposed. The type strain is FP1T (=DSM 18328T=ATCC BAA-1310T).

Five bacterial strains, designated as NUM 1001T, NUM 1002, NUM 1003, NUM 1004 and NUM 1005, were isolated from the oral cavities of pigs. Colonies grown on mitis salivarius agar were similar in morphology to those of mutans streptococci. The novel isolates were analysed biochemically using the Rapid ID 32 Strep microsystem, subjected to DNA–DNA hybridization with oral streptococci and had their 16S rRNA genes sequenced. On the basis of the phylogenetic and phenotypic evidence obtained, the strains represent a novel species of the genus Streptococcus, for which the name Streptococcus orisuis sp. nov. is proposed. The type strain is NUM 1001T (=JCM 14035T=DSM 18307T).

The taxonomic status of six strains of Acinetobacter obtained from meat samples, collected from supermarkets in Porto, Portugal, was investigated using polyphasic analysis. Partial rpoB sequence similarities lower than 95 % to other Acinetobacter species with validly published names led to the hypothesis that these strains represented novel species. This was confirmed based on comparative multilocus sequence analysis, which included the gyrB, recA and 16S rRNA genes, revealing that these strains represented two coherent lineages that were distinct from each other and from all known species. The names Acinetobacter portensis sp. nov. (comprising four strains) and Acinetobacter guerrae sp. nov. (comprising two strains) are proposed for these novel species. The species status of these two groups was confirmed by low (below 95 %) whole-genome sequence average nucleotide identity values and low (below 70 %) digital DNA–DNA hybridization similarities between the whole-genome sequences of the proposed type strains of each novel species and the representatives of the known Acinetobacter species. Phylogenomic treeing from core genome analysis supported these results. The coherence of each new species lineage was supported by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry differentiation of the species at the protein level, by cellular fatty acid profiles, and by unique and differential combinations of metabolic and physiological properties shared by each novel species. The type strain of A. portensis sp. nov. is AC 877T (=CCUG 68672T=CCM 8789T) and the type strain of A. guerrae sp. nov. is AC 1271T (=CCUG 68674T=CCM 8791T).

A number of bacteria were isolated from sea water in Skagerrak, Denmark, at 30 m depth. Two of the isolates, strains D28 and D30T, belonged to the Flavobacteriaceae within the Cytophaga–Flavobacterium–Bacteroides group. Sequencing of 16S rRNA genes of the two strains indicated strongly that they belonged to the genus Tenacibaculum and that they showed greatest similarity to the species Tenacibaculum amylolyticum and Tenacibaculum mesophilum. DNA–DNA hybridization values, DNA base composition and phenotypic characteristics separated the Skagerrak strains from the other species within Tenacibaculum. Thus, it is concluded that the strains belong to a novel species within the genus Tenacibaculum, for which the name Tenacibaculum skagerrakense sp. nov. is proposed, with strain D30T (=ATCC BAA-458T=DSM 14836T) as the type strain.

An extremely halophilic archaeon, strain Fa-1T, was isolated from a marine solar saltern in Fujian, China. Strain Fa-1T required Mg2+ and at least 2.0 M NaCl for growth. It was able to grow at pH 6.5–9.0 (optimally at pH 7.0–7.5) and at 20–55 °C (optimally at 37–42 °C). The major polar lipids of strain Fa-1T were phosphatidylglycerol, phosphatidylglycerol phosphate methyl ester, phosphatidylglycerol sulfate and a sulfated diglycosyl diether. On the basis of a 16S rRNA gene sequence analysis, strain Fa-1T was closely related to nine species of the genus Halorubrum, showing sequence similarities of 97.4–98.4 %. The G+C content of the DNA of strain Fa-1T is 64.9 mol% (T m). DNA–DNA hybridization values between strain Fa-1T and the most closely related members of the genus Halorubrum were below 51 %. On the basis of the data from this study, strain Fa-1T represents a novel species of the genus Halorubrum, for which the name Halorubrum litoreum sp. nov. is proposed. The type strain is Fa-1T (=CGMCC 1.5336T =JCM 13561T).

An extremely halophilic archaeon (strain 7-3T) was isolated from Aibi salt lake in Xin-Jiang, China. It formed small (<1.0 mm), red, elevated colonies on salt-milk agar medium. Strain 7-3T was able to grow at pH 6.5–9.5 (optimum pH 8.0) and at 20–50 °C (optimum 45 °C). Analysis of its 16S rRNA gene sequence indicated that strain 7-3T was phylogenetically related to members of the genus Natronorubrum, with sequence similarities of 97.0 % to Natronorubrum bangense A33T and 95.2 % to Natronorubrum tibetense GA33T. The G+C content of its DNA was 61.2 mol% (T m). Levels of DNA–DNA relatedness to Nrr. bangense and Nrr. tibetense were 45 and 37 %, respectively. It is concluded that strain 7-3T (=CGMCC 1.4299T=JCM 13488T) should be classified as the type strain of a novel species of the genus Natronorubrum, for which the name Natronorubrum aibiense sp. nov. is proposed. Based on the properties of Nrr. aibiense given here and of those of Nrr. bangense and Nrr. tibetense described previously, an emended description of the genus Natronorubrum is presented.

A novel actinomycete, designated Z4T, was isolated from soil in Yaan, Sichuan Province, south China. The taxonomic status of this strain was established using a polyphasic approach. The morphological and chemotaxonomic characteristics of the organism are typical of the members of the genus Streptomyces . Phylogenetic analysis based on the almost complete 16S rRNA gene sequence indicated that strain Z4T belonged to the genus Streptomyces , branching off next to Streptomyces durhamensis ATCC 23194T (98.7 %), Streptomyces puniciscabiei KACC 20253T (98.7 %) and Streptomyces filipinensis ATCC 23905T (98.6 %). However, DNA–DNA hybridization studies and phenotypic differences between strain Z4T and closely related species of the genus Streptomyces suggested that strain Z4T represented a different genomic species. It is therefore proposed that Z4T ( = CGMCC 4.7035T = KCTC 29111T) represents the type strain of a novel species of the genus Streptomyces , for which the name Streptomyces yaanensis sp. nov. is proposed.

A new mayfly species, Bleptus michinokuensis sp. nov. (Ephemeroptera: Heptageniidae) is described on the basis of specimens of male and female adults and mature nymphs collected at a seepage zone of a small freshwater branch of the ‘Tachiya-zawa-gawa’ River located amongst the northern foothills of Mt. Gassan (Shonai-machi Town, Yamagata Prefecture, Japan). This new Bleptus species is characterized by its clear fore and hind wings. That is, they neither exhibit the distinct black band on the fore wings, nor the characteristic darkened margins along the edges of both the fore and hind wings. Rather it has a blackish colored terminal half of its fore legs (i.e., tibial, tarsal and pretarsal segments). These features differ clearly when comparing them to the other known species, Bleptus fasciatus Eaton. The information and data describing the habitat and distribution range of this new species are also noted. We also examined and discussed the genetic relationship of two Bleptus mayflies to settle the taxonomic status, inferred from the partially sequenced cytochrome c oxidase subunit I (COI) and large mitochondrial ribosomal subunit (16S rRNA) genes, and also the nuclear internal transcribed spacer 2 (ITS2) gene sequences. Consequently, phenetic and molecular phylogenetic analyses agreed well in terms of clustering.

Strain CC65T, a novel extremely halophilic archaeon, was isolated from a brine sample of a salt lake in Iran. The novel strain was light yellow-pigmented, non-motile, pleomorphic and required at least 1.7 M NaCl and 0.02 M MgCl2 for growth. Optimal growth was achieved at 3.5 M NaCl and 0.4 M MgCl2. The optimum pH and temperature for growth were pH 7.5 and 40 °C, respectively, while it was able to grow over a pH and a temperature range of pH 6.5–9.0 and 30–50 °C, respectively. Analysis of 16S rRNA gene sequence revealed that strain CC65T clustered with the sole member of the genus Halopenitus , Halopenitus persicus DC30T with a sequence similarity of 98.0 %. The polar lipid profile of strain CC65T consisted of phosphatidylglycerol and phosphatidylglycerol phosphate methyl ester. An unidentified glycolipid and two minor phospholipids were also observed. The only quinone present was MK-8(II-H2). The DNA G+C content of strain CC65T was 63.8 mol%. On the basis of the biochemical and physiological characteristics, as well as DNA–DNA hybridization (44 % with Halopenitus persicus IBRC 10041T), strain CC65T is classified as a novel species of the genus Halopenitus , for which the name Halopenitus malekzadehii sp. nov. is proposed. The type strain is CC65T ( = IBRC-M 10418T = KCTC 4045T).

Strain SH-6T was isolated from the sediment of Lake Shangmatala, a saline lake in Inner Mongolia (China). Cells were pleomorphic. The organism was neutrophilic and required at least 2.5 M (15 %) NaCl, but not MgCl2, for growth; optimal growth occurred at 4.3 M (25 %) NaCl. The G+C content of its DNA was 63.1 mol%. 16S rRNA gene sequence analysis revealed that strain SH-6T is a member of the family Halobacteriaceae, but there was a low level of similarity with other members of this family. Highest sequence similarity (94.6 %) was obtained with the 16S rRNA genes of the type strains of Natronolimnobius innermongolicus and Natronolimnobius baerhuensis. Polar lipid analyses revealed that strain SH-6T contains phosphatidylglycerol and phosphatidylglyceromethylphosphate, derived from both C20C20 and C20C25 glycerol diethers together with the glycolipid S2-DGD-1. On the basis of the data obtained, the new isolate could not be classified in any recognized genus. Strain SH-6T is thus considered to represent a novel species in a new genus within the family Halobacteriaceae, order Halobacteriales, for which the name Halopiger xanaduensis gen. nov., sp. nov. is proposed. The type strain of Halopiger xanaduensis is SH-6T (=CECT 7173T=CGMCC 1.6379T=JCM 14033T).
