Diterpenoid 3-epicaryoptin (C26H36O9) is abundant in leaves of Clerodendrum inerme, a traditionally used medicinal plant, having insect antifeedant activities. Here, we aim to explore its cell cycle delay and cytotoxic effects in Allium cepa root apical meristem cells. Allium cepa roots were treated with 3-epicaryoptin and colchicine for 2, 4, 4+16 h (4 h treatment followed by 16 h recovery) and the induced cell cycle delay and cytotoxic effects were analyzed. The highest metaphase cells frequency for 3-epicaryoptin and colchicine treated samples were found to be 66.2% and 82.35% respectively at 4 h treatment. Treatment of 3-epicaryoptin and colchicine increased the aberrant cells %, CA, MN, c-metaphase, and PP cells. Therefore, indiscriminate use of C. inerme in traditional medicine should be restricted and the cell cycle delay and cytotoxicity inducing effects of 3-epicaryoptin needs further exploration for its cancer chemotherapeutic applications.

Onions (Allium cepa L.) is one of the leading horticultural commodities in Indonesia and is often used as seasoning and traditional medicine. Onion has a high economic value and fluctuating prices so that domestic onion production needs to be improved, one of them with a presence of endophytic bacteria that act as plant growth promoting agent or Plant Growth Promoting Bacteria (PGPB). Endophytic bacteria isolated from the root, leaves, and bulbs. In this research has been tested endophytic bacteria of onion plants from Garut regency which has PGPB factors such as able to dissolve phosphate, and produce HCN. The results obtained 251 isolates of endophytic bacteria. Based on the characterization results, the superior isolates capable of dissolving phosphate with an average diameter of 0.45 cm is isolate II.B.1D.3, and 11 isolates capable of producing high HCN. These isolates can be used as PGPB agents so that they can be useful in increasing plant growth and onion production and biocontrol in suppressing pathogens. Keywords: PGPB, endophyte, onion, phosphate, HCN

The influence of increasing cytosine arabinoside (ara-C) concentration (50, 100, 300 and 500 μg/ml) on the mitotic activity of the apical meristem of onion adventitious roots was investigated during 24-h incubation in ara-C and postincubation in water. Incubation in ara-C inhibits reversibly mitosis, the degree of inhibition being dependent on the concentration used. 50 μg/ml ara-C causes only a slight and transitional mitotic depression, while 100—500 μg/ml reduces the per cent of mitoses in various degrees after 12-h incubation whereas after 24 h all concentrations (100—500) reduce mitosis to the same level of about 2 per cent (80% inhibition). During postincubation in water there occurs after treatment with 100—500 μg/ml ara-C, a wave of highly synchronized mitoses with a peak after 12 h. The highest frequency of synchronized mitoses (60°/o) appeared in roots treated with 300 μg/ml ara-C. The main wave is preceded by a small one with a peak after 2-4 h of postincubation. This seems to be the consequence of phases S and G<sub>2</sub> inhibition by ara-C. It was found that under the influence of ara-C the per cent of mitoses with chromosome aberration and of cells with micronuclei increased only slightly.

(CEBOLLA) EFFECT OF POTASSIUm SORBATE AT DIFFERENT CONCENTRATIONS AND ExPOSURE TImE ON THE CELL CyCLE AND THE GENETIC mATERIAL IN ROOT mERISTEmS OF AllIuM CEPA l “ONION” Joselyne Quispe, José Saldaña, Tony Verde y Shirley Valderrama Universidad Nacional de Trujillo, Trujillo,Perú DOI: https://doi.org/10.33017/RevECIPeru2010.0011/ RESUMEN Con el propósito de determinar el efecto del Sorbato de Potasio sobre el ciclo celular en meristemos radiculares de Allium cepa L. “cebolla” se establecieron tres grupos experimentales que se expusieron a las soluciones de Sorbato de Potasio en concentraciones de 0.5, 1 y 2 g/L; por 6, 7 y 8 horas cada una y un grupo control. Luego se realizó la respectiva coloración en Orceína acética al 2% de acuerdo a la técnica de Tjio y Levan, observándose con el microscopio compuesto un total de 1500 células meristemáticas en cada uno de los tratamientos. Los resultados aplicando tratamiento después de 8 horas de exposición a la concentración de 2 g/L mostraron una disminución del índice mitótico de 11.26 %, mientras que se incrementó el índice profásico en 95.57 %. Se produjeron también alteraciones como rupturas cromosómicas, puentes anafásicos y células binucleadas. Las pruebas y análisis de varianza, mostraron diferencias significativas entre los diferentes tratamientos, confirmado mediante la prueba de comparación múltiple de promedios de Duncan. Se evidencia un probable efecto genotóxico en el material genético por el Sorbato de Potasio y una alteración en el ciclo celular de los meristemos de A. cepa L.; así mismo, se comprueba la función de biosensor de A. cepa L. en el estudio de sustancias que puedan afectar el ciclo celular. ABSTRACT In order to determine the effect of Potassium Sorbate on the cell cycle in root meristems of Allium cepa L. “onion” set three experimental groups were exposed to solutions of Potassium Sorbate at concentrations of 0.5, 1 and 2g /l, for 6, 7 and 8 hours each and a control group. Later the respective color in 2% Acetic orcein according to the technique of Tjio and Levan, observed with the compound microscope a total of 1500 meristematic cells in each of the treatments. The appropriate treatment results after 8 hours of exposure to the concentration of 2 g / L showed a decrease in mitotic index of 11.26%, while it increased the rate at 95.57% prophasic index. There were also alterations as chromosome breaks, anaphasic bridges and binucleated cells. The tests and analysis of variance showed significant differences between the different treatments, as confirmed by multiple comparison test of Duncan averages. It shows a probable genotoxic effect in the genetic material by Potassium Sorbate and an alteration in the cell cycle of meristems of A. cepa L., likewise, is found biosensor function of A. cepa L. in the study of substances that may affect the cell cycle.
