Background Levels of 50% neutralizing titer (NT50) reflect the vaccine-induced humoral immunity after the vaccination against the severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2). Measurements of NT50 are difficult to implement in large quantities. A high-throughput laboratory test is expected for determining the level of herd immunity against SARS-CoV-2. Methods We analyzed samples from 168 Japanese healthcare workers who had completed two doses of the BNT162b2 vaccine. We analyzed immunoglobulin G (IgG) index values against spike protein (SP) using automated chemiluminescent enzyme immunoassay system AIA-CL and analyzed the background factors affecting antibody titer. SP IgG index was compared with 50% neutralization titers. Results The median SP IgG index values of the subjects (mean age = 43 years; 75% female) were 0.1, 1.35, 60.80, and 97.35 before and at 2, 4, and 6 weeks after the first dose, respectively. At 4 and 6 weeks after the first dose, SP IgG titers were found to have positive correlation with NT50 titer (r=0.7535 in 4 weeks; r=0.4376 in 6 weeks). Proportions of the SP IgG index values against the Alpha, Beta, Gamma, and Delta variants compared with the original strain were 2.029, 0.544, 1.017, and 0.6096 respectively. Older age was associated with lower SP IgG titer index 6 weeks after the first dose. Conclusions SP IgG index values were raised at 3 weeks after two doses of BNT162b2 vaccination and have positive correlation with NT50. SP IgG index values were lower in the older individuals and against Beta and Delta strain.

The recent spreading of new SARS-CoV-2 variants, carrying several mutations in the spike protein, could impact immune protection elicited by natural infection or conferred by vaccination. In this study, we evaluated the neutralizing activity against the viral variants that emerged in the United Kingdom (B.1.1.7), Brazil (P.1), and South Africa (B.1.351) in human serum samples from hospitalized patients infected by SARS-CoV-2 during the first pandemic wave in Italy in 2020. Of the patients studied, 59.5% showed a decrease (≥2 fold) in neutralizing antibody titer against B.1.1.7, 83.3% against P.1, and 90.5% against B.1.351 with respect to the original strain. The reduction in antibody titers against all analyzed variants, and in particular P.1 and B.1.351, suggests that previous symptomatic infection might be not fully protective against exposure to SARS-CoV-2 variants carrying a set of relevant spike mutations.

AbstractAvian cholera, caused by the bacterium Pasteurella multocida, is a common and important infectious disease of wild birds in North America. Between 2005 and 2012, avian cholera caused annual mortality of widely varying magnitudes in Northern common eiders (Somateria mollissima borealis) breeding at the largest colony in the Canadian Arctic, Mitivik Island, Nunavut. Although herd immunity, in which a large proportion of the population acquires immunity to the disease, has been suggested to play a role in epidemic fadeout, immunological studies exploring this hypothesis have been missing. We investigated the role of three potential drivers of fadeout of avian cholera in eiders, including immunity, prevalence of infection, and colony size. Each potential driver was examined in relation to the annual real-time reproductive number (Rt) of P. multocida, previously calculated for eiders at Mitivik Island. Each year, colony size was estimated and eiders were closely monitored, and evaluated for infection and serological status. We demonstrate that acquired immunity approximated using antibody titers to P. multocida in both sexes was likely a key driver for the epidemic fadeout. This study exemplifies the importance of herd immunity in influencing the dynamics and fadeout of epidemics in a wildlife population.

Systemic Inflammatory Response Syndrome (SIRS) mempunyai kebahayaan tinggi terjadi sepsis dan kematian. Nilai jumlahkeseluruhan leukosit merupakan salah satu peramal pasien SIRS dengan bakteriemia. Pemeriksaan jumlah monosit, angka bandinglimfosit Monocyte-Lymphocyte Ratio (MLR), Neutrophil-Lymphocyte Count Ratio (NLCR) dapat diketahui dengan pemeriksaan leukosit.Presepsin telah diteliti untuk mencerminkan kondisi sepsis. Penelitian ini bertujuan untuk mengetahui keberadaan hubungan jumlahmonosit, MLR dan NLCR dengan presepsin di SIRS lewat pembuktian. Ada 34 pasien SIRS di ICU RSUP Dr. Kariadi, diambil secaraberturutan antara selama bulan Januari−Februari 2014. Pemeriksaan darah rutin dengan hematology analyzer. MLR dan NLCR dihitung secara manual. Kadar presepsin ditentukan dengan metode Chemiluminescent Enzyme Immunoassay (CLEIA). Uji kenasabanPearson untuk hubungan MLR dan NLCR dengan presepsin. Uji kenasaban Spearman untuk jumlah monosit dengan presepsin. Kadarpresepsin subjek penelitian 286–15687 pg/mL. Terdapat 23(67,8%) subjek yang mempunyai jumlah monosit dalam rentang nilai rujukan.24(70,6%) dan memiliki jumlah neutrofil absolut lebih besar dari rentang nilai rujukan, sedangkan 21(61,8%) mempunyai jumlahlimfosit absolut dalam rentang nilai rujukan. Hubungan jumlah monosit dengan presepsin mempunyai nilai r= -0,204; p=0,247;yang terkait MLR dengan presepsin r=0,163; p=0,358; sedangkan NLCR dengan presepsin r=0,345; p=0,046. Didasari telitian ini,dapat disimpulkan tidak terdapat hubungan bermakna antara jumlah monosit dan MLR dengan presepsin, selain itu didapatkan pulahubungan positif berarti antara NLCR dan presepsin di SIRS.
